En todos estos procesos las c\u00e9lulas reorganizan su estructura global o citoesqueleto en cuesti\u00f3n de minutos. Para aumentar su contractilidad, las c\u00e9lulas aumentan la actividad de la GTPasa RhoA en la membrana accionando localmente su activador, la mol\u00e9cula GEF.<\/p>\n<p>Los investigadores del IBEC han conseguido dirigir con luz el activador (GEF) de la contractilidad a la membrana celular. Para ello, han utilizado un sistema de dos prote\u00ednas que se unen con gran afinidad cuando son expuestas a un haz azul. Una de las prote\u00ednas del par se une a la membrana y la otra se ancla al activador GEF. Cuando la c\u00e9lula es iluminada con luz azul, la prote\u00edna de la membrana secuestra al activador GEF, promoviendo la contracci\u00f3n de la c\u00e9lula. \u201cLa presencia de este factor GEF en la membrana es suficiente para aumentar hasta en un 50% la contractilidad de la c\u00e9lula iluminada\u201d, se\u00f1ala L\u00e9o Valon, investigador del IBEC y primer autor del estudio.<\/p>\n<h6 class=\"caption\" style=\"float: right; width: 50%; padding: 0px 0px 0px 0px;\"><img decoding=\"async\" class=\"wp-image-27469 alignright lazyload\" data-src=\"https:\/\/ibecbarcelona.eu\/wp-content\/uploads\/2017\/02\/FINAL_COMBINED_.gif\" alt=\"\" width=\"3780\" height=\"258\" src=\"data:image\/svg+xml;base64,PHN2ZyB3aWR0aD0iMSIgaGVpZ2h0PSIxIiB4bWxucz0iaHR0cDovL3d3dy53My5vcmcvMjAwMC9zdmciPjwvc3ZnPg==\" style=\"--smush-placeholder-width: 3780px; --smush-placeholder-aspect-ratio: 3780\/258;\" \/>Activador de la contractilidad GEF encendi\u00e9ndose y apag\u00e1ndose por luz.<\/h6>\n<p>El aumento en las fuerzas que ejercen las c\u00e9lulas es completamente reversible. \u201cEsto permite generar patrones locales y peri\u00f3dicos de contracci\u00f3n y relajaci\u00f3n, simplemente alternando la exposici\u00f3n de las c\u00e9lulas a la luz con periodos de oscuridad\u201d, contin\u00faa L\u00e9o. Los investigadores has sido capaces de contraer un epitelio en tan s\u00f3lo 5 minutos y a partir de un pulso de luz.<\/p>\n<p>\u201cLas herramientas que hemos desarrollado tienen una amplia aplicabilidad en mecanobiolog\u00eda\u201d, comenta Xavier Trepat, investigador ICREA en el IBEC y responsable de la investigaci\u00f3n. \u201cEl control de las fuerzas que hacen las c\u00e9lulas permitir\u00e1 el estudio detallado de fen\u00f3menos como la adhesi\u00f3n celular y la mecanotransducci\u00f3n a nivel subcelular, y de la deformaci\u00f3n y remodelaci\u00f3n de tejidos a nivel supracelular\u201d.<\/p>\n<p>Adem\u00e1s de sus aplicaciones en el campo de la mecanobiolog\u00eda, los investigadores ven posible que esta nueva herramienta se aplique en el estudio de la morfog\u00e9nesis embrionaria. \u201cDado que estas herramientas optogen\u00e9ticas solo implican el uso de equipamiento y manipulaciones gen\u00e9ticas habituales en cualquier laboratorio, esperamos que se conviertan en una t\u00e9cnica referente para el control de las fuerzas celulares en diversos modelos\u201d \u2013 se\u00f1ala Xavier.<\/p>\n<p>En este estudio han participado L\u00e9o Valon, Ariadna Mar\u00edn-Llaurad\u00f3 y Xavier Trepat del IBEC, as\u00ed como Thomas Wyatt y Guillaume Charras del London Centre for Nanotechnology.
