Malgrat la seva alta incid\u00e8ncia, el c\u00e0ncer de mama continua sent un repte per a metges i investigadors, ja que, a causa de la gran complexitat del teixit tumoral i del \u201cmicroambient&#8221; del tumor, \u00e9s molt dif\u00edcil reproduir les condicions reals en el laboratori per al seu estudi i tractament.<\/p>\n<p>Ara, investigadors de l&#8217;Institut de Bioenginyeria de Catalunya (IBEC) han desenvolupat un nou model de c\u00e0ncer de mama que recrea la complexitat de la composici\u00f3 de tumors de mama. Es tracta d&#8217;una \u201cbiotinta\u201d que est\u00e0 formada per teixits de mama de porcs als quals se&#8217;ls ha eliminat les c\u00e8l\u00b7lules, i que serveix de base per al creixement de c\u00e8l\u00b7lules canceroses humanes. El treball, dirigit per Elisabeth Engel, l\u00edder del grup de recerca \u201c<a href=\"https:\/\/ibecbarcelona.eu\/biomaterials\">Biomaterials per a Ter\u00e0pies Regeneratives<\/a>\u201d i Catedr\u00e0tica de la Universitat Polit\u00e8cnica de Catalunya (UPC), s&#8217;acaba de publicar en la revista cient\u00edfica <a href=\"https:\/\/pubs.acs.org\/doi\/full\/10.1021\/acsami.2c00920\">ACS Appl. Mater. Interf\u00edcies<\/a>.<\/p>\n<p><strong>Impressi\u00f3 en 3D del tumor utilitzant la nova biotinta a partir de mames de porc<\/strong><\/p>\n<p>Un tumor est\u00e0 format per c\u00e8l\u00b7lules canceroses, que es comporten i es divideixen de manera anormal i que es troben immerses en el microambient que els envolta. El microambient tumoral est\u00e0 format per la matriu extracel\u00b7lular (de les sigles en angl\u00e8s, ECM), gran varietat de c\u00e8l\u00b7lules epitelials, c\u00e8l\u00b7lules del sistema immune, c\u00e8l\u00b7lules de greix (adip\u00f2cits), factors solubles de creixement i hormones, entre altres. La matriu extracel\u00b7lular \u00e9s una part important d&#8217;aquest microambient, ja que est\u00e0 implicada en el creixement del tumor i en l&#8217;efic\u00e0cia dels medicaments. A m\u00e9s, la seva complexitat \u00e9s un dels factors que m\u00e9s limiten l&#8217;estudi del c\u00e0ncer de mama.<\/p>\n<p>La matriu extracel\u00b7lular vindria a ser com una esp\u00e8cie de suport altament complex i amb caracter\u00edstiques mec\u00e0niques, biol\u00f2giques i qu\u00edmiques molt precises, on es desenvolupen les c\u00e8l\u00b7lules canceroses. Fent un s\u00edmil amb un niu d&#8217;ocells, la palla, les fulles, les branques i altres materials que formen el niu serien l\u2019ECM que permet el desenvolupament dels ous, en aquest exemple, les c\u00e8l\u00b7lules canceroses.<\/p>\n<p>Amb l&#8217;objectiu de recrear aquesta complexa ECM, els investigadors de l&#8217;IBEC han utilitzat teixit de mames de porcs femella, aprofitant-se de la gran similitud entre els genomes de porcs i humans i que \u00e9s un teixit que es pot obtenir de manera senzilla i en gran quantitat. A trav\u00e9s de diferents procediments han eliminat les c\u00e8l\u00b7lules de porc del teixit mamari, deixant nom\u00e9s la ECM. Aquest material, al qual anomenen biotinta, \u00e9s el que servir\u00e0 de base per als passos seg\u00fcents i el desenvolupament del tumor en el laboratori.<\/p>\n<blockquote><p><em>\u201cHem pogut desenvolupar, per primera vegada, una biotinta a partir de teixits mamaris desprove\u00efts de les seves c\u00e8l\u00b7lules capa\u00e7 de mimetitzar les caracter\u00edstiques mec\u00e0niques i bioqu\u00edmiques de la matriu extracel\u00b7lular de la mama humana.\u201d<\/em><\/p><\/blockquote>\n<p style=\"text-align: right;\"><em>B\u00e0rbara Blanco-Fern\u00e1ndez, primera autora de l&#8217;estudi<\/em><\/p>\n<p><em><img decoding=\"async\" class=\"alignnone size-full wp-image-92738 lazyload\" data-src=\"http:\/\/ibecbarcelona.eu\/wp-content\/uploads\/2022\/07\/esquema.jpg\" alt=\"\" width=\"1407\" height=\"492\" src=\"data:image\/svg+xml;base64,PHN2ZyB3aWR0aD0iMSIgaGVpZ2h0PSIxIiB4bWxucz0iaHR0cDovL3d3dy53My5vcmcvMjAwMC9zdmciPjwvc3ZnPg==\" style=\"--smush-placeholder-width: 1407px; --smush-placeholder-aspect-ratio: 1407\/492;\"><\/em>Una vegada que els investigadors han aconseguit aquesta biotinta, formada per l\u2019ECM de teixit mamari de porcs, l&#8217;han emprat per a generar un tumor de mama hum\u00e0 per bioimpresi\u00f3n 3D. Per a aix\u00f2, a la biotinta han afegit c\u00e8l\u00b7lules canceroses humanes, factors com el col\u00b7lagen tipus 1 (present en grans quantitats en els tumors de mama) i altres components que mantenen l&#8217;estructura i duresa ideals del tumor. Aquesta mescla s&#8217;imprimeix en 3D i al cap d&#8217;uns dies d&#8217;incubaci\u00f3 en condicions adequades i perfectament controlades s&#8217;obt\u00e9 un tumor de mama hum\u00e0.<\/p>\n<p>La nova biotinta desenvolupada pels investigadors de l&#8217;IBEC posa en relleu la import\u00e0ncia de recrear la complexitat de la matriu extracel\u00b7lular i alhora el gran potencial d&#8217;aquests materials per a la fabricaci\u00f3 de models tumorals per bioimpresi\u00f3n 3D per a l&#8217;estudi del c\u00e0ncer.
