Aquests mini ronyons diab\u00e8tics obren les portes a estudiar, entre altres, la relaci\u00f3 entre la diabetis i la COVID-19.<\/h4>\n\n\n\n<p>Fa dos anys que milers de cient\u00edfics i metges de tot el m\u00f3n treballen per a entendre com es desenvolupa la COVID-19 i quina relaci\u00f3 t\u00e9 amb una altra mena de malalties. Diversos estudis han assenyalat, que les persones diab\u00e8tiques eren m\u00e9s propenses a desenvolupar una COVID-19 severa, aix\u00ed com que m\u00e9s del 20% dels pacients hospitalitzats per COVID-19 patien problemes renals aguts. No obstant aix\u00f2, fins avui, es desconeixia com era el factor causal que originava que aix\u00f2 succe\u00eds.\u00a0<\/p>\n\n\n\n<div class=\"wp-block-image is-style-default\"><figure class=\"aligncenter\"><img decoding=\"async\" data-src=\"http:\/\/ibecbarcelona.eu\/wp-content\/uploads\/2022\/05\/grup-NMontserrat-scaled-e1652369113651.jpg\" alt=\"\" class=\"wp-image-91720 lazyload\" src=\"data:image\/svg+xml;base64,PHN2ZyB3aWR0aD0iMSIgaGVpZ2h0PSIxIiB4bWxucz0iaHR0cDovL3d3dy53My5vcmcvMjAwMC9zdmciPjwvc3ZnPg==\" \/><\/figure><\/div>\n\n\n\n<p>Ara, un equip internacional liderat per Nuria Montserrat, professora de recerca ICREA en l&#8217;Institut de Bioenginyeria de Catalunya (IBEC) i investigadora principal del grup \u201c<a href=\"https:\/\/ibecbarcelona.eu\/ipscs\" target=\"_blank\" rel=\"noreferrer noopener\">Pluripotencia per a la regeneraci\u00f3 d&#8217;\u00f2rgans<\/a>\u201d, en col\u00b7laboraci\u00f3, entre altres, amb investigadors de la Universitat de Florida, el Life Sciences Institute de la Universitat de British Columbia al Canad\u00e0, Karolinska Institute i Karolinska University Hospital a Su\u00e8cia, ha utilitzat la <strong>bioenginyeria per a desenvolupar mini ronyons que simulen el rony\u00f3 de pacients en les fases inicials de la diabetis.<\/strong>\u00a0<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-style-large is-layout-flow wp-block-quote-is-layout-flow\" style=\"font-size:11px\"><p>El nostre model de organoide renal diab\u00e8tic ens ha perm\u00e8s observar que els mini ronyons diab\u00e8tics, amb major nombre de receptors ACE2, presenten una major susceptibilitat a la infecci\u00f3 viral.<\/p><cite>Elena Garreta, investigadora en l&#8217;IBEC i primera coautora de l&#8217;estudi\u00a0<\/cite><\/blockquote>\n\n\n\n<p>En el treball, Montserrat tamb\u00e9 ha comptat amb la col\u00b7laboraci\u00f3 d&#8217;investigadors espanyols del Cl\u00ednic CIMA i la Universitat de Navarra, l&#8217;Hospital Cl\u00ednic de Barcelona, l&#8217;Hospital de la Santa Creu i Sant Pau a Barcelona i de la Universitat de Barcelona. Aquest estudi ha rebut finan\u00e7ament de l&#8217;Institut de Salut Carles III (ISCIII), a trav\u00e9s de les ajudes FONS COVID19: Convocat\u00f2ria Finan\u00e7ament Extraordinari de Projectes de Recerca sobre el SARS-CoV-2 i la malaltia COVID-19 i PLATAFORMA ISCIII BIOBANCOS I BIOMODELOS: Plataformes ISCIII de suport a la I+D+I en Biomedicina i Ci\u00e8ncies de la Salut.&nbsp;<\/p>\n\n\n\n<p>Aquesta \u00e9s la primera vegada que els investigadors utilitzen organoides de rony\u00f3 per a entendre les primeres fases de la diabetis en aquest \u00f2rgan.&nbsp;<\/p>\n\n\n\n<p>Amb la finalitat de <strong>demostrar que el receptor ACE2 t\u00e9 un paper essencial per a la infecci\u00f3 de SARS-CoV-2 en el rony\u00f3<\/strong>, els investigadors tamb\u00e9 van generar, mitjan\u00e7ant enginyeria gen\u00e8tica, organoides defectius per a altres receptors descrits fins avui com a \u201cportes d&#8217;entrada\u201d per al virus. Aquesta fase de l&#8217;estudi es va realitzar amb finan\u00e7ament de les Ajudes Fundaci\u00f3 BBVA a equips de recerca cient\u00edfica SARS-CoV-2 i COVID-19.