Hasta ahora, la expresi\u00f3n de prote\u00ednas en c\u00e9lulas seleccionadas se activaba mediante 2 fotones, pero su uso quedaba limitado a c\u00e9lulas pr\u00f3ximas a la superficie debido a la elevada dispersi\u00f3n de los fotones rojos utilizados. Sin embargo, para investigar los \u00f3rganos internos durante el desarrollo es necesario, no solo mantener la viabilidad de las c\u00e9lulas marcadas, sino poder acceder a estas cuando se encuentran enmascaradas en tejidos m\u00e1s complejos o profundos.<\/p>\n<p>\u00abEsta innovadora t\u00e9cnica, basada en la fotoactivaci\u00f3n de tres fotones de longitud de onda m\u00e1s larga, supone una soluci\u00f3n al problema de la dispersi\u00f3n y, adem\u00e1s, es capaz de activar las prote\u00ednas que se encuentran en las c\u00e9lulas de los tejidos m\u00e1s profundos sin interferir en la esperanza de vida del animal\u00bb, comenta Dobryna Zalvidea, responsable del estudio en el Instituto de Bioingenier\u00eda de Catalunya (IBEC).<\/p>\n<p>Los investigadores marcaron las c\u00e9lulas musculares del coraz\u00f3n de embriones de pez cebra y descubrieron que la progenie de estas c\u00e9lulas en peces adultos estaba marcada de manera permanente sin causar ning\u00fan da\u00f1o remarcable en los tejidos. Esta metodolog\u00eda, tambi\u00e9n puede extenderse a otros tejidos y modelos animales, as\u00ed como ser de utilidad para activar, con resoluci\u00f3n espacial y temporal, la expresi\u00f3n de diferentes prote\u00ednas para controlar el comportamiento a largo plazo de c\u00e9lulas individuales o de grupos celulares <em>in vivo<\/em>.<\/p>\n<p>El sistema propuesto en este estudio basado en la fotoactivaci\u00f3n utiliza un inductor inactivo encapsulado (Cre-loxP) que penetra en el interior de todo el organismo pero que solo se vuelve funcional cuando se activa mediante la luz. Una vez activado, el inductor es capaz de modificar lugares determinados del ADN de las c\u00e9lulas de las que se quiere conocer su comportamiento. De este modo, se controla la expresi\u00f3n de las prote\u00ednas y se pueden dirigir los cambios a tipos celulares espec\u00edficos utilizando \u00fanicamente un est\u00edmulo externo como desencadenante.
