A l&#8217;intest\u00ed prim, les vellositats i criptes intestinals proporcionen al teixit una gran superf\u00edcie, el que incrementa el volum i temps de resid\u00e8ncia de fluids, millorant la seva funci\u00f3 absortiva de nutrients i f\u00e0rmacs.<\/p>\n<p>No obstant aix\u00f2, la majoria dels models in vitro no implementen aquestes caracter\u00edstiques 3D. Els models convencionals es basen en monocapes planes de c\u00e8l\u00b7lules sobre substrats durs, el que no resulta \u00f2ptim per a la capacitat predictiva dels assaigs precl\u00ednics actuals. Fins ara, el problema principal era la disponibilitat de t\u00e8cniques de fabricaci\u00f3 simples que poguessin reproduir les geometries complexes dels teixits en materials polim\u00e8rics tous, com s\u00f3n els hidrogels. Els hidrogels s\u00f3n pol\u00edmers hidrof\u00edlics amb alt contingut d&#8217;aigua que mimetitzen les propietats mec\u00e0niques dels teixits i s&#8217;utilitzen com a substrats per al cultiu cel\u00b7lular principalment en 2D.<\/p>\n<p>Aquest m\u00e8tode presenta una nova tecnologia per a fabricar hidrogels en 3D mimetitzant l&#8217;arquitectura de l&#8217;intest\u00ed prim. Aquest nou m\u00e8tode utilitza la fotolitografia convencional per a fabricar estructures 3D amb geometries complexes en hidrogels de polietilenglicol (PEGDA) en un \u00fanic pas i sense la necessitat d&#8217;utilitzar motlles. Aquest nou m\u00e8tode \u00e9s simple i rendible per a produir models d&#8217;epiteli intestinal que poden integrar-se f\u00e0cilment en plataformes est\u00e0ndard de cultiu cel\u00b7lular.<\/p>\n<p>Amb aquest nou procediment, s\u2019ha demostrat la idone\u00eftat de la t\u00e8cnica de fotolitografia per a la producci\u00f3 de microestructures 3D amb morfologies sense la necessitat de \u00a0crear motlles.<\/p>\n<p>En aquest nou proc\u00e9s s\u2019aconsegueixen gradients controlats d\u2019oxigen durant la fotopolimeritzaci\u00f3. Al controlar els par\u00e0metres de fabricaci\u00f3, com la cin\u00e8tica de difusi\u00f3 i consum d\u2019oxigen, la dist\u00e0ncia de la font de llum i la dosis d\u2019exposici\u00f3, les dimensions i geometria de les microestructures es poden definir a la perfecci\u00f3. A m\u00e9s, la copolimeritzaci\u00f3 del polietilenglicol amb \u00e0cid acr\u00edlic millora el control de la din\u00e0mica dels processos de reacci\u00f3-difusi\u00f3 que governen la polimeritzaci\u00f3. Per una altra banda, l\u2019\u00e0cid acr\u00edlic permet la incorporaci\u00f3 controlada de lligands per a l\u2019adhesi\u00f3 cel\u00b7lular mentre que conserva les propietats mec\u00e0niques dels hidrogels.<\/p>\n<p>Els pol\u00edmers sint\u00e8tics, com per exemple PEGDA, tenen avantatges davant els pol\u00edmers naturals, com per exemple, la possibilitat d&#8217;ajustar les propietats o el fet que no s\u00f3n biodegradables i per tant, poden suportar condicions de cultiu cel\u00b7lular a llarg termini. Mitjan\u00e7ant l&#8217;ajust adequat de la composici\u00f3 del pol\u00edmer i els par\u00e0metres de fabricaci\u00f3, \u00e9s possible imitar fidelment l&#8217;arquitectura 3D de c\u00e8l\u00b7lules de l&#8217;epiteli de l&#8217;intest\u00ed prim en hidrogels sint\u00e8tics. A m\u00e9s, aquest m\u00e8tode permet la fabricaci\u00f3 d&#8217;aquests hidrogels en membranes poroses que s&#8217;adapten a les plaques de cultiu cel\u00b7lular convencional i permeten mesurar les propietats funcionals de la barrera cel\u00b7lular intestinal, com per exemple la seva integritat i permeabilitat, mitjan\u00e7ant els m\u00e8todes est\u00e0ndard.
