Si avancem fins a l&#8217;any 2021, no nom\u00e9s disposem de m\u00e8todes de microsc\u00f2pia \u00f2ptica que ens permeten veure part\u00edcules diminutes amb m\u00e9s resoluci\u00f3 que mai, sin\u00f3 que tamb\u00e9 tenim t\u00e8cniques no \u00f2ptiques, com els microscopis de for\u00e7a de rastreig, amb els quals els investigadors poden construir mapes detallats d&#8217;una s\u00e8rie de propietats f\u00edsiques i qu\u00edmiques.\u00a0<\/span><strong>El grup de\u00a0<a href=\"https:\/\/ibecbarcelona.eu\/nanobioelec\" target=\"_blank\" rel=\"noopener\">Caracteritzaci\u00f3\u00a0Bioel\u00e8ctrica\u00a0a\u00a0Nanoescala<\/a>\u00a0de l&#8217;IBEC, dirigit pel professor de la UB\u00a0<a href=\"https:\/\/ibecbarcelona.eu\/member\/23\/Gabriel+Gomila+Lluch\/\" target=\"_blank\" rel=\"noopener\">Gabriel\u00a0Gomila<\/a>, en col\u00b7laboraci\u00f3 amb els membres del grup de Nanoscopia per la nanomedicina de l&#8217;IBEC, han estat analitzant les c\u00e8l\u00b7lules utilitzant un tipus especial de microsc\u00f2pia anomenada Microsc\u00f2pia Diel\u00e8ctrica de Rastreig<\/strong><span data-contrast=\"auto\">, una t\u00e8cnica avan\u00e7ada desenvolupada en els darrers anys amb la qual poden crear mapes d&#8217;una propietat f\u00edsica el\u00e8ctrica anomenada constant diel\u00e8ctrica. Cadascuna de les biomol\u00e8cules que componen les c\u00e8l\u00b7lules \u00e9s a dir, l\u00edpids, prote\u00efnes i \u00e0cids nucleics- t\u00e9 una constant diel\u00e8ctrica diferent, de manera que un mapa d&#8217;aquesta propietat \u00e9s b\u00e0sicament un mapa de la composici\u00f3 cel\u00b7lular.\u00a0<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:160,&quot;335559740&quot;:259}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">La t\u00e8cnica que han desenvolupat t\u00e9 un avantatge sobre l&#8217;actual m\u00e8tode \u00f2ptic de refer\u00e8ncia, que consisteix a aplicar un colorant fluorescent que pot alterar la c\u00e8l\u00b7lula estudiada. El seu m\u00e8tode no requereix l&#8217;addici\u00f3 de cap agent extern potencialment pertorbador. No obstant aix\u00f2, l&#8217;aplicaci\u00f3 d&#8217;aquesta t\u00e8cnica requereix un complex proc\u00e9s de\u00a0postprocessament\u00a0per convertir els observables mesurats en les magnituds f\u00edsiques, el que en el cas de les c\u00e8l\u00b7lules eucariotes implica enormes quantitats de temps de c\u00e0lcul. Aquesta vegada, van utilitzar algoritmes d&#8217;aprenentatge autom\u00e0tic en lloc de m\u00e8todes computacionals convencionals. El resultat va ser dr\u00e0stic: un cop entrenat, l&#8217;algoritme d&#8217;aprenentatge autom\u00e0tic va ser capa\u00e7 de produir un mapa de composici\u00f3\u00a0bioqu\u00edmica diel\u00e8ctrica de les c\u00e8l\u00b7lules en tan sols uns segons. En l&#8217;estudi no es van afegir subst\u00e0ncies externes a la mostra, un objectiu llargament buscat en l&#8217;obtenci\u00f3 d&#8217;imatges de composici\u00f3 en biologia cel\u00b7lular. Van aconseguir aquests r\u00e0pids resultats utilitzant un potent tipus d&#8217;algorisme anomenat xarxes neuronals, que imita la manera en qu\u00e8 operen les neurones de cervell hum\u00e0.<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:160,&quot;335559740&quot;:259}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">L&#8217;estudi va ser\u00a0descrit per Mart\u00ed\u00a0Checa,\u00a0que el va portar a terme com a part del seu doctorat en el grup de\u00a0Gomila\u00a0a l&#8217;IBEC. Ara \u00e9s investigador\u00a0postdoctoral\u00a0a l&#8217;<\/span><a href=\"https:\/\/icn2.cat\/en\/\" target=\"_blank\" rel=\"noopener\"><span data-contrast=\"none\">Institut Catal\u00e0 de\u00a0Nanoci\u00e8ncia\u00a0i\u00a0Nanotecnologia\u00a0(ICN2)<\/span><\/a><span data-contrast=\"auto\">. &#8220;\u00c9s un dels primers estudis que proporciona un mapa de composici\u00f3 bioqu\u00edmica de c\u00e8l\u00b7lules eucariotes seques tan r\u00e0pid i sense etiquetes&#8221;, explica\u00a0Checa. De fet, en aquest estudi de prova de concepte, els investigadors van utilitzar c\u00e8l\u00b7lules seques, per evitar els enormes efectes de l&#8217;aigua en els mesuraments diel\u00e8ctrics\u00a0per la seva elevada constant diel\u00e8ctrica. En un\u00a0<\/span><a href=\"https:\/\/www.mdpi.com\/2079-4991\/11\/6\/1402\/htm\" target=\"_blank\" rel=\"noopener\"><span data-contrast=\"none\">estudi de seguiment publicat recentment<\/span><\/a><span data-contrast=\"auto\">, tamb\u00e9 van analitzar c\u00e8l\u00b7lules fixes en el seu estat natural en l\u00edquid. Aqu\u00ed van poder comparar els valors obtinguts en els models sec i l\u00edquid per obtenir un mapa prec\u00eds de les biomol\u00e8cules que componen les c\u00e8l\u00b7lules eucariotes. Es tracta de les c\u00e8l\u00b7lules\u00a0multiestructurades\u00a0de les quals es componen els\u00a0animals, plantes, fongs i altres organismes. &#8220;El seg\u00fcent pas en aquesta investigaci\u00f3 \u00e9s aplicar el m\u00e8tode a c\u00e8l\u00b7lules vives el\u00e8ctricament excitables, com les neurones, on es produeix una intensa activitat el\u00e8ctrica.
