Tot i no tenir una elevada mortalitat, gaireb\u00e9 la meitat dels tumors de bufeta tornen a apar\u00e8ixer al cap de 5 anys, de manera que es requereix una vigil\u00e0ncia continuada del pacient, amb freq\u00fcents visites a l&#8217;hospital i la necessitat de repetir el tractament. Per tot aix\u00f2, el c\u00e0ncer de bufeta \u00e9s un dels m\u00e9s costosos de curar.<\/p>\n\n\n\n<p>Els tractaments actuals que impliquen l&#8217;administraci\u00f3 de f\u00e0rmacs directament a l&#8217;interior de la bufeta han demostrat bones taxes de superviv\u00e8ncia, per\u00f2 una efic\u00e0cia terap\u00e8utica baixa. Una alternativa prometedora \u00e9s l&#8217;\u00fas de nanopart\u00edcules, capaces de fer arribar l&#8217;agent terap\u00e8utic directament al tumor. En especial, destaquen els nanorobots, nanopart\u00edcules amb capacitat d&#8217;autopropulsar-se per l&#8217;interior del cos.<\/p>\n\n\n\n<p>Ara, un estudi publicat a la prestigiosa revista <em>Nature Nanotechnology<\/em> mostra com un equip de recerca ha estat capa\u00e7 de reduir en un 90% la mida de tumors de bufeta en ratolins administrant una sola dosi de nanorobots propulsats per urea.<\/p>\n\n\n\n<p>Aquestes diminutes nanom\u00e0quines estan formades per una esfera porosa de s\u00edlice. En la seva superf\u00edcie, incorporen diversos components amb funcions espec\u00edfiques. Un d&#8217;ells \u00e9s l&#8217;enzim ureasa, una prote\u00efna que reacciona amb la urea, present a l&#8217;orina, fent que la nanopart\u00edcula sigui capa\u00e7 de propulsar-se. Un altre component clau \u00e9s el iode radioactiu, un radiois\u00f2top utilitzat comunament per al tractament localitzat de tumors.<a><\/a><\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>&#8220;Amb una sola dosi veiem una disminuci\u00f3 del 90% del volum del tumor. \u00c9s molt m\u00e9s eficient, tenint en compte que l&#8217;habitual en pacients amb aquest tipus de tumors \u00e9s que vagin entre 6 i 14 vegades a l&#8217;hospital.&#8221;<\/p>\n<cite>Samuel S\u00e1nchez<\/cite><\/blockquote>\n\n\n\n<p>El treball, liderat per l&#8217;Institut de Bioenginyeria de Catalunya (IBEC) i el CIC biomaGUNE i desenvolupat amb la col\u00b7laboraci\u00f3 de l&#8217;Institut de Recerca Biom\u00e8dica (IRB Barcelona) i de la Universitat Aut\u00f2noma de Barcelona (UAB), obre la porta a nous tractaments per al c\u00e0ncer de bufeta, que redueixin el temps d&#8217;hospitalitzaci\u00f3, cosa que implicaria un menor cost i una major comoditat per al pacient.<\/p>\n\n\n\n<p>&#8220;Amb una sola dosi veiem una disminuci\u00f3 del 90% del volum del tumor. \u00c9s molt m\u00e9s eficient, tenint en compte que l&#8217;habitual en pacients amb aquest tipus de tumors \u00e9s que vagin entre 6 i 14 vegades a l&#8217;hospital. Aquesta caracter\u00edstica suposa un avantatge respecte al procediment actual, on un cop administrat el tractament directament a la bufeta, el pacient ha de canviar de posici\u00f3 cada mitja hora per aconseguir que el f\u00e0rmac arribi a totes les parets.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>&#8220;Aquests nanorobots tenen la capacitat de descompondre la matriu extracel\u00b7lular del tumor fet que podria afavorir una major penetraci\u00f3 tumoral.&#8221;<\/p>\n<cite>Meritxell Serra Casablancas<\/cite><\/blockquote>\n\n\n\n<p>El nou treball va m\u00e9s enll\u00e0 en demostrar no nom\u00e9s la mobilitat de les nanopart\u00edcules a la bufeta, sin\u00f3 tamb\u00e9 la seva acumulaci\u00f3 espec\u00edfica en el tumor. Aix\u00f2 va ser possible gr\u00e0cies a diferents t\u00e8cniques, incloses imatges m\u00e8diques de tomografia per emissi\u00f3 de positrons (PET) dels ratolins, aix\u00ed com imatges de microsc\u00f2pia sobre els teixits extirpats despr\u00e9s de la finalitzaci\u00f3 de l&#8217;estudi. Aquestes \u00faltimes es van prendre mitjan\u00e7ant un sistema de microsc\u00f2pia de fluoresc\u00e8ncia desenvolupat espec\u00edficament per a aquest projecte a l&#8217; IRB Barcelona. El sistema permet observar la bufeta completa, escanejant les diferents capes de l&#8217;\u00f2rgan per despr\u00e9s obtenir una reconstrucci\u00f3 en 3D.<\/p>\n\n\n\n<p>&#8220;L&#8217;innovador sistema \u00f2ptic que hem desenvolupat, ens va permetre, anul\u00b7lar la llum que reflectia el mateix tumor i aix\u00ed identificar i localitzar les nanopart\u00edcules en tot l&#8217;\u00f2rgan, sense un marcatge previ, a una resoluci\u00f3 sense precedents. Els nanorobots no contenen anticossos espec\u00edfics per recon\u00e8ixer el tumor, i normalment, el teixit tumoral \u00e9s m\u00e9s r\u00edgid que el teixit sa.