Les darreres troballes del grup han donat com a resultat nanomotors autopropulsats impulsats per enzims com la ureasa o la glucosa oxidasa, per\u00f2 aix\u00ed i tot, els par\u00e0metres clau que subjeuen el moviment assolit per aquestes m\u00e0quines amb enzims encara no es coneixen per complet.<\/p>\n<p>En treballar juntament amb el grup Nanoscopy for Nanomedicine de Lorenzo Albertazzi, expert en STORM &#8211; un m\u00e8tode de microsc\u00f2pia de s\u00faper resoluci\u00f3 que va m\u00e9s enll\u00e0 de la resoluci\u00f3 de la microsc\u00f2pia \u00f2ptica convencional -, el grup del Samuel va poder investigar el paper que juga la distribuci\u00f3 i la quantitat d&#8217;enzims en la generaci\u00f3 de moviment actiu.<\/p>\n<p>&#8220;La t\u00e8cnica de microsc\u00f2pia STORM ens va permetre realitzar un mapeig en 3D dels enzims ureasa unides a la superf\u00edcie dels micromotors amb una resoluci\u00f3 d&#8217;una sola mol\u00e8cula&#8221;, diu Tania Pati\u00f1o, investigadora postdoctoral en el grup Smart Nano-Bio-Devices i primera autora de l&#8217;article. &#8220;Estudiem dos tipus de nanomotors: un fet amb poliestir\u00e8 i l&#8217;altre amb poliestir\u00e8 recobert amb una capa rugosa de di\u00f2xid de silici&#8221;.<\/p>\n<p>Els investigadors havien vist que el segon tipus de motors tenia una propulsi\u00f3 direccional m\u00e9s r\u00e0pida en comparaci\u00f3 dels seus hom\u00f2legs de poliestir\u00e8. Usant STORM, van poder detectar mol\u00e8cules individuals del combustible, l&#8217;enzim ureasa, en les superf\u00edcies dels micromotors i veure quines difer\u00e8ncies o variacions podrien estar contribuint a les difer\u00e8ncies de velocitat.<\/p>\n<p>&#8220;Tots dos tipus de motors tenien una distribuci\u00f3 asim\u00e8trica d&#8217;enzims al voltant de les seves superf\u00edcies, per la qual cosa sab\u00edem que no era solament la seva distribuci\u00f3 la que estava afectant al moviment&#8221;, diu l&#8217;investigador principal del IBEC i professor de recerca ICREA Samuel S\u00e1nchez, qui va dirigir la recerca. &#8220;No obstant aix\u00f2, quan expliquem les mol\u00e8cules de ureasa, que vam poder fer gr\u00e0cies a STORM, vam veure que hi havia un augment de 10 vegades en el nombre d&#8217;enzims en els nanomotors amb recobriment de di\u00f2xid de silici en comparaci\u00f3 dels que solament contenien poliestir\u00e8; en altres paraules, les superf\u00edcies rugoses permeten una major uni\u00f3 de l&#8217;enzim, la qual cosa condueix a un moviment m\u00e9s actiu&#8221;.<\/p>\n<p>Per comprendre millor el paper del nombre d&#8217;enzims en l&#8217;autopropulsi\u00f3 de micromotors, els investigadors van correlacionar la quantitat d&#8217;enzims, quantificades per STORM, amb la velocitat i la for\u00e7a de propulsi\u00f3, trobant un llindar espec\u00edfic de nombre de mol\u00e8cules perqu\u00e8 els motors nedessin. &#8220;Aquests resultats proporcionen nous coneixements sobre les caracter\u00edstiques de disseny dels micro i nanomotors, i ens ajudaran a aconseguir transportadors de c\u00e0rrega que es puguin utilitzar de forma segura i de manera espec\u00edfica i eficient en el cos&#8221;, diu Samuel.<\/p>\n<p>&#8212;<\/p>\n<p>Tania Pati\u00f1o, Natalia Feiner-Gracia, Xavier Arqu\u00e9, Albert Miguel-L\u00f3pez, Anita Jannasch, Tom Stumpp, Erik Sch\u00e4ffer, Lorenzo Albertazzi, Samuel S\u00e1nchez (2018). <a href=\"https:\/\/pubs.acs.org\/doi\/10.1021\/jacs.8b03460\" target=\"_blank\" rel=\"noopener\">Influence of enzyme quantity and distribution on the self-propulsion of non-Janus urease powered micromotors.<\/a> <em>J.
