
{"id":11841,"date":"2015-01-23T11:08:56","date_gmt":"2015-01-23T11:08:56","guid":{"rendered":"http:\/\/ibecbarcelona.eu\/new\/?p=11841"},"modified":"2015-09-28T12:40:39","modified_gmt":"2015-09-28T12:40:39","slug":"nuevo-paso-hacia-los-farmacos-controlados-por-lu","status":"publish","type":"post","link":"https:\/\/ibecbarcelona.eu\/es\/nuevo-paso-hacia-los-farmacos-controlados-por-lu\/","title":{"rendered":"Nuevo paso hacia los f\u00e1rmacos controlados por luz"},"content":{"rendered":"<h6 class=\"caption\" style=\"width: 230px; float: left; padding: 0px 10px 10px 0px;\">Imagen: Esquema de la interacci\u00f3n prote\u00edna-prote\u00edna<\/h6>\n<p>El descubrimiento signific\u00f3 un gran paso hacia el desarrollo de f\u00e1rmacos dirigidos y controlados por luz, ya que los efectos del f\u00e1rmaco se limitar\u00edan a un \u00e1rea y a un tiempo espec\u00edfico, reduciendo as\u00ed los efectos secundarios.<\/p>\n<p>Ahora, en experimentos recientes, los mismos investigadores han estado utilizando sus mol\u00e9culas foto sensibles para controlar la endocitosis -el proceso mediante el cual las c\u00e9lulas absorben ciertas sustancias que necesitan para funcionar correctamente-, mediada por clatrina, con lo que han descubrimiento que se ampl\u00eda el campo de los inhibidores potenciales.<\/p>\n<p>\u201cComo muchas de las interacciones prote\u00edna-prote\u00edna est\u00e1n mediadas por p\u00e9ptidos lineares peque\u00f1os, a menudo helicoidales (prote\u00ednas peque\u00f1as), nosotros pens\u00e1bamos que nuestras mol\u00e9culas foto conmutables ten\u00edan que ser dise\u00f1adas para imitar a estos p\u00e9ptidos r\u00edgidos para que as\u00ed fueran capaces de inhibir el PPI,\u201d explica Pau Gorostiza del grupo de Nanosondas y Nanoconmutadores del IBEC. \u201cPero ahora hemos encontrado que no necesitan tener la misma estructura r\u00edgida, y que las mol\u00e9culas con estructuras flexibles muestran una gran capacidad inhibitoria, as\u00ed como una mejor capacidad foto conmutadora\u201d.<\/p>\n<p>La ausencia de una estructura r\u00edgida y helicoidal, por lo tanto, no es una limitaci\u00f3n para seleccionar candidatos para ser inhibidores foto conmutables del PPI, lo que significa que cuando se est\u00e1n desarrollando potenciales adicionales inhibitorios, los investigadores pueden mirar entre el amplio grupo de p\u00e9ptidos m\u00e1s flexibles para encontrar su candidato.<\/p>\n<p>\u201cEstos resultados son un avance relevante de las interacciones intracelulares prote\u00edna-prote\u00edna hacia futuras terapias controladas por luz\u201d dice Ernest Giralt, investigador principal del grupo de P\u00e9ptidos y Prote\u00ednas del IRB Barcelona. \u201cEl comportamiento que muestran estos p\u00e9ptidos resalta la ventaja que puede proceder de la flexibilidad parcial de algunos inhibidores de la interacci\u00f3n prote\u00edna-prote\u00edna, lo que permite al p\u00e9ptido \u2018adaptarse\u2019 estructuralmente a la prote\u00edna diana,\u201d a\u00f1ade Laura Nevola, co-autora del art\u00edculo del IRB Barcelona.<\/p>\n<p>Las aplicaciones terap\u00e9uticas m\u00e1s inmediatas de los inhibidores foto conmutables son dirigidas a enfermedades que afectan a tejido superficial como por ejemplo la piel, la retina o las membranas mucosas externas. En el futuro, las mol\u00e9culas foto sensibles podr\u00e1n ser utilizadas para estudiar, por ejemplo, la endocitosis in vitro de las c\u00e9lulas cancer\u00edgenas \u2013en las que este es un proceso incontrolado\u2013 lo que podr\u00e1 permitir la inhibici\u00f3n selectiva de la proliferaci\u00f3n celular.<\/p>\n<p>Otra \u00e1rea de aplicaci\u00f3n para estas mol\u00e9culas puede encontrarse en la biolog\u00eda del desarrollo, donde las c\u00e9lulas requieren de la endocitosis para cambiar de forma y de funci\u00f3n. Su desarrollo tambi\u00e9n abre las puertas a un nuevo campo de investigaci\u00f3n como la optofarmacolog\u00eda o la optogen\u00e9tica.<\/p>\n<p>&#8212;<\/p>\n<p><em><strong>Art\u00edculo de