<\/p>\n\n\n\n<p>&#8220;No obstant aix\u00f2, observem que aquests nanorobots tenen la capacitat de descompondre la matriu extracel\u00b7lular del tumor en augmentar localment el pH mitjan\u00e7ant una reacci\u00f3 qu\u00edmica d&#8217;autopropulsi\u00f3. Aquest fenomen podria afavorir una major penetraci\u00f3 tumoral i va resultar ser benefici\u00f3s per assolir una acumulaci\u00f3 preferencial en el tumor&#8221;, detalla Meritxell Serra Casablancas, co-primera autora de l&#8217;estudi i investigadora de l&#8217;IBEC.<\/p>\n\n\n\n<p>Aix\u00ed, els cient\u00edfics van arribar a la conclusi\u00f3 que els nanorobots xoquen amb l&#8217;uroteli com si fos una paret, per\u00f2 en el tumor, en ser m\u00e9s esponj\u00f3s, penetren i s&#8217;acumulen al seu interior. Part de les dades deriven de les tesis doctorals de Meritxell Serra i Ana Hortelao, ambdues investigadores en el grup de <a href=\"https:\/\/ibecbarcelona.eu\/es\/nanodevices\">Nano biodispositius intel\u00b7ligents<\/a> de&nbsp; l&#8217;IBEC, liderat per S\u00e1nchez. Tamb\u00e9 de la tesi de Cristina Sim\u00f3, co-primera autora de l&#8217;estudi, qui va dur a terme la seva recerca predoctoral al Laboratori de Radioqu\u00edmica i Imatge Nuclear liderat per Jordi Llop al CIC biomaGUNE. S&#8217;afegeix l&#8217;experi\u00e8ncia en el model animal de la malaltia del grup de la UAB liderat per Esther Juli\u00e1n. A m\u00e9s, el projecte ha rebut finan\u00e7ament del Consell de Recerca Europeu (ERC) i la Fundaci\u00f3 &#8220;La Caixa&#8221;.<\/p>\n\n\n\n<p>La tecnologia en la qual es basen aquests nanorobots, en els quals Samuel S\u00e1nchez i el seu equip han treballat durant m\u00e9s de set anys, ha estat patentada recentment i \u00e9s la base de Nanobots Therapeutics, spin-off de l&#8217;IBEC i ICREA creada el gener del 2023.<\/p>\n\n\n\n<p>L&#8217;empresa, fundada per S\u00e1nchez, representa un pont entre la investigaci\u00f3 i l&#8217;aplicaci\u00f3 cl\u00ednica: &#8220;Obtenir un bon finan\u00e7ament per a la spin-off \u00e9s crucial per poder continuar desenvolupant aquesta tecnologia i, si tot va b\u00e9, que pugui arribar al mercat i a la societat. Al juny, nom\u00e9s despr\u00e9s de 5 mesos des de la creaci\u00f3 de Nanobots Tx, tanquem la primera ronda de finan\u00e7ament amb \u00e8xit, i estem emocionats pel futur.&#8221;, ressalta S\u00e1nchez.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\"><strong><strong>Innovaci\u00f3 tecnol\u00f2gica en microsc\u00f2pia per localitzar els nanorobots<\/strong><\/strong><\/h4>\n\n\n\n<p>El treball amb nanorobots ha suposat un important desafiament cient\u00edfic en t\u00e8cniques de bioimatge per a la visualitzaci\u00f3 d&#8217;aquests elements en els teixits i el mateix tumor. Les t\u00e8cniques no invasives m\u00e9s habituals utilitzades en l&#8217;\u00e0mbit cl\u00ednic -com PET- no tenen la resoluci\u00f3 necess\u00e0ria per ubicar a escala microsc\u00f2pica aquestes part\u00edcules de mida molt redu\u00efda. Per aix\u00f2, la Plataforma Cient\u00edfica de Microsc\u00f2pia de l&#8217;IRB Barcelona, va fer servir una t\u00e8cnica de microsc\u00f2pia que implica l&#8217;\u00fas d&#8217;una l\u00e0mina de llum l\u00e0ser per il\u00b7luminar les mostres i permet obtenir imatges tridimensionals per dispersi\u00f3 de la llum en xocar amb teixits i part\u00edcules. En observar que el mateix tumor dispersava part de la llum, generant interfer\u00e8ncies, van desenvolupar una nova t\u00e8cnica basada en llum polaritzada que anul\u00b7la tota la dispersi\u00f3 procedent del teixit i les c\u00e8l\u00b7lules del mateix tumor. Aix\u00f2 va permetre visualitzar i localitzar els nanorobots sense necessitat d&#8217;haver-los marcat pr\u00e8viament amb t\u00e8cniques moleculars.<\/p>\n\n\n\n<p><\/p>\n\n\n\n<p><strong>Sobre l&#8217;IBEC<\/strong><br>L&#8217;Institut de\u00a0Bioenginyeria\u00a0de Catalunya (IBEC) \u00e9s un centre CERCA, dues vegades reconegut com a Centre d&#8217;Excel\u00b7l\u00e8ncia Severo Ochoa, i amb el segell\u00a0TECNIO, desenvolupador de tecnologia i facilitador d&#8217;empreses. L&#8217;IBEC\u00a0\u00e9s membre de l&#8217;Institut de Ci\u00e8ncia i Tecnologia de Barcelona (BIST) i realitza una recerca multidisciplin\u00e0ria d&#8217;excel\u00b7l\u00e8ncia a la frontera entre l&#8217;enginyeria i les ci\u00e8ncies de la vida per a generar coneixement, integrant camps com la nanomedicina, la biof\u00edsica, la biotecnologia, l&#8217;enginyeria de teixits i les aplicacions de les tecnologies de la informaci\u00f3 en el camp de la salut.
