Estos compuestos, que muestran efectos analg\u00e9sicos al activarlos por luz, pueden inhibir las se\u00f1ales nerviosas de manera localizada y bajo demanda. Este enfoque innovador implica modificar la estructura qu\u00edmica de un f\u00e1rmaco agreg\u00e1ndole un conmutador molecular activado por la luz, como el azobenceno. De esta manera se puede conseguir que el f\u00e1rmaco se active solo cuando se expone a un color de luz particular, y no en la oscuridad.<\/p>\n\n\n\n<p>Bas\u00e1ndose en estos principios, un equipo de investigaci\u00f3n liderado por el Instituto de Bioingenier\u00eda de Catalu\u00f1a (IBEC) ha desarrollo derivados fotoconmutables de la carbamazepina, un medicamento antiepil\u00e9ptico ampliamente usado en medicina, para combatir algunos tipos de dolor neurop\u00e1tico, como la neuralgia del nervio trig\u00e9mino. Estos compuestos, que tienen un efecto analg\u00e9sico tras ser activados con luz, son capaces de inhibir las se\u00f1ales nerviosas de forma local y bajo demanda. Los derivados sintetizados por los investigadores se activan en longitudes de onda correspondientes al color \u00e1mbar, lo que permite atravesar tejido y hueso usando l\u00e1mparas hal\u00f3genas convencionales.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>Cuando iluminamos las larvas que han absorbido estos compuestos con una longitud de onda espec\u00edfica, el f\u00e1rmaco se activa y las larvas se mueven m\u00e1s r\u00e1pido. Al cambiar la longitud de onda, su movimiento se reduce nuevamente, lo que demuestra el efecto reversible del compuesto sobre el sistema nervioso.<\/p>\n<cite>Luisa Camerin<\/cite><\/blockquote>\n\n\n\n<p>Los dos compuestos sintetizados, carbazopine-1 y carbadiazocine, muestran actividad fotofarmacol\u00f3gica, permitendo controlar con luz de forma reversible la actividad de las neuronas del hipocampo y la locomoci\u00f3n de larvas de pez cebra. Estos experimentos <em>in vivo<\/em> permiten observar comportamientos relacionados con la ansiedad, que se reflejan en movimientos de nado repentinos. Tal y como explica Luisa Camerin, investigadora del IBEC y primera autora del estudio: \u201cCuando iluminamos las larvas que han absorbido estos compuestos con una longitud de onda espec\u00edfica, el f\u00e1rmaco se activa y las larvas se mueven m\u00e1s r\u00e1pido. Al cambiar la longitud de onda, su movimiento se reduce nuevamente, lo que demuestra el efecto reversible del compuesto sobre el sistema nervioso.\u201d<\/p>\n\n\n\n<figure class=\"wp-block-video\"><video controls src=\"https:\/\/ibecbarcelona.eu\/wp-content\/uploads\/2024\/07\/anie202403636-s2-camerin_2024_supporting_movie_m2_carbadiazocine8_low.mp4\"><\/video><figcaption class=\"wp-element-caption\"><em>V\u00eddeo representativo del movimiento fotocontrolado en peces cebra. Los animales tratados con el veh\u00edculo (l\u00ednea negra) muestran menor actividad de nado y casi ninguna respuesta a la luz en comparaci\u00f3n con los animales no tratados. En los peces tratados con Carbadiazocina, se observan respuestas comportamentales a la luz, donde la luz de 420 nm aumenta la locomoci\u00f3n y la luz de 500 nm la disminuye.<\/em><\/figcaption><\/figure>\n\n\n\n<p>Adem\u00e1s, la carbadiazocine ha demostrado tambi\u00e9n propiedades analg\u00e9sicas: \u201cHemos observado, en modelos de rata desarrollados en el laboratorio de Esther Berrocoso en la Universidad de C\u00e1diz, que la carbadiazocine tiene un efecto analg\u00e9sico contra el dolor neurop\u00e1tico sin signos de anestesia, sedaci\u00f3n o toxicidad. Estos resultados permiten demostrar un tratamiento simple y convincente con iluminaci\u00f3n no invasiva,\u201d detalla Pau Gorostiza, profesor de investigaci\u00f3n ICREA, investigador principal en el IBEC y miembro del CIBER-BBN.<\/p>\n\n\n\n<p>El dolor neurop\u00e1tico est\u00e1 causado por lesiones o enfermedades del sistema somatosensorial, como la radiculopat\u00eda lumbar (\u00abci\u00e1tica\u00bb), la neuropat\u00eda diab\u00e9tica y el dolor cr\u00f3nico postquir\u00fargico. El tratamiento de este tipo de dolor a menudo requiere opioides, analg\u00e9sicos m\u00e1s potentes que los antiinflamatorios no esteroideos habituales &#8211; como el paracetamol y el ibuprofeno-. Sin embargo, su uso es controvertido debido a su eficacia inconsistente, la necesidad de dosis altas que pueden causar tolerancia y adicci\u00f3n, y efectos secundarios sist\u00e9micos como estre\u00f1imiento, n\u00e1useas, mareos y somnolencia.<a id=\"_msocom_1\"><\/a><\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p>Hemos observado, en modelos de rata desarrollados en el laboratorio de Esther Berrocoso en la Universidad de C\u00e1diz, que la carbadiazocine tiene un efecto analg\u00e9sico contra el dolor neurop\u00e1tico sin signos de anestesia, sedaci\u00f3n o toxicidad. Estos resultados permiten demostrar un tratamiento simple y convincente con iluminaci\u00f3n no invasiva<\/p>\n<cite>Pau Gorostiza<\/cite><\/blockquote>\n\n\n\n<p>En este contexto, las terapias basadas en la luz est\u00e1n cobrando importancia en medicina debido a su capacidad para dirigirse a regiones espec\u00edficas del cuerpo, aumentando la eficacia del tratamiento y reduciendo los efectos secundarios de los f\u00e1rmacos sist\u00e9micos.<\/p>\n\n\n\n<p>El equipo ya est\u00e1 trabajando en el pr\u00f3ximo paso previsto para este proyecto, que es activar los f\u00e1rmacos mediante luz infrarroja, que penetra con mayor profundidad en los tejidos, y utilizar fuentes de luz infrarroja port\u00e1tiles como l\u00e1seres o diodos electroluminiscentes (LEDs).<\/p>\n\n\n\n<p>Este estudio forma parte delproyecto <a href=\"https:\/\/phototheraport.org\/\">PHOTOTHERAPORT<\/a>, coordinado por el Instituto de Bioingenier\u00eda de Catalu\u00f1a (IBEC). El proyecto est\u00e1 financiado por el prestigioso programa Pathfinder Open del Consejo de Innovaci\u00f3n Europeo (EIC, por sus siglas en ingl\u00e9s) y se centra en el desarrollo de implantes luminiscentes y f\u00e1rmacos activados con luz para innovadoras terapias de neuromodulaci\u00f3n ((Programa HORIZON-EIC-2023 PATHFINDEROPEN-01, n\u00ba 101130883).
