Mitjan\u00e7ant l&#8217;estudi de sistemes multicel\u00b7lulars que abasten des del desenvolupament fisiol\u00f2gic fins a la progressi\u00f3 patol\u00f2gica, ALIVE buscar\u00e0 establir un nou marc conceptual per comprendre com es generen i mantenen les funcions biol\u00f2giques col\u00b7lectives.<\/em><\/figcaption><\/figure>\n\n\n\n<p>Al voltant de 100 equips de recerca de tot Europa van competir per l&#8217;ajuda cient\u00edfica m\u00e9s prestigiosa de la Fundaci\u00f3 Novo Nordisk. El finan\u00e7ament ha estat concedit a un consorci liderat per <strong>Amin Doostmohammadi<\/strong>, de la Niels Bohr International Academy, juntament amb tres socis de refer\u00e8ncia internacional: <strong>Xavier Trepat<\/strong>, <strong>de l&#8217;Institut de Bioenginyeria de Catalunya <\/strong>(IBEC) de Barcelona; <strong>Nikta Fakhri<\/strong>, del <strong>Massachusetts Institute of Technology<\/strong> (MIT); i <strong>Erwin Frey<\/strong>, de la <strong>Ludwig Maximilian University<\/strong> de M\u00fanic.<\/p>\n\n\n\n<p><em>&#8220;Estic molt il\u00b7lusionat per iniciar una nova col\u00b7laboraci\u00f3 amb investigadors i investigadores l\u00edders mundials d&#8217;aquest consorci, tant en l&#8217;\u00e0mbit te\u00f2ric com en l&#8217;experimental&#8221;<\/em>, afirma Trepat. <em>&#8220;L&#8217;estudi dels fluxos d&#8217;informaci\u00f3 en sistemes fora de l&#8217;equilibri representa un gran repte per a f\u00edsics i bi\u00f2legs, i estic conven\u00e7ut que aquest finan\u00e7ament ens permetr\u00e0 desenvolupar un marc general per comprendre aquest problema&#8221;.<\/em><\/p>\n\n\n\n<h4 class=\"wp-block-heading\"><strong><strong>El corrent unidireccional de la vida<\/strong><\/strong><\/h4>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p><em>The topic of information flows in non-equilibrium systems is a great challenge for physicists and biologists, and I am quite convinced that this grant will allow us to develop a general framework to understand this problem<\/em>.<\/p>\n\n\n\n<p>Xavier Trepat<\/p>\n<\/blockquote>\n\n\n\n<p>Cada c\u00e8l\u00b7lula del nostre organisme est\u00e0 constantment fent alguna cosa: tira, empeny, envia senyals a les seves ve\u00efnes o adopta un determinat dest\u00ed cel\u00b7lular. D&#8217;alguna manera, a partir de tota aquesta activitat local, emergeixen teixits organitzats. Sense l&#8217;exist\u00e8ncia d&#8217;un controlador central, es formen \u00f2rgans, cicatritzen ferides i els tumors envaeixen teixits. Com passa aix\u00f2? Aquesta \u00e9s la pregunta que ALIVE es proposa respondre.<\/p>\n\n\n\n<p>La idea central del projecte \u00e9s que els teixits vius s\u00f3n sistemes que processen informaci\u00f3. Les forces que exerceixen les c\u00e8l\u00b7lules, els senyals que envien i les identitats que adopten contenen informaci\u00f3, i aquesta informaci\u00f3 flueix en una direcci\u00f3 espec\u00edfica al llarg del temps. De la mateixa manera que un riu t\u00e9 un corrent, la din\u00e0mica dels teixits vius t\u00e9 una direcci\u00f3 temporal preferent: les causes precedeixen als efectes de manera quantificable. Els f\u00edsics denominen a aquest fenomen la <strong>fletxa del temps<\/strong>. ALIVE convertir\u00e0 aquesta fletxa col\u00b7lectiva del temps en una eina quantitativa per comprendre i, en \u00faltima inst\u00e0ncia, controlar el comportament dels teixits.