Aquest proc\u00e9s es coneix com a mecanotransducci\u00f3 i t\u00e9 un paper fonamental en l&#8217;evoluci\u00f3 dels tumors s\u00f2lids, com el c\u00e0ncer de mama.<\/p>\n\n\n\n<p>Se sap que un canvi mec\u00e0nic habitual en processos de c\u00e0ncer \u00e9s l&#8217;enduriment dels teixits. Aquesta rigidesa \u00e9s precisament el que es detecta en fer una autoexploraci\u00f3 o palpaci\u00f3 mam\u00e0ria, per detectar possibles tumors. La rigidesa del teixit mamari provoca una reacci\u00f3 en cadena, generant tensi\u00f3 a l&#8217;interior de les c\u00e8l\u00b7lules i deformant el seu nucli. En \u00faltima inst\u00e0ncia, la deformaci\u00f3 del nucli activa gens que controlen la proliferaci\u00f3 cel\u00b7lular i estan relacionats amb el creixement tumoral.<\/p>\n\n\n\n<p>Un estudi publicat avui a la revista <em>Nature Materials<\/em> demostra un mecanisme cel\u00b7lular que podria ser clau per frenar l&#8217;avan\u00e7 dels tumors de mama. Els resultats del treball, liderat per Pere Roca-Cusachs, investigador principal de l&#8217;IBEC i de la Universitat de Barcelona, indiquen que la laminina, una prote\u00efna que dona consist\u00e8ncia i suport als teixits mamaris sans, dificulta el proc\u00e9s de mecanotransducci\u00f3 a les c\u00e8l\u00b7lules, protegint el nucli de ser deformat.<\/p>\n\n\n\n<p><strong>&#8220;Els nostres resultats mostren que la pres\u00e8ncia de laminina evita els efectes de la rigidesa. \u00c9s a dir, que protegeix les c\u00e8l\u00b7lules contra el creixement tumoral. Demostrem aquest mecanisme <em>in vitro<\/em>, per\u00f2 creiem que tamb\u00e9 podria funcionar <em>in vivo <\/em>ja que existeix una concordan\u00e7a amb l&#8217;observat en mostres de pacients de c\u00e0ncer de mama&#8221;<\/strong>, explica Zanetta Kechagia, investigadora postdoctoral de l&#8217;IBEC i primera autora de l&#8217;estudi.<\/p>\n\n\n\n<p><strong>&#8220;A partir d&#8217;aquest mecanisme, que hem demostrat que preveu la invasi\u00f3 de c\u00e8l\u00b7lules tumorals, es podrien dissenyar eines diagn\u00f2stiques m\u00e9s sensibles o noves ter\u00e0pies potencials contra el c\u00e0ncer de mama. Les m\u00e9s habituals estan relacionades amb canvis en el citoesquelet de les c\u00e8l\u00b7lules per interaccionar amb el teixit i migrar, aix\u00ed com amb l&#8217;activaci\u00f3 de la prote\u00efna YAP, que entra al nucli accionant gens relacionats amb la proliferaci\u00f3 cel\u00b7lular.<\/p>\n\n\n\n<p>Per estudiar el proc\u00e9s de mecanotransducci\u00f3, l&#8217;equip d&#8217;investigaci\u00f3 va sembrar c\u00e8l\u00b7lules de teixit mamari sobre gels amb rigidesa variable per simular teixits sans (tous) i malignes (r\u00edgids).
