L&#8217;estudi, que va utilitzar organoides de tumors a partir de c\u00e8l\u00b7lules de pacients, es va centrar en les c\u00e8l\u00b7lules mare canceroses que expressen la prote\u00efna LGR5, un marcador clau en la senyalitzaci\u00f3 necess\u00e0ria per a la renovaci\u00f3 i diferenciaci\u00f3 de c\u00e8l\u00b7lules mare intestinals i canceroses. Els resultats van revelar difer\u00e8ncies significatives en les propietats mec\u00e0niques entre les c\u00e8l\u00b7lules que expressen LGR5 (LGR5+) i les que no ho fan (LGR5-).<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>This transition between these two cell types that work together is an adaptive mechanism that helps them withstand the physical changes in the microenvironment and facilitates cancer progression. <\/p>\n<cite>Xavier Trepat<\/cite><\/blockquote>\n\n\n\n<p>M\u00e9s concretament, els investigadors van observar que les c\u00e8l\u00b7lules que no expressen LGR5 s\u00f3n m\u00e9s toves, menys adhesives i es mouen m\u00e9s r\u00e0pidament, cosa que les fa m\u00e9s aptes per moure&#8217;s des del tumor principal. En contrast, les c\u00e8l\u00b7lules que s\u00ed que expressen LGR5 s\u00f3n m\u00e9s aptes per adherir-se a les parets dels vasos sanguinis i formar bretxes que els permeten envair altres teixits, cosa que facilitaria el seu creixement en nous llocs del cos, podent formar met\u00e0stasi.<\/p>\n\n\n\n<p>&#8220;Aquesta transici\u00f3 entre aquests dos tipus cel\u00b7lulars, que treballen en equip, \u00e9s un mecanisme adaptatiu que les ajuda a suportar els canvis f\u00edsics del microambient i facilita la progressi\u00f3 del c\u00e0ncer.&#8221; Explica Xavier Trepat, professor de recerca ICREA a l&#8217;IBEC.<\/p>\n\n\n\n<p>El treball, publicat a la prestigiosa revista cient\u00edfica <em>Nature Communications<\/em>, demostra que aquestes difer\u00e8ncies en les propietats mec\u00e0niques de les c\u00e8l\u00b7lules es deuen a la disminuci\u00f3 de les prote\u00efnes ERM (ezrina, radixina i moesina) que ocorre en les c\u00e8l\u00b7lules LGR5+. Aquestes prote\u00efnes connecten la membrana cel\u00b7lular amb l&#8217;esquelet intern de la c\u00e8l\u00b7lula, afectant la seva rigidesa i adhesi\u00f3. Manipulant aquestes prote\u00efnes, l&#8217;equip d&#8217;investigaci\u00f3 va poder fer que les c\u00e8l\u00b7lules LGR5+ i LGR5- es comportessin de manera similar, confirmant el paper crucial de les ERM en les difer\u00e8ncies mec\u00e0niques observades.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>Els nostres resultats demostren el potencial de les prote\u00efnes ERM com a objectiu terap\u00e8utic per interferir en el proc\u00e9s metast\u00e0sic del c\u00e0ncer colorectal.<\/p>\n<cite>Sefora Conti<\/cite><\/blockquote>\n\n\n\n<p>Tal com detalla Sefora Conti, investigadora de l&#8217;IBEC en el grup de Trepat i primera autora de l&#8217;estudi, van dur a terme diversos experiments per comprovar l&#8217;acci\u00f3 d&#8217;ERM:<\/p>\n\n\n\n<p>&#8220;Afegim a les c\u00e8l\u00b7lules LGR5+ una prote\u00efna sint\u00e8tica que imita la funci\u00f3 d&#8217;ERM i observem que llavors es tornaven indistingibles mec\u00e0nicament de les c\u00e8l\u00b7lules LGR5-. D&#8217;altra banda, redu\u00efm l&#8217;expressi\u00f3 de les prote\u00efnes ERM en c\u00e8l\u00b7lules LGR5- i de nou observem que les seves difer\u00e8ncies mec\u00e0niques respecte a LGR5+ es redu\u00efen. Aquests resultats demostren el potencial de les prote\u00efnes ERM com a objectiu terap\u00e8utic per interferir en el proc\u00e9s metast\u00e0sic del c\u00e0ncer colorectal.&#8221;<\/p>\n\n\n\n<p>En col\u00b7laboraci\u00f3 amb el grup de l&#8217;Institut de Recerca Biom\u00e8dica de Barcelona (IRB) liderat per Eduard Batlle, els investigadors tamb\u00e9 van analitzar els patrons d&#8217;expressi\u00f3 de les prote\u00efnes ERM en c\u00e8l\u00b7lules individuals d&#8217;una cohort de pacients, mostrant que aquesta reducci\u00f3 en les ERM \u00e9s una caracter\u00edstica comuna en els tumors colorectals.<\/p>\n\n\n\n<p>Aquests descobriments podrien contribuir al desenvolupament de noves estrat\u00e8gies per tractar i prevenir la met\u00e0stasi en el c\u00e0ncer colorectal, que \u00e9s el tercer tipus de c\u00e0ncer m\u00e9s com\u00fa a tot el m\u00f3n i la segona causa principal de morts relacionades amb el c\u00e0ncer.<\/p>\n\n\n\n<p>Xavier Trepat, l\u00edder de la recerca, tamb\u00e9 \u00e9s professor a la Universitat de Barcelona (UB) i membre del Centre de Recerca Biom\u00e8dica en Xarxa en Bioenginyeria, Biomaterials i Nanomedicina (CIBER-BBN).
