Els m\u00e8todes tradicionals per al seguiment del metabolisme solen basar-se en assajos no integrats al proc\u00e9s experimental o en instruments sofisticats d&#8217;alt camp, que s\u00f3n costosos i requereixen grans volums de mostra. La\u00a0espectroscop\u00eda\u00a0de Resson\u00e0ncia Magn\u00e8tica Nuclear (RMN), amb la seva capacitat inigualable per a diferenciar esp\u00e8cies qu\u00edmiques de forma no invasiva, ha prom\u00e8s durant molt de temps una \u00e0mplia comprensi\u00f3 dels processos bioqu\u00edmics, per\u00f2 la seva baixa sensibilitat inherent ha limitat el seu \u00fas en contextos a microescala. Aquest desafiament \u00e9s especialment significatiu en plataformes microflu\u00eddiques, on els volums de mostra s\u00f3n m\u00ednims i els processos metab\u00f2lics evolucionen din\u00e0micament.<br><br>L&#8217;estudi recentment publicat en la revista <em>Analytical Chemistry<\/em> presenta un espectr\u00f2metre de\u00a0RMN\u00a0de sobretaula adaptat per a la dissoluci\u00f3 de Polaritzaci\u00f3 Nuclear Din\u00e0mica (dDNP), una t\u00e8cnica de\u00a0hiperpolaritzaci\u00f3 que potencia la polaritzaci\u00f3 d&#8217;esp\u00edn nuclear molt m\u00e9s all\u00e0 de l&#8217;equilibri t\u00e8rmic, incrementant la intensitat del senyal de\u00a0RMN\u00a0en diversos ordres de magnitud. Aprofitant aquesta millora, els autors van poder superar les limitacions cl\u00e0ssiques de sensibilitat del maquinari compacte de\u00a0RMN\u00a0i detectar canvis metab\u00f2lics en temps real en cultius cel\u00b7lulars que flueixen a trav\u00e9s de dispositius microflu\u00eddics personalitzats. El desenvolupament d&#8217;aquesta nova tecnologia va ser el principal objectiu del <a href=\"https:\/\/blocproject.eu\/\">BLOC:\u00a0RMN\u00a0de laboratori per a l&#8217;a-xip en laboratori<\/a>, un projecte europeu realitzat de l&#8217;1 de gener de 2020 al 31 de desembre de 2023, coordinat per Irene Marco-Rius, investigadora principal del <a href=\"https:\/\/ibecbarcelona.eu\/molecular-imaging-for-precision-medicine\/\">grup d&#8217;Imatge Molecular per a Medicina de Precisi\u00f3<\/a> en l&#8217;Institut de\u00a0Bioinginyeria\u00a0de Catalunya (IBEC).<br><br>La integraci\u00f3 del\u00a0RMN\u00a0de sobretaula amb microflu\u00eddica va implicar una enginyeria acurada del sistema de detecci\u00f3 i el maneig de fluids, permetent observar metab\u00f2lits\u00a0hiperpolaritzats\u00a0introdu\u00efts a l&#8217;entorn de cultiu cel\u00b7lular mentre eren absorbits i transformats per les c\u00e8l\u00b7lules. Aquest enfocament proporciona dades cin\u00e8tiques sobre reaccions metab\u00f2liques sense interrompre el cultiu ni requerir grans poblacions cel\u00b7lulars, un avantatge clau per a estudis de mostres biol\u00f2giques rares o valuoses. Els investigadors van demostrar que era possible observar cont\u00ednuament les conversions metab\u00f2liques crucials, la qual cosa oferia una visi\u00f3 detallada de l&#8217;estat bioqu\u00edmic de les c\u00e8l\u00b7lules estudiades.<br><br>Els resultats suposen una fita per a la\u00a0metabol\u00f3mica\u00a0en un xip: una plataforma compacta i rendible de\u00a0RMN\u00a0capa\u00e7 de permetre l&#8217;observaci\u00f3 directa de fluxos metab\u00f2lics din\u00e0mics en sistemes microflu\u00eddics. Aprofitant la\u00a0hiperpolaritzaci\u00f3, la tecnologia tanca la bretxa entre el rendiment en\u00a0RMN\u00a0d&#8217;alt camp i les necessitats pr\u00e0ctiques de l&#8217;an\u00e0lisi de laboratori en xip, <strong>declara Marc\u00a0Azagra, primer autor de l&#8217;estudi.<br><\/strong><br>De cara al futur, aquesta recerca obre perspectives prometedores per a una \u00e0mplia gamma d&#8217;aplicacions en ci\u00e8ncies de la vida, descobriment de f\u00e0rmacs i medicina de precisi\u00f3.
