Es poden observar neurones corticals prim\u00e0ries despr\u00e9s de 15 dies de cultiu in vitro. b) Immunolocalitzaci\u00f3 de la versi\u00f3 maligna de l\u2019alfa-sinucle\u00efna (verda) en el dispositiu microflu\u00eddic despr\u00e9s de la inoculaci\u00f3 amb l\u2019alfa-sinucle\u00efna recombinant. c) Fotomicrografia mostrant exemples de neurones corticals adultes amb un contingut significatiu de sinucle\u00efna degenerada 45 dies despr\u00e9s de la injecci\u00f3 de l\u2019alfa-sinucle\u00efna en l&#8217;estriat.<\/h6>\n<p>En el seu estudi, publicat aquesta setmana, els investigadors revelen que, a m\u00e9s de dur a terme aquestes funcions, la PrP<sup>c<\/sup> tamb\u00e9 s&#8217;uneix a una forma pat\u00f2gena de l\u2019alfa-sinucle\u00efna i fomenta la seva propagaci\u00f3 entre neurones. Aquesta prote\u00efna, quan est\u00e0 fosforil\u00b7lada &#8211; o el que \u00e9s el mateix, a la seva forma maligna -, causa la degeneraci\u00f3 i mort neuronal en malalties com el Parkinson. El resultat dels investigadors apunta a la PrP<sup>c<\/sup> com una possible diana terap\u00e8utica per frenar l&#8217;avan\u00e7 en aquest tipus de patologies.<\/p>\n<p>Aquesta descoberta cient\u00edfica, fruit de la col\u00b7laboraci\u00f3 entre tres grups de l\u2019IBEC, aix\u00ed com d&#8217;investigadors del T\u00f2quio Metropolitan Institute of Medical Science, l&#8217;Hospital de Bellvitge i l&#8217;Institut de Recerca de la Vall d\u2019Hebron a Barcelona, i l&#8217;Institut de Recerca en Enginyeria d&#8217;Arag\u00f3 a la Universitat de Saragossa, podria ser rellevant en altres patologies neurodegeneratives, incloent alguns tipus de dem\u00e8ncia, l&#8217;Alzheimer o l&#8217;atr\u00f2fia multisist\u00e8mica.<\/p>\n<p>El Parkinson \u00e9s una malaltia neurodegenerativa que afecta al moviment. S&#8217;origina quan un tipus particular de neurones es degeneren i moren, afectant a les funcions motores i cognitives del sistema nervi\u00f3s central. Els responsables d&#8217;aquesta degeneraci\u00f3 s\u00f3n els cossos de Lewy, uns agregats an\u00f2mals que es formen a l&#8217;interior de les neurones a base d&#8217;una versi\u00f3 maligna de l\u2019alfa-sinucle\u00efna, una prote\u00efna que abunda al citoplasma de les neurones. Quan l\u2019alfa-sinucle\u00efna no es pot degradar correctament, la seva forma canvia, la prote\u00efna es fosforil\u00b7la i comen\u00e7a a aglomerar-se, creant aquests cossos de Lewy densos i insolubles.<\/p>\n<p>Aquests agregats proliferen a l&#8217;interior de la neurona i desperten la seva resposta autoimmune, que acaba causant el su\u00efcidi de la pr\u00f2pia c\u00e8l\u00b7lula. El proc\u00e9s \u00e9s r\u00e0pid i devastador, at\u00e8s que la forma pat\u00f2gena de l\u2019alfa-sinucle\u00efna \u00e9s capa\u00e7 de propagar-se a trav\u00e9s de les connexions entre neurones, creuant diferents regions del cervell i formant nous cossos de Lewy al seu pas, la qual cosa causa un dany irreversible en un nombre creixent de c\u00e8l\u00b7lules al llarg del temps.