&nbsp;<\/p>\n\n\n\n<p>El treball ha utilitzat, a m\u00e9s, mostres de pacients, posant en relleu el paper del metabolisme energ\u00e8tic en la infecci\u00f3 per SARS-CoV-2. Aquestes troballes obren la porta a la identificaci\u00f3 de noves intervencions terap\u00e8utiques per a tractar la COVID-19. Aquest estudi trencador s&#8217;acaba de publicar en la prestigiosa revista<em> Cell Metabolism<\/em>.&nbsp;<\/p>\n\n\n\n<div style=\"height:10px\" aria-hidden=\"true\" class=\"wp-block-spacer\"><\/div>\n\n\n\n<h3 class=\"wp-block-heading\"><strong><strong>Els mini ronyons diab\u00e8tics tenen m\u00e9s portes d&#8217;entrada per al SARS-CoV-2\u00a0<\/strong><\/strong><\/h3>\n\n\n\n<blockquote class=\"wp-block-quote has-text-align-right is-style-large is-layout-flow wp-block-quote-is-layout-flow\" style=\"font-size:11px\"><p>\u00c9s absolutament imperatiu entendre els mecanismes moleculars que subjauen a un COVID-19 m\u00e9s sever en pacients amb diabetis i altres comorbiditats metab\u00f2liques. El desenvolupament d&#8217;un organoide de rony\u00f3 diab\u00e8tic \u00e9s un gran pas cap a la dissecci\u00f3 experimental de com els canvis metab\u00f2lics poden afectar les infeccions SARS-CoV-2. Les dades tornen a demostrar que l&#8217;ACE2 \u00e9s el receptor essencial del SARS-CoV-2, fins i tot en condicions de comorbiditat.<\/p><cite>Josef Penninger, Institut de Biotecnologia Molecular<\/cite><\/blockquote>\n\n\n\n<p>Per a crear mini ronyons amb les mateixes caracter\u00edstiques cel\u00b7lulars, i les mateixes alteracions metab\u00f2liques que les que es troben en els ronyons d&#8217;una persona diab\u00e8tica en la fase inicial de la malaltia, els investigadors van desenvolupar mini ronyons en el laboratori a partir de c\u00e8l\u00b7lules mare humanes pluripotents que van ser sotmesos a condicions de cultiu en el laboratori amb la finalitat de reproduir l&#8217;entorn diab\u00e8tic.&nbsp;<\/p>\n\n\n\n<p>Posteriorment, utilitzant diferents t\u00e8cniques de biologia molecular, com l&#8217;edici\u00f3 gen\u00e8tica, els investigadors van observar que, en els mini ronyons diab\u00e8tics, l&#8217;abund\u00e0ncia de receptor ACE2 era la que determinava la susceptibilitat a la infecci\u00f3 viral, establint una relaci\u00f3 causal entre la diabetis i la pres\u00e8ncia d&#8217;un dels receptors decisius en la infecci\u00f3 per SARS-CoV-2.\u00a0<\/p>\n\n\n\n<p>A m\u00e9s, utilitzant t\u00e8cniques capdavanteres com la seq\u00fcenciaci\u00f3 de RNA, els investigadors van identificar que els mini ronyons diab\u00e8tics posseeixen una signatura metab\u00f2lica que podria explicar perqu\u00e8 els mini ronyons diab\u00e8tics s&#8217;infecten m\u00e9s.\u00a0<\/p>\n\n\n\n<div class=\"wp-block-image is-style-default\"><figure class=\"aligncenter\"><img decoding=\"async\" data-src=\"http:\/\/ibecbarcelona.eu\/wp-content\/uploads\/2022\/05\/organoides.png\" alt=\"\" class=\"wp-image-91723 lazyload\" src=\"data:image\/svg+xml;base64,PHN2ZyB3aWR0aD0iMSIgaGVpZ2h0PSIxIiB4bWxucz0iaHR0cDovL3d3dy53My5vcmcvMjAwMC9zdmciPjwvc3ZnPg==\" \/><\/figure><\/div>\n\n\n\n<h6 class=\"has-small-font-size wp-block-heading\">Imatge de microsc\u00f2pia \u00f2ptica d&#8217;un organoide de rony\u00f3 generat a partir de c\u00e8l\u00b7lules mare pluripotents humanes [imatge de l&#8217;esquerra]. Aquesta tecnologia millorar\u00e0 la nostra capacitat per investigar com el virus interacciona amb diferents \u00f2rgans en el cos hum\u00e0.<\/em><\/p><cite>Ali Mirazimi, professor adjunt a l&#8217;Institut Karolinska<\/cite><\/blockquote>\n\n\n\n<p>Per a verificar si els resultats obtinguts amb els mini ronyons s&#8217;observaven tamb\u00e9 en l&#8217;\u00f2rgan natiu, els investigadors van analitzar c\u00e8l\u00b7lules renals de pacients amb diabetis i d&#8217;individus sense diabetis. Les dades van demostrar que les c\u00e8l\u00b7lules renals de pacients diab\u00e8tics, de la mateixa manera que el que ocorre en els mini ronyons, presentaven m\u00e9s receptors ACE2 i patien susceptibilitat a la infecci\u00f3 per SARS-CoV-2.
