Utilitzant microsc\u00f2pia avan\u00e7ada en temps real, models fets amb enginyeria de teixits i simulacions computacionals, l&#8217;equip va descobrir que aquest proc\u00e9s acaba empenyent el nucli de la c\u00e8l\u00b7lula fora de la g\u00e0bia de queratina que l&#8217;envolta. Es pot observar clarament que, en les c\u00e8l\u00b7lules estirades, els filaments individuals de queratina es reorganitzen gradualment en gruixos feixos en forma d&#8217;estrella que s&#8217;estenen entre c\u00e8l\u00b7lules ve\u00efnes, formant xarxes\u00a0supracel\u00b7lulars al llarg del teixit.<\/figcaption><\/figure>\n\n\n\n<p>Com se les arreglen els teixits per a suportar un estirament durant hores o fins i tot dies? Un estudi dirigit per investigadors de <strong>l&#8217;Institut de&nbsp;Bioingenyeria&nbsp;de Catalunya (IBEC)<\/strong> ha descobert una resposta inesperada: les c\u00e8l\u00b7lules reorganitzen radicalment la seva estructura interna i, en el proc\u00e9s, alliberen els seus nuclis d&#8217;una xarxa protectora de queratina. El treball, publicat en <a href=\"https:\/\/doi.org\/10.1038\/s41567-026-03371-8\"><em>Nature&nbsp;Physics<\/em><\/a>, revela una nova forma en la qual els teixits s&#8217;adapten a forces mec\u00e0niques prolongades.<br><br>La queratina \u00e9s un dels principals components estructurals del citoesquelet, la xarxa de filaments proteics que confereix a les c\u00e8l\u00b7lules la seva forma i la seva resist\u00e8ncia mec\u00e0nica. Encara que des de fa temps se sap que la queratina protegeix els teixits de grans deformacions, no quedava clar com respon exactament a l&#8217;estirament prolongat.<\/p>\n\n\n\n<p>Per a investigar aquesta q\u00fcesti\u00f3, els investigadors van combinar teixits epitelials modificats gen\u00e8ticament, un dispositiu microflu\u00eddic d&#8217;estirament dissenyat a mesura, microsc\u00f2pia avan\u00e7ada en temps real i modelitzaci\u00f3 computacional a m\u00faltiples escales. Van observar que la queratina no reacciona de manera immediata quan s&#8217;estiren els teixits. En canvi, al llarg de v\u00e0ries hores, els filaments individuals es reorganitzen gradualment en gruixos feixos en forma d&#8217;estrella que s&#8217;estenen entre c\u00e8l\u00b7lules ve\u00efnes, formant xarxes&nbsp;supracel\u00b7lulars al llarg del teixit.<br><br>\u00ab<em>La simulaci\u00f3 de la din\u00e0mica intracel\u00b7lular va revelar que el mateix proc\u00e9s que forma aquests cridaners feixos tamb\u00e9 genera forces que, en \u00faltima inst\u00e0ncia, empenyen al nucli fora de la seva g\u00e0bia<\/em>\u00bb, afirma <strong>Marco&nbsp;Pensalfini<\/strong>, professor adjunt de la <strong>Universitat Queen Mary de Londres, qui va desenvolupar el model computacional juntament amb<\/strong> <strong>Marino Arroyo<\/strong>, catedr\u00e0tic del Departament d&#8217;Enginyeria Civil i Mediambiental de la UPC i investigador principal del grup de recerca \u00ab <a href=\"https:\/\/cimne.com\/ca\/recerca\/clusters-de-recerca\/mecanica-computacional-enginyeria-medica-materia-viva\/interficies-materials-tous-vius\/\">Interf\u00edcies de materials tous i vius<\/a>\u00bb del&nbsp;CIMNE. \u00ab<em>La modelitzaci\u00f3 computacional ens va permetre relacionar el que observem en els experiments amb els mecanismes f\u00edsics subjacents<\/em>\u00bb.