<\/p>\n\n\n\n<p><em>&#8220;El flux d&#8217;informaci\u00f3 dins dels sistemes vius, i el fet que aquest flux tingui una direcci\u00f3, \u00e9s la idea que es troba al cor del nostre enfocament&#8221;,<\/em> explica Doostmohammadi. <em>&#8220;Si som capa\u00e7os de mesurar i cartografiar aquest flux, podrem comen\u00e7ar a predir quan un teixit funcionar\u00e0 amb normalitat, quan fallar\u00e0 i com podr\u00edem intervenir&#8221;.<\/em><\/p>\n\n\n\n<h4 class=\"wp-block-heading\"><strong><strong>Quatre sistemes, des d&#8217;esponges marines fins a embrioides humans<\/strong><\/strong><\/h4>\n\n\n\n<p>El consorci estudiar\u00e0 quatre sistemes biol\u00f2gics que abasten diferents formes de vida multicel\u00b7lular: les <strong>esponges marines<\/strong>, que representen els or\u00edgens m\u00e9s primerencs de la multicel\u00b7lularitat animal; els <strong>organoides intestinals<\/strong>, com a model de renovaci\u00f3 tissular saludable; els <strong>organoides de c\u00e0ncer colorrectal<\/strong>, on es trenca l&#8217;organitzaci\u00f3 col\u00b7lectiva; i els <strong>embrioides humans<\/strong> derivats de c\u00e8l\u00b7lules mare pluripotents indu\u00efdes, en els quals l&#8217;aparici\u00f3 del pla corporal pot observar-se i controlar-se experimentalment.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p><em>If we can measure and map the <em>flow of information within living systems<\/em>, we can begin to predict when tissues will function normally, when they will fail, and how we might intervene<\/em>.<\/p>\n\n\n\n<p>Amin Doostmohammadi<\/p>\n<\/blockquote>\n\n\n\n<p>En tots ells, l&#8217;equip combinar\u00e0 mesuraments de forces mec\u00e0niques, microsc\u00f2pia en viu, eines moleculars i nous marcs te\u00f2rics procedents de la f\u00edsica fora de l&#8217;equilibri per extreure, modelitzar i, en \u00faltima inst\u00e0ncia, dirigir els fluxos d&#8217;informaci\u00f3 que governen l&#8217;organitzaci\u00f3 dels teixits.<\/p>\n\n\n\n<p>A l&#8217;IBEC, el grup de <a href=\"https:\/\/ibecbarcelona.eu\/research-groups\/integrative-cell-and-tissue-dynamics\/\">Din\u00e0mica Integrativa de C\u00e8l\u00b7lules i Teixits<\/a>, liderat pel professor d\u2019investigaci\u00f3 ICREA <strong>Xavier Trepat<\/strong>, exercir\u00e0 un paper fonamental en el desenvolupament de les plataformes experimentals que permetran mesurar directament els fluxos d&#8217;informaci\u00f3 en sistemes multicel\u00b7lulars. Aprofitant la seva reconeguda experi\u00e8ncia internacional en mecanobiologia, el grup combinar\u00e0 cartografiat de forces, pertorbacions optogen\u00e8tiques i avan\u00e7ades estrat\u00e8gies d&#8217;enginyeria tissular en 3D per estudiar com es transmet la informaci\u00f3 a trav\u00e9s d&#8217;organoides, embrioides i teixits artificials. Mitjan\u00e7ant la generaci\u00f3 de conjunts de dades a gran escala i alta qualitat que integrin senyals mec\u00e0nics i bioqu\u00edmics, l&#8217;equip treballar\u00e0 estretament amb els te\u00f2rics del consorci per posar a prova nous marcs conceptuals destinats a comprendre l&#8217;organitzaci\u00f3 dels teixits, la invasi\u00f3 tumoral i la regeneraci\u00f3.<\/p>\n\n\n\n<p><em>&#8220;Abordar el problema dels fluxos d&#8217;informaci\u00f3 en sistemes multicel\u00b7lulars ens ajudar\u00e0 a comprendre processos fonamentals del desenvolupament, la regeneraci\u00f3 i la malaltia&#8221;<\/em>, afirma Trepat.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\"><strong><strong>Un programa de sis anys orientat al descobriment<\/strong><\/strong><\/h4>\n\n\n\n<p>ALIVE tindr\u00e0 una durada de sis anys i permetr\u00e0 incorporar un important nombre d&#8217;estudiants de doctorat i investigadors i investigadores postdoctorals en les quatre institucions participants.
