El estudio, que utiliz\u00f3 organoides de tumores a partir de c\u00e9lulas de pacientes, se centr\u00f3 en las c\u00e9lulas madre cancerosas que expresan la prote\u00edna LGR5, un marcador clave en la se\u00f1alizaci\u00f3n necesaria para la renovaci\u00f3n y diferenciaci\u00f3n de c\u00e9lulas madre intestinales y cancerosas. Los resultados revelaron diferencias significativas en las propiedades mec\u00e1nicas entre las c\u00e9lulas que expresan LGR5 (LGR5+) y las que no lo hacen (LGR5-).<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>Esta transici\u00f3n entre estos dos tipos celulares, que trabajan en equipo, es un mecanismo adaptativo que las ayuda a soportar los cambios f\u00edsicos del microambiente y facilita la progresi\u00f3n del c\u00e1ncer.<\/p>\n<cite>Xavier Trepat<\/cite><\/blockquote>\n\n\n\n<p>M\u00e1s concretamente, los investigadores observaron que las c\u00e9lulas que no expresan LGR5 son m\u00e1s blandas, menos adhesivas y se mueven m\u00e1s r\u00e1pido, lo que las hace m\u00e1s aptas para moverse desde el tumor principal. En contraste, las c\u00e9lulas que s\u00ed expresan LGR5 son m\u00e1s aptas para adherirse a las paredes de los vasos sangu\u00edneos y formar brechas que les permiten invadir otros tejidos, lo que facilitar\u00eda su crecimiento en nuevos sitios del cuerpo, pudiendo formar met\u00e1stasis.<\/p>\n\n\n\n<p>\u201cEsta transici\u00f3n entre estos dos tipos celulares, que trabajan en equipo, es un mecanismo adaptativo que las ayuda a soportar los cambios f\u00edsicos del microambiente y facilita la progresi\u00f3n del c\u00e1ncer.\u201d Explica Xavier Trepat, profesor de investigaci\u00f3n ICREA en el IBEC.<\/p>\n\n\n\n<p>El trabajo, publicado en la prestigiosa revista cient\u00edfica <em>Nature Communications<\/em>, demuestra que estas diferencias en las propiedades mec\u00e1nicas de las c\u00e9lulas se deben a la disminuci\u00f3n de las prote\u00ednas ERM (ezrina, radixina y moesina) que ocurre en las c\u00e9lulas LGR5+. Estas prote\u00ednas conectan la membrana celular con el esqueleto interno de la c\u00e9lula, afectando a su rigidez y adhesi\u00f3n. Manipulando estas prote\u00ednas, el equipo de investigaci\u00f3n pudo hacer que las c\u00e9lulas LGR5+ y LGR5- se comportaran de manera similar, confirmando el papel crucial de las ERM en las diferencias mec\u00e1nicas observadas.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>Nuestros resultados demuestran el potencial de las prote\u00ednas ERM como objetivo terap\u00e9utico para interferir en el proceso metast\u00e1sico del c\u00e1ncer colorrectal.<\/p>\n<cite>Sefora Conti<\/cite><\/blockquote>\n\n\n\n<p>Tal y como detalla Sefora Conti, investigadora del IBEC en el grupo de Trepat y primera autora del estudio, llevaron a cabo varios experimentos para comprobar la acci\u00f3n de ERM:<\/p>\n\n\n\n<p>\u201cA\u00f1adimos a las c\u00e9lulas LGR5+ una prote\u00edna sint\u00e9tica que imita la funci\u00f3n de ERM y observamos que entonces se volv\u00edan indistinguibles mec\u00e1nicamente de las c\u00e9lulas LGR5-. Por otro lado, redujimos la expresi\u00f3n de las prote\u00ednas ERM en c\u00e9lulas LGR5- y de nuevo observamos que sus diferencias mec\u00e1nicas respecto a LGR5+ se reduc\u00edan. Estos resultados demuestran el potencial de las prote\u00ednas ERM como objetivo terap\u00e9utico para interferir en el proceso metast\u00e1sico del c\u00e1ncer colorrectal.\u201d<\/p>\n\n\n\n<p>En colaboraci\u00f3n con el grupo del Instituto de Investigaci\u00f3n Biom\u00e9dica de Barcelona (IRB) liderado por Eduard Batlle, los investigadores tambi\u00e9n analizaron los patrones de expresi\u00f3n de las prote\u00ednas ERM en c\u00e9lulas individuales de una cohorte de pacientes, mostrando que esta reducci\u00f3n en las ERM es una caracter\u00edstica com\u00fan en los tumores colorrectales.<\/p>\n\n\n\n<p>Estos descubrimientos podr\u00edan contribuir al desarrollo de nuevas estrategias para tratar y prevenir la met\u00e1stasis en el c\u00e1ncer colorrectal, que es el tercer tipo de c\u00e1ncer m\u00e1s com\u00fan en todo el mundo y la segunda causa principal de muertes relacionadas con el c\u00e1ncer.<\/p>\n\n\n\n<p>Xavier Trepat, l\u00edder de la investigaci\u00f3n, tambi\u00e9n es profesor en la Universidad de Barcelona (UB) y miembro del Centro de Investigaci\u00f3n Biom\u00e9dica en Red en Bioingenier\u00eda, Biomateriales y Nanomedicina (CIBER-BBN).
