El treball s&#8217;ha realitzat en col\u00b7laboraci\u00f3 amb investigadors de l&#8217;IDIBAPS, la UB i la Universitat de Val\u00e8ncia.<\/p>\n<p>L&#8217;equip de del R\u00edo, amb la recentment doctorada \u00c0gata Mata i Vanessa Gil com a primeres autores, ha identificat a Sema3E i el seu receptor PlexinD1 com a prote\u00efnes que regulen la proliferaci\u00f3 de les c\u00e8l\u00b7lules mare adultes de l&#8217;hipocamp. Es tracta d&#8217;un nou rol desconegut per al binomi Sema3E \u2013 PlexinD1, que fins ara era conegut pel seu paper en la formaci\u00f3 de connexions i sinapsis espec\u00edfiques durant el desenvolupament del cervell.<\/p>\n<p>L&#8217;hipocamp \u00e9s una pe\u00e7a essencial per a la mem\u00f2ria a curt i llarg termini, aix\u00ed com per a la mem\u00f2ria espacial. Es localitza a l&#8217;interior del l\u00f2bul temporal del cervell, i li deu el seu nom a la seva forma anat\u00f2mica, que recorda a la del cavallet de mar (o hipocamp). En la malaltia d&#8217;Alzheimer, \u00e9s l&#8217;hipocamp una de les primeres regions cerebrals a patir danys, causant problemes de mem\u00f2ria i desorientaci\u00f3 com a part dels primers s\u00edmptomes de la malaltia.<br \/>\nDurant molt de temps, es va creure que les neurones adultes del sistema nervi\u00f3s central no podien regenerar-se, i que la p\u00e8rdua de mem\u00f2ria era un proc\u00e9s contra el qual l&#8217;organisme no era capa\u00e7 de lluitar. No obstant aix\u00f2, en els \u00faltims anys el descobriment de neurog\u00e8nesi en algunes \u00e0rees cerebrals ha trencat aquest dogma. Actualment, ja s&#8217;ha demostrat que les c\u00e8l\u00b7lules del bulb olfactori es regeneren, aix\u00ed com tamb\u00e9 ho fan les c\u00e8l\u00b7lules de l&#8217;hipocamp, en concret les de la zona subgranular (SGZ, de l&#8217;angl\u00e8s subgranular zone). En aquesta regi\u00f3 de l&#8217;hipocamp, un n\u00ednxol de c\u00e8l\u00b7lules mare s&#8217;encarrega de generar noves neurones i c\u00e8l\u00b7lules glials que posteriorment s&#8217;integren funcionalment. En els pacients d&#8217;Alzheimer, s&#8217;ha descrit que aquesta regeneraci\u00f3 funcional es troba alterada i no \u00e9s suficient com per recuperar el dany neurodegeneratiu present en la malaltia, la qual cosa contribueix, lamentablement, al deteriorament cognitiu dels pacients.<\/p>\n<p>En el seu \u00faltim treball, publicat recentment en la revista Scientific Reports, els investigadors de l&#8217;IBEC desxifren un nou mecanisme molecular que controla la generaci\u00f3 de noves neurones adultes en l&#8217;hipocamp. Es tracta de la prote\u00efna Sema3E i el seu receptor de membrana PlexinD1, coneguts per guiar la formaci\u00f3 de connexions i promoure la reorganitzaci\u00f3 del citosquelet neuronal durant el neurodesenvolupament.<\/p>\n<p>Els assajos desenvolupats en ratol\u00ed han revelat que, en abs\u00e8ncia de Sema3E o el seu receptor PlexinD1 en les neurones de l&#8217;hipocamp, es produeix una sobreproducci\u00f3 de noves neurones en els n\u00ednxols de l&#8217;hipocamp que migren de forma an\u00f2mala, provocant malformacions en aquesta \u00e0rea del cervell. Com a conseq\u00fc\u00e8ncia, les connexions neuronals i les ones gamma generades pels ratolins adults tamb\u00e9 pateixen canvis. \u201cSabem que les ones gamma tenen un paper rellevant en el manteniment del sistema nervi\u00f3s central; el nostre estudi demostra que l&#8217;abs\u00e8ncia de Sema3E o PlexinD1 en l&#8217;hipocamp podria promoure canvis patol\u00f2gics no nom\u00e9s a nivell morfol\u00f2gic, sin\u00f3 tamb\u00e9 a nivell de comportament\u201d, comenta \u00c0gata Mata, coprimera autora de l&#8217;estudi i investigadora predoctoral de l&#8217;IBEC i de la UB.<br \/>\nEl descobriment d&#8217;aquesta nova funci\u00f3 del binomi Sema3E-PlexinD1 podria llan\u00e7ar llum sobre els mecanismes implicats en la mem\u00f2ria o en el desenvolupament de tumors cerebrals causats per la proliferaci\u00f3 descontrolada de diversos tipus cel\u00b7lulars. \u201cSema3E exerceix un paper fonamental en la regulaci\u00f3 de l&#8217;hipocamp en desenvolupament i adult, tenint repercussions a nivell morfol\u00f2gic i fisiol\u00f2gic, que repercuteixen en la mem\u00f2ria a nivell conductual\u201d, afegeix Del R\u00edo.<\/p>\n<p>&#8212;<\/p>\n<p><strong>Article referenciat: <\/strong>Agata Mata, Vanessa Gil, Jes\u00fas P\u00e9rez-Clausell, Miguel Dasilva, Mari Carmen Gonz\u00e1lez-Calixto,\u00a0Eduardo Soriano,\u00a0Jos\u00e9 Manuel Garc\u00eda-Verdugo,\u00a0Maria V.
