Els investigadors han explorat diversos m\u00e8todes per a la visualitzaci\u00f3 en viu i l&#8217;estimulaci\u00f3 de l&#8217;activitat profunda del cervell. Un d&#8217;aquests m\u00e8todes \u00e9s l&#8217;excitaci\u00f3 multifot\u00f3 mitjan\u00e7ant l&#8217;\u00fas de llum infraroja (IR) polsada. Els teixits absorbeixen de forma molt feble aquest tipus de llum, de manera que pot penetrar a trav\u00e9s de l&#8217;os i en profunditat en \u00f2rgans com el cervell. Tanmateix, t\u00e9 limitacions per produir imatges enfocades i controlar l&#8217;activitat cel\u00b7lular amb precisi\u00f3. Per superar aix\u00f2, els cient\u00edfics han estat investigant l&#8217;excitaci\u00f3 mitjan\u00e7ant tres fotons utilitzant polsos ultracurts de llum IR per aconseguir la visualitzaci\u00f3 de tot el c\u00f2rtex cerebral i l&#8217;observaci\u00f3 de l&#8217;activitat neuronal. No obstant aix\u00f2, fins ara, no s&#8217;havia provat l&#8217;estimulaci\u00f3 neuronal in vivo utilitzant l&#8217;excitaci\u00f3 de tres fotons.<\/p>\n\n\n\n<p>Un estudi pioner, liderat per l&#8217;Institut de Bioenginyeria de Catalunya (IBEC) en col\u00b7laboraci\u00f3 amb l&#8217;ICFO &#8211; Institut de Ci\u00e8ncies Fot\u00f2niques, ambd\u00f3s membres del BIST, ha presentat el primer m\u00e8tode per controlar l&#8217;activitat cerebral en organismes vius utilitzant f\u00e0rmacss activats per excitaci\u00f3 de tres fotons i llum infraroja mitjana. Aquest m\u00e8tode es basa en l&#8217; activaci\u00f3 d&#8217; un receptor espec\u00edfic per a l&#8217; acetilcolina, un neurotransmissor vital involucrat en diversos processos cerebrals, com l&#8217; aprenentatge, l&#8217; atenci\u00f3 i la mem\u00f2ria.<\/p>\n\n\n\n<p>Per aconseguir-ho, els investigadors van utilitzar PAI, una mol\u00e8cula sensible a la llum pr\u00e8viament desenvolupada a l&#8217;IBEC, utilitzant la concentraci\u00f3 de f\u00e0rmac m\u00e9s baixa i la longitud d&#8217;ona de fotoactivaci\u00f3 m\u00e9s llarga mai registrada.<\/p>\n\n\n\n<p>L&#8217;estudi, publicat a la revista Angewandte Chemie, va ser dirigit pel Dr. Pau Gorostiza, Professor de Recerca ICREA, membre del CIBER-BBN i l\u00edder del grup de <a href=\"https:\/\/ibecbarcelona.eu\/nanoprobes\">Nanosondas i Nanoconmutadors<\/a> a l&#8217;IBEC, i pel Dr. Pablo Loza-\u00c1lvarez, cap del laboratori de <a href=\"https:\/\/www.icfo.eu\/research-group\/4\/sln\/home\/\">Microsc\u00f2pia de Superresoluci\u00f3 i Nanosc\u00f2pia (SNL)<\/a> de l&#8217;ICFO.<\/p>\n\n\n\n<p><strong>&#8220;La novetat d&#8217;aquests resultats \u00e9s que ens demostren que podem controlar l&#8217;activitat dels f\u00e0rmacss utilitzant llum infraroja. A m\u00e9s, la majoria dels compostos fotosensibles que sol\u00edem usar en fotofarmacologia amb llum ultraviolada i visible s\u00f3n igualment susceptibles a ser activats mitjan\u00e7ant l&#8217;excitaci\u00f3 de tres fotons amb llum infraroja de longitud mitjana, que resulta menys agressiva per als teixits.&#8221;,<\/strong> explica Gorostiza.<\/p>\n\n\n\n<p><strong>A m\u00e9s, aquesta t\u00e8cnica, en ser il\u00b7luminaci\u00f3 IR, ofereix la possibilitat d&#8217;arribar molt profund dins del teixit i amb resolucions submicrom\u00e8triques en les tres dimensions. En termes simples, significa que podem localitzar l&#8217;activaci\u00f3 just en el punt focal del feix l\u00e0ser, en il\u00b7luminar-lo externament a trav\u00e9s del crani&#8221;<\/strong>, agrega Loza-\u00c1lvarez.<\/p>\n\n\n\n<p>Aquests resultats mostren el potencial de la farmacologia de tres fotons i obren noves perspectives per a la recerca fonamental en neurobiologia i el desenvolupament de ter\u00e0pies de neuromodulaci\u00f3 basades en la llum.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\"><strong>El poder de la llum<\/strong><\/h4>\n\n\n\n<p>La llum es pot utilitzar per aconseguir dirigir l&#8217; acci\u00f3 de f\u00e0rmacss en \u00e0rees espec\u00edfiques del cos mitjan\u00e7ant la fotofarmacologia. Aquest enfocament innovador implica modificar l&#8217; estructura qu\u00edmica d&#8217; un f\u00e0rmacs agregant a la seva estructura un interruptor molecular activat per la llum. En el seu nou article cient\u00edfic, els investigadors van rec\u00f3rrer a la llum infraroja de longitud d&#8217;ona m\u00e9s llarga i a c\u00e0lculs te\u00f2rics per millorar l&#8217;activaci\u00f3 de PAI a trav\u00e9s d&#8217;excitacions de m\u00faltiples fotons. Aquests c\u00e0lculs, realitzats pel laboratori de Josep Maria Lluch i Jordi Hernando a la Universitat Aut\u00f2noma de Barcelona, van suggerir que l&#8217;excitaci\u00f3 de tres fotons podria ser m\u00e9s eficient que les alternatives de dos fotons i que el principi podria ser \u00e0mpliament aplicable.<\/p>\n\n\n\n<p>Posteriorment, els investigadors van dur a terme experiments in vivo per validar aquestes prediccions te\u00f2riques. Van utilitzar larves de peix zebra gen\u00e8ticament modificades per expressar un indicador de calci fluorescent. L&#8217; activitat neuronal condueix a r\u00e0pides fluctuacions en els nivells intracel\u00b7lulars de calci lliure, ra\u00f3 per la qual els indicadors de calci s&#8217; utilitzen comunament per visualitzar l&#8217; activitat neuronal.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>La primera vegada que vam veure les respostes cerebrals quedem realment impressionats. Tot el cervell es va il\u00b7luminar<\/p>\n<cite>Rosalba Sortino<\/cite><\/blockquote>\n\n\n\n<p>Aquesta configuraci\u00f3 experimental els va permetre observar i analitzar les alteracions en els nivells de calci quan el cervell va ser exposat a estimulaci\u00f3 lum\u00ednica a trav\u00e9s de l&#8217;excitaci\u00f3 de tres fotons: <strong>&#8220;La primera vegada que vam veure les respostes cerebrals quedem realment impressionats. Administrar-nos el compost PAI en la seva forma inactiva, que va ser captat pel cervell del peix zebra, i quan l&#8217;excitem amb els destells de tres fotons, tot el cervell es va il\u00b7luminar.