<\/p>\n\n\n\n<p>La reorganitzaci\u00f3 estructural comen\u00e7a en les unions on es troben tres c\u00e8l\u00b7lules. Els filaments de queratina desapareixen progressivament d&#8217;aquestes regions i s&#8217;acumulen formant feixos cada vegada m\u00e9s gruixuts. Sorprenentment, el proc\u00e9s \u00e9s col\u00b7lectiu: comen\u00e7a en unes poques c\u00e8l\u00b7lules a\u00efllades i despr\u00e9s es propaga a les seves ve\u00efnes, creant \u00e0rees cada vegada m\u00e9s extenses de c\u00e8l\u00b7lules agrupades en feixos, connectades per una xarxa compartida de queratina.<\/p>\n\n\n\n<p>Les imatges de microsc\u00f2pia en temps real van confirmar aquesta predicci\u00f3: a mesura que els feixos es feien m\u00e9s gruixuts, el nucli es desprenia gradualment de la malla de queratina que l&#8217;envoltava fins a separar-se per complet, quedant unit \u00fanicament a una quantitat residual de filaments. Aquest \u00aballiberament del nucli\u00bb va resultar ser una de les troballes m\u00e9s cridaneres de l&#8217;estudi, estretament vinculat a la pr\u00f2pia formaci\u00f3 dels feixos de queratina.<\/p>\n\n\n\n<p>\u00ab<em>Una de les preguntes m\u00e9s intrigants que planteja el nostre estudi \u00e9s si aquest proc\u00e9s d&#8217;alliberament protegeix en \u00faltima inst\u00e0ncia al nucli o el fa m\u00e9s vulnerable<\/em>\u00bb, afirma <strong>Xavier&nbsp;Trepat<\/strong>, professor de recerca ICREA a l\u2019IBEC, l\u00edder del grup de&nbsp;<a href=\"https:\/\/ibecbarcelona.eu\/ca\/research-groups\/integrative-cell-and-tissue-dynamics\/\">Din\u00e0mica Integrativa de C\u00e8l\u00b7lules i Teixits<\/a>&nbsp;i col\u00edder de l\u2019estudi. \u00ab<em>D&#8217;una banda, l&#8217;eliminaci\u00f3 de la g\u00e0bia de queratina podria exposar al nucli de forma m\u00e9s directa a les forces mec\u00e0niques. D&#8217;altra banda, i encara que pugui semblar contradictori, la desconnexi\u00f3 del nucli d&#8217;una xarxa&nbsp;citoesquel\u00e9tica&nbsp;sotmesa a gran tensi\u00f3 podria, de fet, protegir-lo en impedir la transmissi\u00f3 de la for\u00e7a. Per a comprendre quin d&#8217;aquests efectes predomina, seran necessaris estudis addicionals que examinin com responen els nuclis a una tensi\u00f3 mec\u00e0nica prolongada<\/em>\u00bb.<br><br>Els investigadors tamb\u00e9 van identificar un paper important de l&#8217;actina, un altre component fonamental del citoesquelet. L&#8217;afebliment dels enlla\u00e7os moleculars entre l&#8217;actina i la queratina va accelerar el proc\u00e9s d&#8217;agrupaci\u00f3 gaireb\u00e9 tres vegades, la qual cosa demostra que les interaccions entre tots dos sistemes de filaments s\u00f3n reguladors clau de com els teixits s&#8217;adapten a l&#8217;estirament.<br><br>\u00abAquestes troballes poden ajudar-nos a comprendre millor com s&#8217;adapten els teixits a l&#8217;estirament prolongat en una \u00e0mplia varietat de contextos biol\u00f2gics\u00bb, afirma <strong>Tom&nbsp;Golde<\/strong>, investigador postdoctoral del grup de&nbsp;Trepat&nbsp;i primer autor de l&#8217;estudi. \u00abEs donen condicions mec\u00e0niques similars durant el desenvolupament embrionari i en \u00f2rgans que s&#8217;expandeixen i contreuen repetidament, com la bufeta o la gl\u00e0ndula mam\u00e0ria.