<\/p>\n<p>Els investigadors de l\u2019IBEC i els seus col\u00b7laboradors han observat que la sobre-expressi\u00f3 de la PrP<sup>c<\/sup> a la membrana neuronal incrementa la uni\u00f3 i expansi\u00f3 de l\u2019alfa-sinucle\u00efna pat\u00f2gena entre neurones i, m\u00e9s tard, la formaci\u00f3 dels cossos de Lewy. \u201cHem vist clarament com l\u2019alfa-sinucle\u00efna \u00e9s capa\u00e7 de viatjar d&#8217;una regi\u00f3 cerebral a unes altres, deixant al seu pas un nombre molt alt de neurones mortes i d\u2019altres moribundes\u201d, diu la Laura Urrea, primera autora de l&#8217;estudi.<\/p>\n<p>\u201cLa PrP<sup>c<\/sup> \u00e9s una pe\u00e7a clau per a la propagaci\u00f3 dels cossos de Lewy\u201d, afegeix Jos\u00e9 Antonio Del R\u00edo, coordinado de l&#8217;estudi i investigador principal del grup de Neurobiologia Molecular i Cel\u00b7lular de l&#8217;IBEC i professor de\u00a0la Universitat de Barcelona (UB). \u201cSi pogu\u00e9ssim evitar que aquestes acumulacions malicioses es formessin i propaguessin, podr\u00edem detenir la progressi\u00f3 de la mort neuronal en malalties com el Parkinson\u201d.<\/p>\n<p>A m\u00e9s dels seus experiments <em>in vivo<\/em> i <em>in vitro<\/em>, l&#8217;equip de Del R\u00edo ha desenvolupat un dispositiu microflu\u00eddic capa\u00e7 de distingir la interacci\u00f3 espec\u00edfica entre la PrPc i l\u2019alfa-sinucle\u00efna que facilita el seu transport entre neurones. \u201cA dia d&#8217;avui, hem identificat un domini en la PrP<sup>c<\/sup> que podria ser la diana terap\u00e8utica clau per a contenir el transport de l\u2019alfa-sinucle\u00efna entre neurones i aix\u00ed frenar l&#8217;avan\u00e7 de la malaltia\u201d, afegeix l&#8217;investigador.<\/p>\n<p>Aquest estudi, en el qual tamb\u00e9 han participat els grups de <a href=\"https:\/\/ibecbarcelona.eu\/nanobioengineering\">Nanobioingenieria<\/a>\u00a0i\u00a0<a href=\"https:\/\/ibecbarcelona.eu\/bactinf\">Infeccions bacterianes: terapies antimicrobianes<\/a>\u00a0del IBEC -liderats respectivament per Josep Samitier i\u00a0Eduard Torrents, tots dos\u00a0professores de\u00a0la Universitat de Barcelona (UB)-,ha estat possible gr\u00e0cies al finan\u00e7ament de\u00a0<a href=\"http:\/\/www.ccma.cat\/tv3\/marato\/recerca\/projectes-financats\/\" target=\"_blank\">La Marat\u00f3 de TV3<\/a>, el <a href=\"http:\/\/www.idi.mineco.gob.es\/portal\/site\/MICINN\/menuitem.00d7c011ca2a3753222b7d1001432ea0\/?vgnextoid=33881f4368aef110VgnVCM1000001034e20aRCRD\" target=\"_blank\">MINECO<\/a>\u00a0i el Centre de Recerca Biom\u00e8dica en Xarxa sobre Malalties Neurodegeneratives (<a href=\"https:\/\/ciberned.es\/\" target=\"_blank\">CIBERNED<\/a>), entre altres.<\/p>\n<p>&nbsp;<\/p>\n<p>&#8212;<\/p>\n<p><em><strong>Article:<\/strong> <\/em>Urrea L, Segura-Feliu M, Masuda-Suzukake M, Hervera A, Pedraz L, Aznar JM, Vila M, Samitier J, Torrents E, Ferrer I, Gav\u00edn R, Hagesawa M, Del R\u00edo JA.
