Els axons que regeneren s\u00f3n capa\u00e7os de formar noves sinapsis (blau) amb interneurones de la medul\u00b7la (verd).<\/figcaption><\/figure>\n<p>Entendre els mecanismes que duen a la regeneraci\u00f3 medul\u00b7lar, aix\u00ed com identificar possibles dianes terap\u00e8utiques que es podrien utilitzar per al tractament de lesions traum\u00e0tiques del sistema nervi\u00f3s central, s\u00f3n q\u00fcestions de gran rellev\u00e0ncia en la recerca biom\u00e8dica.<\/p>\n<p>En un article publicat recentment a la revista EMBO Journal, fruit de la col\u00b7laboraci\u00f3 entre els investigadors de l\u2019Institut de Bioenginyeria de Catalunya (IBEC) i l\u2019Imperial College de Londres, els investigadors han analitzat\u00a0 la capacitat regenerativa del sistema sensorial dels ganglis de l&#8217;arrel dorsal (DRG). La ra\u00f3 \u00e9s que aquestes neurones tenen una branca axonal en el sistema nervi\u00f3s central i un altre en el perif\u00e8ric, que surten d&#8217;un sol cos cel\u00b7lular, permetent estudiar els mecanismes implicats en la capacitat regenerativa dels nervis perif\u00e8rics <em>versus<\/em> la incapacitat del sistema nervi\u00f3s central. \u201cLes lesions al sistema nervi\u00f3s perif\u00e8ric responen diferent de com ho fan al sistema nervi\u00f3s central. Identificar difer\u00e8ncies en la resposta cel\u00b7lular en ambdues branques del sistema sensorial ens permet identificar possibles dianes per al tractament de lesions medul\u00b7lars\u201d comenta el Prof. Di Giovanni, responsable de la recerca.<\/p>\n<p>Els investigadors van trobar que les lesions en la branca perif\u00e8rica, per\u00f2 no en la central, desencadenaven una onada de calci que a la vegada activava una cascada de reaccions bioqu\u00edmiques que acabava inactivant un enzim anomenat\u00a0 HDAC3. Aquest enzim \u00e9s un regulador epigen\u00e8tic qu\u00e8, mitjan\u00e7ant estudis de seq\u00fcenciaci\u00f3 massiva i an\u00e0lisis funcionals bioinform\u00e0tics, els investigadors van identificar que es trobava silenciant l\u2019expressi\u00f3 de gens essencials per la regeneraci\u00f3 axonal. \u201cLa inhibici\u00f3 terap\u00e8utica de l\u2019enzim HDAC3 en models animals de lesi\u00f3 medul\u00b7lar permet augmentar la capacitat regenerativa despr\u00e9s de lesions del sistema nervi\u00f3s central, obrint aix\u00ed la porta a futurs tractaments per pacients amb lesions medul\u00b7lars\u201d conclou el Dr. Arnau Hervera, autor principal de l\u2019article i investigador a l\u2019IBEC.<\/p>\n<p>Finalment, els investigadors van comprovar que la inhibici\u00f3, gen\u00e8tica o farmacol\u00f2gica de l\u2019enzim HDAC3 provoca un creixement regeneratiu tamb\u00e9 en un model de lesi\u00f3 medul\u00b7lar en ratolins, identificant aix\u00ed una nova diana terap\u00e8utica per al tractament de les lesions medul\u00b7lars,<\/p>\n<p>L\u2019estudi va ser liderat pel doctor Arnau Hervera i dut a terme entre els laboratoris del professor Jos\u00e9 Antonio del R\u00edo a l&#8217;IBEC i del professor Simone Di Giovanni a l&#8217;Imperial College de Londres.
