1. Estructura i funci\u00f3 de les prote\u00efnes bacterianes que regulen l\u2019expressi\u00f3 de la virul\u00e8ncia<\/strong>
\nLes interaccions prote\u00efna-prote\u00efna i prote\u00efna-ADN tenen un paper important en la capacitat dels bacteris virulents d\u2019adaptar-se a l\u2019entorn de l\u2019hoste i causar malalties. Actualment, la nostra recerca se centra en un grup de prote\u00efnes: les prote\u00efnes associades al nucleoide (NAP), que contribueixen a l\u2019estructura de l\u2019ADN i regulen l\u2019expressi\u00f3 g\u00e8nica. <\/p>\n

Ens interessa aclarir el paper que tenen dues d\u2019aquestes prote\u00efnes (Hha i H-NS) en la regulaci\u00f3 de la virul\u00e8ncia i en la transfer\u00e8ncia de plasmidis. Les variants patog\u00e8niques d\u2019Escherichia coli, com E. coli enteroagregatiu, s\u00f3n el subjecte de la nostra recerca. A causa de la seva funci\u00f3 essencial, aquestes prote\u00efnes mostren un potencial excel\u00b7lent per combatre la infecci\u00f3 bacteriana.<\/p>\n

2. Plasmidis bacterians i el seu paper en la transmissi\u00f3 de marcadors de resist\u00e8ncia a m\u00faltiples f\u00e0rmacs<\/strong>
\nUna de les preocupacions principals en relaci\u00f3 amb les infeccions bacterianes \u00e9s la selecci\u00f3 d\u2019a\u00efllats que siguin resistents a diversos f\u00e0rmacs antimicrobians. En molts casos, la transmissi\u00f3 de la capacitat de les c\u00e8l\u00b7lules bacterianes de resistir simult\u00e0niament a diversos f\u00e0rmacs \u00e9s deguda als plasmidis. Aquests elements gen\u00e8tics es poden transmetre d\u2019una c\u00e8l\u00b7lula a una altra i poden modificar el fenotip de la c\u00e8l\u00b7lula receptora. <\/p>\n

Recentment, hem demostrat que els plasmidis presents en la resist\u00e8ncia a m\u00faltiples f\u00e0rmacs de la Salmonella requereixen que es mantinguin de manera estable prote\u00efnes de plasmidi espec\u00edfiques en aquest microorganisme. Aquestes prote\u00efnes es podrien considerar dianes per combatre la resist\u00e8ncia a m\u00faltiples f\u00e0rmacs.<\/p>\n

3. Aplicaci\u00f3 de nanoeines de biotecnologia bacteriana<\/strong><\/p>\n

3.1. Dielectrophoresis. Ja hem mostrat que la dielectroforesi pot ser una eina valuosa per a la classificaci\u00f3 i la caracteritzaci\u00f3 dels bacteris. Actualment usem diferents dissenys de xips (el\u00e8ctrodes de carb\u00f3 2D i 3D) per: a) estudiar l\u2019efecte dels camps el\u00e8ctrics en la fisiologia cel\u00b7lular bacteriana; b) combinar la DEP amb altres protocols moleculars per a la detecci\u00f3 i identificaci\u00f3 de diversos tipus de c\u00e8l\u00b7lules. Els resultats recents han demostrat que els xips de la DEP es poden utilitzar per incrementar la detecci\u00f3 de la PCR de les c\u00e8l\u00b7lules de llevats. <\/p>\n

3.2. Microsc\u00f2pia de forces at\u00f2miques (AFM). Els enfocaments convencionals d\u2019AFM han mostrat que s\u00f3n t\u00e8cniques potents per caracteritzar els biomaterials i les biomol\u00e8cules. En un projecte conjunt amb el grup de Caracteritzaci\u00f3 bioel\u00e8ctrica a la nanoescala (p\u00e0gina 56), tenim la intenci\u00f3 d\u2019emprar l\u2019AFM el\u00e8ctrica per caracteritzar l\u2019embolcall de les c\u00e8l\u00b7lules bacterianes. Tamb\u00e9 planegem usar aquest enfocament per analitzar les propietats estructurals i fisiol\u00f2giques de les c\u00e8l\u00b7lules bacterianes vives.\u00a0 <\/p>\n\n<\/div>

Two groups working together at IBEC demonstrate the potential of electrical studies of single bacterial cells in a paper published in ACS Nano. Gabriel Gomila\u2019s Nanoscale Bioelectrical Characterization group and that of Antonio Ju\u00e1rez, Microbial Biotechnology and Host-pathogen Interaction, combined their expertise on microscopic electrical measurements and bacteria respectively to come up with a way to study the response to external electrical fields of just a single bacterial cell.<\/p>\n

IBEC Senior Researcher Eduard Torrents participated in a conference to mark the National Day for Cystic Fibrosis last Wednesday 25 April. This event, which took place at the Spanish Society of Pneumology and Thoracic Surgery (SEPAR), also included the official presentation of funds from the Associaci\u00f3 Catalana de Fibrosis Qu\u00edstica (Catalan Association of Cystic Fibrosis) to research groups specializing in the disease.<\/p>\n

An IBEC group has embarked on a technology transfer venture together with two biopharmaceutical companies. Antonio Ju\u00e1rez\u2019s Microbial Technology and Host-Pathogen Interaction lab has formed a consortium with CZV Veterinaria, a leader in the manufacture of veterinary products based in Porri\u00f1o, Galicia, and Valls Companys\u2019 pharmaceutical arm MEVET in Lleida. Their two-year project aims to obtain strains of Salmonella with weakened virulence, which can then be used to develop a vaccine to reduce the incidence of the infection in poultry farms. Salmonella, a leading cause of food poisoning, is zoonotic, able to spread to man through contaminated animal food products.<\/p>\n
