Aquests &#8220;assembloides&#8221; es creen mitjan\u00e7ant la uni\u00f3 d&#8217;organoides de rony\u00f3 en 3D amb organoides endotelials.&nbsp;<\/p>\n\n\n\n<p>Els organoides s\u00f3n estructures tridimensionals cultivades <em>in vitro<\/em> que imiten les funcions dels \u00f2rgans i s\u00f3n una eina prometedora per al modelatge de malalties i el cribratge de f\u00e0rmacs. Tanmateix, aquests petits \u00f2rgans sovint no tenen sistemes nervi\u00f3s, immunol\u00f2gic o vascular a causa de la seva complexitat.&nbsp;<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>Quan aquests dos tipus d&#8217;organoides estan junts, inicien un proc\u00e9s d&#8217;interacci\u00f3 en a penes tres hores. El resultat d&#8217;aquest procediment \u00e9s veritablement espectacular.<\/p>\n<cite>N\u00faria Montserrat Pulido<\/cite><\/blockquote>\n\n\n\n<p>Reproduir al laboratori el complex sistema de vascularitzaci\u00f3 dels ronyons, un teixit altament irrigat, ha estat un desafiament per als cient\u00edfics durant m\u00e9s d&#8217;una d\u00e8cada. Ara, aquest treball publicat a la prestigiosa revista cient\u00edfica <a href=\"https:\/\/onlinelibrary.wiley.com\/doi\/10.1002\/adma.202400306\" target=\"_blank\" rel=\"noreferrer noopener\"><em>Advanced Materials<\/em><\/a>, descriu una aproximaci\u00f3 que no s&#8217;havia provat abans:&nbsp;<\/p>\n\n\n\n<p>&#8220;Hem acoblat organoides de rony\u00f3 amb organoides vasculars. Per dur a terme amb \u00e8xit aquest proc\u00e9s, \u00e9s essencial identificar el moment idoni per unir-los, tenint en compte el moment de desenvolupament de cadascun dels organoides&#8221;, explica N\u00faria Montserrat Pulido, professora de recerca ICREA, investigadora principal de l&#8217;IBEC i l\u00edder de l&#8217;estudi. &#8220;Quan aquests dos tipus d&#8217;organoides estan junts, inicien un proc\u00e9s d&#8217;interacci\u00f3 en a penes tres hores. El resultat d&#8217;aquest procediment \u00e9s veritablement espectacular&#8221;, destaca.&nbsp;<\/p>\n\n\n\n<p>Aquestes troballes no nom\u00e9s proporcionen informaci\u00f3 valuosa sobre el desenvolupament dels organoides renals, sin\u00f3 que tamb\u00e9 estableixin les bases per dissenyar nous procediments de vascularitzaci\u00f3 d&#8217;aquests organoides. Aix\u00f2 podria tenir aplicacions en trasplantaments d&#8217;organoides en entorns cl\u00ednics i en investigacions sobre disfuncions vasculars en malalties humanes.&nbsp;&nbsp;<\/p>\n\n\n\n<p>&#8220;Necessitem tenir aquest sistema vascular en els nostres organoides si volem modelar patologies sist\u00e8miques que afecten el rony\u00f3 a trav\u00e9s del torrent sanguini, com pot ser una malaltia autoimmune o una diabetis, per exemple. Creiem, a m\u00e9s, que l&#8217;abordatge que hem utilitzat en aquest estudi es podria aplicar tamb\u00e9 a altres models d&#8217;organoide.&#8221;, detalla Elena Garreta Bahima, investigadora al grup de Montserrat a l&#8217;IBEC i primera autora de l&#8217;estudi.&nbsp;<\/p>\n\n\n\n<p>El seg\u00fcent pas, comenta Montserrat, ser\u00e0 introduir aquests organoides vascularitzats en un xip microflu\u00eddic per poder fer estudis encara m\u00e9s especialitzats per tal de mimetitzar condicions que s&#8217;esdevenen en malalties complexes aix\u00ed com permetre la connexi\u00f3 vascular amb altres organoides, com els organoides card\u00edacs, treball que avui dia estan desenvolupant al laboratori a IBEC.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\"><strong>Imprimir amb c\u00e8l\u00b7lules<\/strong>\u00a0<\/h4>\n\n\n\n<p>L&#8217;equip d&#8217;investigaci\u00f3 ja havia vist en estudis anteriors que els organoides de rony\u00f3 que generaven contenien un component vascular: c\u00e8l\u00b7lules d&#8217;endoteli, que s\u00f3n les que acaben donant lloc als vasos sanguinis. Utilitzant teixit renal d&#8217;origen porc\u00ed o hum\u00e0, van crear un hidrogel biocompatible, que podria ser utilitzat com a tinta per a impressi\u00f3 3D.\u00a0<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>Necessitem tenir aquest sistema vascular en els nostres organoides si volem modelar patologies sist\u00e8miques que afecten el rony\u00f3 a trav\u00e9s del torrent sanguini, com pot ser una malaltia autoimmune o una diabetis, per exemple.<\/p>\n<cite>Elena Garreta Bahima<\/cite><\/blockquote>\n\n\n\n<p>Amb aquesta base, van decidir simplificar el sistema per fer-lo m\u00e9s accessible i econ\u00f2mic. D&#8217;entre totes les prote\u00efnes que conformen l&#8217;hidrogel, van seleccionar el col\u00b7lagen I, una de les prote\u00efnes m\u00e9s abundants en la matriu extracel\u00b7lular del teixit renal, i van generar un gel biocompatible amb el qual van validar els seus resultats experimentals.&nbsp;<\/p>\n\n\n\n<p>Aquest treball representa una fita significativa en el camp de les ter\u00e0pies emergents i avan\u00e7ades, una de les principals \u00e0rees de recerca de l&#8217;IBEC. Aquestes ter\u00e0pies s\u00f3n tractaments m\u00e8dics innovadors que empren tecnologies d&#8217;avantguarda per abordar malalties i condicions complexes.&nbsp;<\/p>\n\n\n\n<p>L&#8217;estudi a m\u00e9s ha aglutinat investigadors d&#8217;altres centres que formen part de la Plataforma Nacional de Biomodals i Biobancs de l&#8217;Institut de Salut Carles III (PISCIII-BB), posant en relleu la import\u00e0ncia de la col\u00b7laboraci\u00f3 en xarxa per a la definici\u00f3 de solucions en biomedicina basades en bioenginyeria.
