Des de f\u00e0rmacs que actuen \u00fanicament en el punt del nostre cos que \u00e9s il\u00b7luminat i, per tant, lliures d&#8217;efectes secundaris no desitjats en altres regions; fins al control espacial i temporal de qualsevol prote\u00efna de la qual vulguem estudiar el funcionament en el context d&#8217;un organisme&#8221;, comenta Pau Gorostiza, professor d&#8217;investigaci\u00f3 ICREA i responsable del Grup de Nanosondes i Nanocommutadors de l&#8217;IBEC. L&#8217;estudi s&#8217;ha publicat recentment a la revista <em>Nature Communications<\/em>.<\/p>\n<p><strong>Interruptors fotosensibles de gran precisi\u00f3<br \/>\n<\/strong>La mol\u00e8cula fotocommutable que han utilitzat els investigadors \u00e9s una nova variant de l\u2019azobenz\u00e8, un compost qu\u00edmic que a les fosques t\u00e9 forma plana i, quan hi incideix la llum, es pin\u00e7a. La fotofarmacologia vol aprofitar aquesta peculiar propietat per controlar l&#8217;activitat de f\u00e0rmacs: s&#8217;introdueix a l&#8217;organisme un f\u00e0rmac inactiu combinat amb azobenz\u00e8. El disseny del f\u00e0rmac nom\u00e9s permet el seu funcionament quan l\u2019azobenz\u00e8 t\u00e9 forma de pin\u00e7a. D&#8217;aquesta manera, tot i tenir un f\u00e0rmac distribu\u00eft per tot el cos, aquest nom\u00e9s far\u00e0 efecte en aquells punts on s\u2019irradi\u00ef la llum que estimula l\u2019azobenz\u00e8, tot evitant els efectes secundaris associats a l&#8217;actuaci\u00f3 del f\u00e0rmac en zones en qu\u00e8 no \u00e9s necess\u00e0ria.<\/p>\n<p>Fins fa poc, les t\u00e8cniques basades en mol\u00e8cules fotocommutables empraven l\u00e0sers de llum cont\u00ednua violeta o blava (estimulaci\u00f3 d&#8217;un fot\u00f3) per activar aquests compostos, un m\u00e8tode que no permet focalitzar l&#8217;est\u00edmul. \u201cVol\u00edem que la mol\u00e8cula s&#8217;activ\u00e9s en un punt concret, no en tot el feix de llum que irradiem. Vam veure que les transicions amb dos fotons, que utilitzen llum infraroja, permetien aconseguir-ho, per\u00f2 l&#8217;efic\u00e0cia era molt baixa i les aplicacions, limitades. Les mol\u00e8cules que hem desenvolupat ara aconsegueixen aquest efecte amb una efic\u00e0cia del 100%. \u00c9s una tecnologia molt robusta i precisa per manipular l&#8217;activitat neuronal\u201d, Comenten Jordi Hernando i Ramon\u00a0Alib\u00e9s, investigadors del Departament de Qu\u00edmica de la UAB que han dirigit part d\u2019aquest estudi juntament amb Josep Maria Lluch i F\u00e9lix\u00a0Busqu\u00e9\u201d.<\/p>\n<p>Els investigadors han comprovat l&#8217;efectivitat de la t\u00e8cnica en neurones de ratol\u00ed i en un model animal per a l&#8217;estudi de circuits neuronals, el cuc <em>Caenorhabditis elegans<\/em>. \u201cTot i que les c\u00e8l\u00b7lules en un teixit neuronal estan molt pr\u00f2ximes, hem aconseguit seleccionar aquelles sobre les que vol\u00edem activar la mol\u00e8cula fotocommutable\u201d.<\/p>\n<p>L&#8217;estimulaci\u00f3 de dos fotons, predita per Maria G\u00f6ppert-Mayer i demostrada gr\u00e0cies als l\u00e0sers polsats desenvolupats pels guanyadors del Nobel de F\u00edsica del 2018, Donna Strickland i G\u00e9rard Mourou, ha representat una revoluci\u00f3 per a la visualitzaci\u00f3 i la manipulaci\u00f3 de l&#8217;activitat neuronal.<\/p>\n<p>Els resultats d&#8217;aquesta recerca tenen un gran potencial, ja que obren la porta a noves l\u00ednies de recerca en l&#8217;\u00e0mbit molecular. Amb la t\u00e8cnica descrita, els cient\u00edfics gaudiran d&#8217;un control espaciotemporal sense precedents sobre qualsevol mol\u00e8cula fotocommutable que desitgin investigar.<\/p>\n<p>L&#8217;estudi ha estat liderat per l&#8217;IBEC, centre d\u2019Excel\u00b7l\u00e8ncia Severo Ochoa i membre de l&#8217;Institut de Ci\u00e8ncia i Tecnologia de Barcelona (BIST), juntament amb el Departament de Qu\u00edmica de la UAB, i ha comptat amb la participaci\u00f3 del grup de Michael Krieg a l&#8217;Institut Catal\u00e0 de Ci\u00e8ncies Fot\u00f2niques (ICFO).<\/p>\n<p><em><strong>Imatge<\/strong>: Control de l&#8217;activitat de neurones individuals de cuc mitjan\u00e7ant estimulaci\u00f3 de dos fotons.
