La majoria de les eines per aconseguir aquesta &#8220;pertorbaci\u00f3&#8221; es basen en enfocaments gen\u00e8tics que interfereixen en l&#8217;acci\u00f3 d&#8217;una prote\u00efna o es basen en l&#8217;administraci\u00f3 local de compostos. No obstant aix\u00f2, cap d&#8217;aquests enfocaments s\u00f3n adequats per investigar esdeveniments transitoris o irreversibles, com en el cas de l&#8217;endocitosi, perqu\u00e8 \u00e9s dif\u00edcil o impossible controlar la din\u00e0mica de prote\u00efnes, la ubicaci\u00f3 espacial i el temps de l&#8217;activitat. Aquestes limitacions tamb\u00e9 poden afectar l&#8217;efic\u00e0cia i especificitat dels medicaments utilitzats en la farmacoter\u00e0pia.<\/p>\n<p>Ara, el grup &#8220;Nanoprobes and nanoswitches&#8221; de l&#8217;IBEC liderat pel Professor de Recerca ICREA Pau Gorostiza en col\u00b7laboraci\u00f3 amb l&#8217;IQAC-CSIC, IPMC, IRB, IDIBELL, UB i UAB, publiquen a la revista Chemical Science un estudi on <strong>desenvolupen les primeres petites mol\u00e8cules inhibidores de l&#8217;endocitosi controlades per llum: els dynazos<\/strong>. Aquestes mol\u00e8cules, anomenades dynazos, s\u00f3n capaces d&#8217;inhibir la dinamina, una prote\u00efna clau en endocitosi, en condicions de &#8220;no llum&#8221;. Al contrari, aplicant la llum visible, els dynazos s\u00f3n inactivats com a inhibidors de la dinamina i l&#8217;endocitosi pot tenir lloc. L&#8217;endocitosi \u00e9s un mecanisme utilitzat per totes les c\u00e8l\u00b7lules eucariotes per internalitzar material extracel\u00b7lular, i regula processos fisiol\u00f2gics clau com la captaci\u00f3 de nutrients i de factors de creixement. A m\u00e9s, tamb\u00e9 constitueix el cam\u00ed d&#8217;entrada d&#8217;una varietat de pat\u00f2gens. L&#8217;\u00fas de dynazos permetr\u00e0 controlar amb precisi\u00f3 la ubicaci\u00f3 espacial i la din\u00e0mica d&#8217;aquest proc\u00e9s cel\u00b7lular estrat\u00e8gic.<\/p>\n<h4>Dynazos: els primers inhibidors de l&#8217;endocitosi controlats per la llum<\/h4>\n<p>Els investigadors han utilitzat un disseny racional i escalonat per obtenir la primera petita mol\u00e8cula regulada per la llum capa\u00e7 d\u2019inhibir l\u2019endocitosi mitjan\u00e7ant el control de la dinamina.<\/p>\n<blockquote>\n<p style=\"text-align: center;\">Hem combinat les propietats farmacol\u00f2giques del dynasore, un inhibidor de la dinamina, amb les propietats fotocr\u00f2miques d&#8217;un grup azobenz\u00e8, una de les mol\u00e8cules sensibles a la llum (fotoswitcherable) m\u00e9s estudiades.<\/p>\n<p>&nbsp;<\/p>\n<p style=\"text-align: right;\">N\u00faria Camarero, primera coautora de l\u2019estudi.<\/p>\n<\/blockquote>\n<p>Els investigadors van caracteritzar en detall les propietats fotocrom\u00e0tiques dels dynazos sota diferents espectres de llum i espectrosc\u00f2pia d&#8217;absorci\u00f3 transit\u00f2ria, i han obtingut mol\u00e8cules que responen a la llum visible violeta, el que minimitza els efectes negatius potencials de l&#8217;\u00fas de la llum UV en sistemes vius. A m\u00e9s, l&#8217;activitat biol\u00f2gica dels dynazos, a concentracions micromolars, va ser confirmada per microsc\u00f2pia de fluoresc\u00e8ncia confocal, citometria de flux i microsc\u00f2pia de reflexi\u00f3 interna total (TIRFM) utilitzant diferents l\u00ednies cel\u00b7lulars.<\/p>\n<p>L&#8217;eina recentment desenvolupada, que permet activar i desactivar de forma remota i reversible l&#8217;activitat biol\u00f2gica de la dinamina, ser\u00e0 sens dubte molt rellevant per desxifrar el paper de l&#8217;endocitosi en processos cel\u00b7lulars com el creixement cel\u00b7lular, la diferenciaci\u00f3, la motilitat i la modulaci\u00f3 de la senyalitzaci\u00f3 dels receptors transmembrana i la transmissi\u00f3 sin\u00e0ptica, entre d&#8217;altres.
