L\u2019estudi, publicat a la revista <em>Nature Biomedical Engineering<\/em>, representa una fita en el camp de la medicina regenerativa i personalitzada, establint les bases per a la utilitzaci\u00f3 d\u2019organoides &nbsp;de rony\u00f3 derivats de c\u00e8l\u00b7lules mare humanes per a ter\u00e0pia cel\u00b7lular en assajos cl\u00ednics.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p><em>L\u2019objectiu a llarg termini \u00e9s poder regenerar o reparar un \u00f2rgan abans del trasplantament. Aix\u00f2 podria reduir el temps d\u2019espera dels pacients cr\u00f2nics i augmentar el nombre d\u2019\u00f2rgans viables per al trasplantament<\/em>.<\/p>\n\n\n\n<p>N\u00faria Montserrat<\/p>\n<\/blockquote>\n\n\n\n<p>Aquest treball, liderat per la <strong>Dra. N\u00faria Montserrat,<\/strong> investigadora ICREA a l\u2019IBEC en el moment de l\u2019estudi i actual consellera del Departament de Recerca i Universitats de la Generalitat de Catalunya, \u00e9s el resultat d\u2019una traject\u00f2ria cient\u00edfica de m\u00e9s d\u2019una d\u00e8cada dedicada a la medicina regenerativa i a la bioenginyeria d\u2019\u00f2rgans. Fruit d\u2019aquesta l\u00ednia de recerca sostinguda, l\u2019equip d\u2019investigaci\u00f3 ha aconseguit combinar per primera vegada organoides renals humans amb ronyons porcins vius, connectats a m\u00e0quines de perfusi\u00f3 normot\u00e8rmica. L\u2019\u00fas d\u2019aquests dispositius, utilitzats habitualment en el quir\u00f2fan per mantenir l\u2019\u00f2rgan viu i oxigenat fora del cos abans d\u2019un transplantament, ha perm\u00e8s inserir organoides humans dins els ronyons porcins i monitorar en temps real la seva integraci\u00f3 i funci\u00f3.<\/p>\n\n\n\n<p>\u201c<em>La nostra recerca demostra que la combinaci\u00f3 de tecnologies d\u2019organoides i perfusi\u00f3 ex vivo pot permetre intervencions cel\u00b7lulars en condicions totalment controlades.<\/em>\u201d explica la Dra. Montserrat. \u201c<em>L\u2019objectiu a llarg termini \u00e9s poder regenerar o reparar un \u00f2rgan abans del trasplantament. Aix\u00f2 podria reduir el temps d\u2019espera dels pacients cr\u00f2nics i augmentar el nombre d\u2019\u00f2rgans viables per al trasplantament<\/em>.\u201d<strong><br><\/strong><\/p>\n\n\n\n<h4 class=\"wp-block-heading\"><strong>Una tecnologia escalable i segura<\/strong><\/h4>\n\n\n\n<p>Un organoide de rony\u00f3 \u00e9s una estructura tridimensional, d\u2019unes micres de mida, cultivada al laboratori a partir de c\u00e8l\u00b7lules mare humanes. Tot i que no \u00e9s un \u00f2rgan complet, reprodueix moltes de les seves estructures i funcions principals. Gr\u00e0cies a aquestes caracter\u00edstiques, els organoides permeten estudiar com es desenvolupa el rony\u00f3, provar nous f\u00e0rmacs i, en el futur, s\u2019espera que puguin ser utilitzats per a reparar teixits renals danyats o millorar \u00f2rgans destinats al trasplantament.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p><em><em>Ara, amb el nostre nou m\u00e8tode, podem generar milers d\u2019organoides renals en condicions controlades i en poc temps, amb una gran precisi\u00f3 i sense necessitat de components complexos. Aix\u00f2 obre la porta a aplicacions com la detecci\u00f3 de f\u00e0rmacs o l\u2019estudi de malalties.<\/em><\/em><\/p>\n\n\n\n<p>Elena Garreta<\/p>\n<\/blockquote>\n\n\n\n<p>L\u2019estudi descriu, per primera vegada, un m\u00e8tode sistem\u00e0tic i escalable per produir milers d\u2019aquests organoides renals humans, utilitzant t\u00e8cniques de microagregaci\u00f3 i enginyeria gen\u00e8tica.<\/p>\n\n\n\n<p><em>&#8220;Tot i el gran potencial cl\u00ednic dels organoides, fins ara un dels grans reptes per aplicar aquesta tecnologia en tractaments m\u00e8dics reals era aconseguir produir aquests organoides de manera escalable, uniforme i assequible. Ara, amb el nostre nou m\u00e8tode, podem generar milers d\u2019organoides renals en condicions controlades i en poc temps, amb una gran precisi\u00f3 i sense necessitat de components complexos. Aix\u00f2 obre la porta a aplicacions com la detecci\u00f3 de f\u00e0rmacs o l\u2019estudi de malalties.&#8221;, <\/em>afegeix la <strong>Dra. Elena Garreta<\/strong>, investigadora s\u00e8nior del grup de <a href=\"https:\/\/ibecbarcelona.eu\/ipscs\/\">Puripot\u00e8ncia per a la regeneraci\u00f3 d\u2019\u00f2rgans<\/a> de l\u2019IBEC i co-primera autora de l\u2019estudi.<\/p>\n\n\n\n<p>D\u2019altra banda, la utilitzaci\u00f3 de les m\u00e0quines mencionades, per portar a terme la perfusi\u00f3 dels organoides dins els ronyons, ofereix un avantatge clau: permet mesurar par\u00e0metres fisiol\u00f2gics de l\u2019\u00f2rgan en temps real i detectar de manera immediata qualsevol signe de dany o rebuig. Els experiments es van realitzar tant <em>ex vivo<\/em> (fora de l\u2019organisme) com <em>in vivo<\/em> (en el mateix animal), utilitzant un model porc\u00ed de trasplantament molt similar al rony\u00f3 hum\u00e0.<\/p>\n\n\n\n<p>L\u2019equip de recerca va observar que, despr\u00e9s de 24 i 48 hores del trasplantament, els organoides humans persistien integrats en el teixit renal porc\u00ed, mantenien la seva viabilitat i no desencadenaven cap resposta immune significativa. El rony\u00f3 trasplantat continuava funcionant amb normalitat, i no es van detectar signes de dany ni toxicitat.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\"><strong>Cap a la regeneraci\u00f3 d\u2019\u00f2rgans abans del trasplantament<\/strong><\/h4>\n\n\n\n<p>Segons els autors i les autores, aquesta metodologia permet imaginar un escenari cl\u00ednic en qu\u00e8 els \u00f2rgans destinats al trasplantament puguin ser tractats i condicionats abans d\u2019implantar-los.
