Aix\u00ed es poden generar estructures de diversos components amb una gran precisi\u00f3 que s&#8217;utilitzen per a la creaci\u00f3 de superf\u00edcies sensores o en sistemes d&#8217;encaminament de medicaments, entre altres aplicacions. No obstant aix\u00f2, encara no s&#8217;ha aconseguit un m\u00e8tode per integrar de forma f\u00e0cil aquestes estructures d&#8217;ADN dins d&#8217;altres dispositius microelectr\u00f2nics.<\/p>\n<p>Ara, un treball liderat pel CSIC, amb la participaci\u00f3 de cient\u00edfics de l&#8217;Institut de Bioenginyeria de Catalunya (IBEC) i de la Universitat de Barcelona (UB), la majoria d&#8217;ells integrats en el Centre d&#8217;Investigaci\u00f3 Biom\u00e8dica en Xarxa de Bioenginyeria, Biomaterials i Nanomedicina (CIBER-BBN), i publicat a la revista Advanced Materials, ha aconseguit un important aven\u00e7 en aquesta l\u00ednia. L&#8217;equip, liderat per Ramon Eritja, professor d&#8217;investigaci\u00f3 del CSIC a l&#8217;Institut de Qu\u00edmica Avan\u00e7ada de Catalunya (IQAC), ha ideat un proc\u00e9s de litografia molecular per crear patrons en superf\u00edcies d&#8217;or. Aquest proc\u00e9s pot ser implementat en la creaci\u00f3 de petits circuits dins dels dispositius microelectr\u00f2nics.<\/p>\n<p><strong>Doblegar l&#8217;ADN com si fos paper<\/strong><br \/>\nEl m\u00e8tode es basa en l&#8217;\u00fas de la t\u00e8cnica de la papirofl\u00e8xia l&#8217;ADN (en angl\u00e8s DNA origami), en la qual es parteix d&#8217;una gran cadena d&#8217;ADN d&#8217;origen viral que es va plegant de manera natural gr\u00e0cies a altres petits segments d&#8217;ADN (oligonucle\u00f2tids) que actuen com si fossin grapes, fins a obtenir dissenys diversos. D&#8217;aqu\u00ed ve precisament el seu nom, pres del tradicional art oriental que crea figures plegant un full de paper en diferents formes.<\/p>\n<p>Les estructures obtingudes poden emprar-se com a plantilles per col\u00b7locar prote\u00efnes, nanopart\u00edcules, enzims o qualsevol altra mol\u00e8cula funcional seguint un patr\u00f3 predeterminat.<br \/>\nUsant aquesta t\u00e8cnica, els cient\u00edfics han fixat primer, en una superf\u00edcie d&#8217;or, un conjunt de mol\u00e8cules d&#8217;ADN d&#8217;uns 10 micr\u00f2metres quadrats que cont\u00e9 uns 250 segments d&#8217;ADN de forma ordenada. Entre cada un dels segments d&#8217;ADN hi ha una dist\u00e0ncia de pocs nan\u00f2metres.<\/p>\n<p>Per a la generaci\u00f3 del patr\u00f3 -en aquest cas, una l\u00ednia- s&#8217;han modificat 12 dels 250 oligonucle\u00f2tids grapa amb un compost qu\u00edmic (un grup tiol) que reacciona amb la superf\u00edcie d&#8217;or. No obstant aix\u00f2, es podria &#8220;dibuixar&#8221; qualsevol altra forma com a patr\u00f3. &#8220;D&#8217;aquesta forma, aquestes mol\u00e8cules marcades d&#8217;ADN i nom\u00e9s aquestes reaccionen quan s\u00f3n fixades sobre la superf\u00edcie d&#8217;or, que \u00e9s la base habitual dels microdispositius&#8221;, explica Ramon Eritja, cient\u00edfic del CSIC a l&#8217;Institut de Qu\u00edmica Avan\u00e7ada de Catalunya.<\/p>\n<p>Quan les mol\u00e8cules d&#8217;ADN marcades s&#8217;han unit a la superf\u00edcie d&#8217;or, s&#8217;eliminen les altres que no estan marcades, aix\u00ed com l&#8217;ADN viral. \u00c9s aix\u00ed com s&#8217;aconsegueix transferir a la superf\u00edcie d\u2019or el patr\u00f3 lineal d&#8217;oligonucle\u00f2tids, els qual, al seu torn, pot atraure i unir nanomaterials funcionalitzats amb cadenes complement\u00e0ries a les grapes d\u2019ADN.<\/p>\n<p>\u00c9s com crear un segell o un tamp\u00f3, que es pot integrar en els processos de fotolitografia que s&#8217;usen habitualment per a crear circuits microelectr\u00f2nics, i que permet &#8220;estampar&#8221; i integrar, sobre les superf\u00edcies d&#8217;or dels circuits, estructures o patrons d&#8217;ADN a una escala inferior als 10 nan\u00f2metres. Una cosa que fins ara no s&#8217;havia pogut aconseguir.<\/p>\n<p><strong>Aplicacions<\/strong><br \/>\nLa tecnologia actual permet sintetitzar i marcar centenars d&#8217;oligonucle\u00f2tids amb gran rapidesa, el que permetria traslladar aquest m\u00e8tode a escala industrial.<\/p>\n<p>Aquest m\u00e8tode permet l&#8217;obtenci\u00f3 de superf\u00edcies amb moltes mol\u00e8cules disposades de forma ordenada, seguint patrons moleculars de mida nanom\u00e8trica. Aquestes superf\u00edcies es podrien utilitzar per crear circuits m\u00e9s petits que els actuals (per exemple, llapis de mem\u00f2ria amb una capacitat 100 vegades m\u00e9s gran que els actuals) o superf\u00edcies sensores d&#8217;alta resoluci\u00f3. En aquesta \u00faltima aplicaci\u00f3, es podria aprofitar la petita grand\u00e0ria dels patrons obtinguts per incorporar de forma ordenada molts sensors en una superf\u00edcie molt petita.
