La pared interna del intestino, el epitelio intestinal, tiene una peculiar estructura 3D organizada en forma de dedos o de \u201cvalles y cimas\u201d, y se sabe que esta estructura 3D es fundamental para su correcto funcionamiento. Por otro lado, estos valles y cimas est\u00e1n recubiertos por una mucosa, que entre otras funciones abriga a nuestra microbiota intestinal, formada por entre 500 y 1000 especies distintas de microorganismos.&nbsp;<\/p>\n\n\n\n<p>Esta misma mucosa es tambi\u00e9n la primera barrera de protecci\u00f3n justamente para impedir que otros microorganismos potencialmente patog\u00e9nicos, o sustancias qu\u00edmicas nocivas, puedan entrar en nuestro organismo y causar enfermedades. Estos dos factores, estructura 3D + mucosa, son esenciales en la fisiolog\u00eda intestinal, sin embargo, los modelos de intestino desarrollados hasta la fecha para su estudio en laboratorio se basan en estructuras en 2D que no producen mucosidad, lo que limita mucho su relevancia y utilizaci\u00f3n.&nbsp;<\/p>\n\n\n\n<p>Ahora, <strong>investigadores del Instituto de Bioingenier\u00eda de Catalu\u00f1a (IBEC) han creado un modelo de intestino que presenta la caracter\u00edstica estructura 3D de valles y cimas, y adem\u00e1s puede generar la mucosa. Han utilizado este nuevo modelo para estudiar la interacci\u00f3n con pat\u00f3genos intestinales. <\/strong>Concretamente han podido<strong> <\/strong>simular con \u00e9xito la infecci\u00f3n bacteriana producida por una cepa de <em>E. coli<\/em> que caracteriza la enfermedad de Crohn. El trabajo ha sido dirigido por Elena Mart\u00ednez, l\u00edder del grupo \u201c<a href=\"https:\/\/ibecbarcelona.eu\/es\/biomsyscelleng\" target=\"_blank\" rel=\"noreferrer noopener\">Sistemas Biomim\u00e9ticos para Ingenier\u00eda Celular<\/a>\u201d en colaboraci\u00f3n con el grupo \u201c<a href=\"https:\/\/ibecbarcelona.eu\/es\/bactinf\" target=\"_blank\" rel=\"noreferrer noopener\">Infecciones Bacterianas y Terapias Antomicrobianas<\/a>\u201d liderado por Eduard Torrents. Los resultados se han publicado recientemente en la revista <a href=\"https:\/\/www.mdpi.com\/1999-4923\/14\/8\/1552\" target=\"_blank\" rel=\"noreferrer noopener\">Pharmaceutics<\/a>.&nbsp;<\/p>\n\n\n\n<p><strong>Fabricando mini-intestinos 3D<\/strong>&nbsp;<\/p>\n\n\n\n<p>Para construir la estructura 3D de valles y cimas del intestino, los investigadores han utilizado el m\u00e9todo de la fotopolimerizaci\u00f3n, es decir, han usado un material que adquiere su forma y consistencia finales al ser iluminado con un tipo concreto de luz UV. Con este sistema han construido una estructura 3D similar al intestino, en forma de dedos, y los diferentes tipos de c\u00e9lulas que forman ese \u00f3rgano han crecido, se han posicionado de forma similar a lo que ocurre en el intestino, y m\u00e1s importante, han sido capaces de producir mucosidad intestinal. Utilizando esta tecnolog\u00eda los investigadores han podido crear al detalle una estructura 3D similar al intestino.&nbsp;&nbsp;<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\"><p><em>\u201cLas c\u00e9lulas han colonizado nuestra estructura de mini-intestino 3D y han sido capaces de secretar la mucosidad y responder al fluido intestinal que caracteriza el intestino, dos aspectos esenciales para su correcto funcionamiento\u201d.&nbsp;<\/em>&nbsp;<em>Mar\u00eda Garc\u00eda-D\u00edaz (IBEC), primera autora del estudio&nbsp;<\/em>&nbsp;<\/p><\/blockquote>\n\n\n\n<p><strong>Un nuevo modelo para estudiar infecciones intestinales<\/strong>&nbsp;<\/p>\n\n\n\n<p>Los mini-intestinos desarrollados por los investigadores del IBEC presentan tanto la estructura 3D caracter\u00edstica de valles y cimas, como la capacidad de producir la mucosidad intestinal, siendo por tanto un modelo muy \u00fatil para estudios de intestino en el laboratorio.&nbsp;&nbsp;<\/p>\n\n\n\n<p>Para analizar la eficacia de este modelo en estudios de infecciones bacterianas, los investigadores se han centrado en la enfermedad de Crohn. Esta enfermedad pertenece al grupo de enfermedades inflamatorias intestinales, y hasta la fecha se desconocen sus causas. Afecta el tracto intestinal y se cree que ese trastorno est\u00e1 relacionado con un ataque del sistema inmune a algunos componentes bacterianos presentes en el intestino. Actualmente no existe cura para esa enfermedad y los tratamientos se limitan a paliar los s\u00edntomas.&nbsp;&nbsp;<\/p>\n\n\n\n<p>Al inocular en los mini-intestinos 3D una cepa de la bacteria <em>E. coli<\/em> relacionada con la enfermedad de Crohn, los investigadores han observado una respuesta inflamatoria similar a la que ocurre en pacientes, poniendo de manifiesto la importancia de recrear un ambiente lo m\u00e1s fidedigno posible para simular en laboratorio el comportamiento de las bacterias. En otras palabras, estos prometedores resultados refuerzan la utilidad y validez de ese nuevo modelo para el estudio de esta, y posiblemente de otras enfermedades intestinales causadas por pat\u00f3genos.
