Aquesta propietat, coneguda com <strong>a taxa de recanvi endoc\u00edtic <\/strong>(del&#8217;angl\u00e8s <em>endoc\u00edtic <\/em><em>turnover rate<\/em><em>, <\/em>ETOR), emergeix com un factor clau en l&#8217;especialitzaci\u00f3 de la barrera i es veu <strong>alterada en condicions inflamat\u00f2ries<\/strong>.<\/p>\n\n\n\n<p>\u00ab<em>Mitjan\u00e7ant una col\u00b7laboraci\u00f3 que integra biologia cel\u00b7lular, prote\u00f2mica, bioinform\u00e0tica i matem\u00e0tiques, ens preguntem si l&#8217;especialitzaci\u00f3 de la barrera hematoencef\u00e0lica es deu no nom\u00e9s a les prote\u00efnes que expressa, sin\u00f3 tamb\u00e9 a com les utilitza<\/em>\u00ab, explica <strong>Daniel Gonzalez-Carter<\/strong>, investigador s\u00e8nior del <a href=\"https:\/\/ibecbarcelona.eu\/es\/research-groups\/molecular-bionics\/\">grup de Bi\u00f2nica Molecular<\/a> de l&#8217;IBEC i l\u00edder de l&#8217;estudi.<\/p>\n\n\n\n<p><em>\u00abAquestes troballes poden ajudar a identificar noves estrat\u00e8gies terap\u00e8utiques per restaurar la salut neurovascular\u00bb<\/em>, afegeix <strong>Giuseppe Battaglia<\/strong>, professor de recerca ICREA a l&#8217;IBEC, investigador principal del grup de Bi\u00f2nica Molecular i coautor de l&#8217;estudi.<\/p>\n\n\n\n<p>La barrera hematoencef\u00e0lica \u00e9s una interf\u00edcie altament selectiva que protegeix el cervell alhora que permet el pas de nutrients i senyals essencials. Tradicionalment, la seva funci\u00f3 s&#8217;ha explicat a partir de la identitat i l&#8217;abund\u00e0ncia de les prote\u00efnes presents a la superf\u00edcie de les c\u00e8l\u00b7lules endotelials.<\/p>\n\n\n\n<p>No obstant aix\u00f2, encara no \u00e9s clar si la regulaci\u00f3 din\u00e0mica d&#8217;aquestes prote\u00efnes, l&#8217;anomenat perfil ETOR, contribueix a les propietats \u00faniques de les c\u00e8l\u00b7lules endotelials cerebrals. Aix\u00ed mateix, es desconeix si aquesta regulaci\u00f3 din\u00e0mica canvia en condicions patol\u00f2giques i afecta el funcionament de la barrera.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote is-layout-flow wp-block-quote-is-layout-flow\">\n<p><em>Aquestes troballes poden ajudar a identificar noves estrat\u00e8gies terap\u00e8utiques per restaurar la salut neurovascular<\/em><\/p>\n\n\n\n<p>Giuseppe Battaglia<\/p>\n<\/blockquote>\n\n\n\n<p>Utilitzant t\u00e8cniques avan\u00e7ades de prote\u00f2mica, l&#8217;equip va analitzar com es comporten al llarg del temps gaireb\u00e9 1.000 prote\u00efnes de membrana en c\u00e8l\u00b7lules endotelials de rata procedents del cervell i d&#8217;altres \u00f2rgans. Van descobrir que <strong>el temps que aquestes prote\u00efnes romanen a la superf\u00edcie cel\u00b7lular<\/strong> constitueix una capa addicional i independent de regulaci\u00f3.<\/p>\n\n\n\n<p>Els investigadors van identificar un <strong>perfil ETOR espec\u00edfic<\/strong> en les c\u00e8l\u00b7lules endotelials cerebrals que les diferencia clarament de les d&#8217;altres \u00f2rgans, fins i tot quan la composici\u00f3 proteica \u00e9s relativament similar.<\/p>\n\n\n\n<p>En concret, les prote\u00efnes implicades en el transport van mostrar taxes de recanvi elevades, cosa que probablement afavoreix un subministrament eficient de nutrients al cervell. Per contra, les prote\u00efnes d&#8217;uni\u00f3 cel\u00b7lular, essencials per mantenir l&#8217;estanque\u00eftat de la barrera, van presentar taxes de recanvi baixes, cosa que garanteix la seva estabilitat estructural.<\/p>\n\n\n\n<p>De forma crucial, aquestes din\u00e0miques <strong>no estaven correlacionades amb l&#8217;abund\u00e0ncia de prote\u00efnes, <\/strong>la qual cosa demostra que l&#8217;ETOR constitueix una caracter\u00edstica reguladora independent de la funci\u00f3 endotelial.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\"><strong><strong><strong>La inflamaci\u00f3 reconfigura el comportament de la barrera<\/strong><\/strong><\/strong><\/h4>\n\n\n\n<p>L&#8217;estudi tamb\u00e9 mostra que la patologia altera aquestes din\u00e0miques. Quan s&#8217;exposen a senyals inflamatoris, les c\u00e8l\u00b7lules endotelials cerebrals mantenen una composici\u00f3 proteica global similar, per\u00f2 el seu comportament canvia: el seu perfil ETOR es modifica significativament.<\/p>\n\n\n\n<p><em>\u00abObservem que les c\u00e8l\u00b7lules endotelials cerebrals gestionen les prote\u00efnes d&#8217;una forma molt espec\u00edfica que les distingeix dels vasos sanguinis de la resta de l&#8217;organisme. L&#8217;important \u00e9s que aquest comportament especialitzat s&#8217;altera durant la inflamaci\u00f3, fent que es comportin m\u00e9s com a vasos sanguinis perif\u00e8rics t\u00edpics i debilitant potencialment la seva capacitat per protegir el cervell\u00bb<\/em>, explica Gonzalez-Carter.<\/p>\n\n\n\n<p>Aix\u00ed mateix, els investigadors van comprovar que els canvis en l&#8217;abund\u00e0ncia de prote\u00efnes reflecteixen principalment <strong>respostes immunit\u00e0ries<\/strong>. En canvi, els canvis en l&#8217;ETOR revelen processos relacionats amb la <strong>remodelaci\u00f3 vascular<\/strong>. Aix\u00f2 suggereix que centrar-se \u00fanicament en els nivells de prote\u00efnes pot fer que es passin per alt mecanismes clau de la malaltia.<\/p>\n\n\n\n<h4 class=\"wp-block-heading\"><strong><strong><strong>Implicacions per a la salut cerebral i el desenvolupament de ter\u00e0pies<\/strong><\/strong><\/strong><\/h4>\n\n\n\n<p>Aquestes troballes obren noves vies per comprendre com es regula la barrera hematoencef\u00e0lica tant en condicions normals com patol\u00f2giques.
