Fins ara, l&#8217;expressi\u00f3 de prote\u00efnes en c\u00e8l\u00b7lules seleccionades s&#8217;activava mitjan\u00e7ant 2 fotons, per\u00f2 el seu \u00fas quedava limitat a c\u00e8l\u00b7lules properes a la superf\u00edcie a causa de l&#8217;elevada dispersi\u00f3 dels fotons vermells utilitzats. No obstant aix\u00f2, per investigar els \u00f2rgans interns durant el desenvolupament \u00e9s necessari, no solament mantenir la viabilitat de les c\u00e8l\u00b7lules marcades, sin\u00f3 poder accedir a aquestes quan es troben emmascarades en teixits m\u00e9s complexos o profunds.<\/p>\n<p>&#8220;Aquesta innovadora t\u00e8cnica, basada en la fotoactivaci\u00f3 de tres fotons de longitud d&#8217;ona m\u00e9s llarga, suposa una soluci\u00f3 al problema de la dispersi\u00f3 i, a m\u00e9s, \u00e9s capa\u00e7 d&#8217;activar les prote\u00efnes que es troben en les c\u00e8l\u00b7lules dels teixits m\u00e9s profunds sense interferir en l&#8217;esperan\u00e7a de vida de l&#8217;animal&#8221;, comenta Dobryna Zalvidea, responsable de l&#8217;estudi en l&#8217;Institut de Bioinginyeria de Catalunya (IBEC).<\/p>\n<p>Els investigadors van marcar les c\u00e8l\u00b7lules musculars del cor d&#8217;embrions de peix zebra i van descobrir que la prog\u00e8nie d&#8217;aquestes c\u00e8l\u00b7lules en peixos adults estava marcada de manera permanent sense causar cap dany remarcable als teixits. Aquesta metodologia, tamb\u00e9 pot estendre&#8217;s a altres teixits i models animals, aix\u00ed com ser d&#8217;utilitada per activar, amb resoluci\u00f3 espacial i temporal, l&#8217;expressi\u00f3 de diferents prote\u00efnes per controlar el comportament a llarg termini de c\u00e8l\u00b7lules individuals o de grups cel\u00b7lulars <em>in vivo<\/em>.<\/p>\n<p>El sistema proposat en aquest estudi basat en la fotoactivaci\u00f3 utilitza un inductor inactiu encapsulat (Cre-loxP) que penetra a l&#8217;interior de tot l&#8217;organisme per\u00f2 que solament es torna funcional quan s&#8217;activa mitjan\u00e7ant la llum. Una vegada activat, l&#8217;inductor \u00e9s capa\u00e7 de modificar llocs determinats de l&#8217;ADN de les c\u00e8l\u00b7lules de les quals es vol con\u00e8ixer el seu comportament. D&#8217;aquesta manera, es controla l&#8217;expressi\u00f3 de les prote\u00efnes i es poden dirigir els canvis a tipus cel\u00b7lulars espec\u00edfics utilitzant \u00fanicament un est\u00edmul extern com a desencadenant.
