Se pueden observar neuronas corticales primarias despu\u00e9s de 15 d\u00edas de cultivo in vitro. b) Immunolocalizaci\u00f3n de la versi\u00f3n maligna de la \u03b1-sinucle\u00edna (verde) en el dispositivo microflu\u00eddico despu\u00e9s de la inoculaci\u00f3n con la \u03b1-sinucle\u00edna recombinante. c) Fotomicrograf\u00eda mostrando ejemplos de neuronas corticales adultas con un contenido significativo de sinucle\u00edna degenerada 45 d\u00edas despu\u00e9s de la inyecci\u00f3n de la \u03b1-sinucle\u00edna en el estriado.<\/h6>\n<p>En su estudio, publicado esta semana, los investigadores revelan que, adem\u00e1s de llevar a cabo esas funciones, la PrP<sup>c<\/sup> tambi\u00e9n se une a una forma pat\u00f3gena de la \u03b1-sinucle\u00edna y fomenta su propagaci\u00f3n entre neuronas. Esta prote\u00edna, cuando est\u00e1 fosforilada &#8211; o lo que es lo mismo, en su su forma maligna -, causa la degeneraci\u00f3n y muerte neuronal en enfermedades como el Parkinson. El resultado de los investigadores apunta a la PrP<sup>c<\/sup> como una posible diana terap\u00e9utica para frenar el avance en este tipo de patolog\u00edas<\/p>\n<p>Este avance cient\u00edfico, fruto de la colaboraci\u00f3n entre tres grupos del IBEC , as\u00ed como de investigadores del Tokyo Metropolitan Institute of Medical Science, el Hospital de Bellvitge y el Instituto de Investigaci\u00f3n de la Vall d\u2019Hebron en Barcelona, y el Instituto de Investigaci\u00f3n en Ingenier\u00eda de Arag\u00f3n en la Universidad de Zaragoza, podr\u00eda ser relevante en otras patolog\u00edas neurodegenerativas, incluyendo algunos tipos de demencia, el Alzheimer o la atrofia multisist\u00e9mica.<\/p>\n<p>El Parkinson es una enfermedad neurodegenerativa que afecta al movimiento. Se origina cuando un tipo particular de neuronas se degeneran y mueren, afectando a las funciones motoras y cognitivas del sistema nervioso central. Los responsables de esta degeneraci\u00f3n son los cuerpos de Lewy, unos agregados an\u00f3malos que se forman en el interior de las neuronas a base de una versi\u00f3n maligna de la \u03b1-sinucle\u00edna, una prote\u00edna que abunda en el citoplasma de las neuronas. Cuando la \u03b1-sinucle\u00edna no se puede degradar correctamente, su forma cambia, la prote\u00edna se fosforila y empieza a aglomerarse, creando estos cuerpos de Lewy densos e insolubles.<\/p>\n<p>Estos agregados proliferan en el interior de la neurona y despiertan su respuesta autoinmune, que termina causando el suicidio de la propia c\u00e9lula. El proceso es r\u00e1pido y devastador, dado que la forma pat\u00f3gena de la \u03b1-sinucle\u00edna es capaz de propagarse a trav\u00e9s de las conexiones entre neuronas, cruzando diferentes regiones del cerebro y formando nuevos cuerpos de Lewy a su paso, lo cual causa un da\u00f1o irreversible en un n\u00famero creciente de c\u00e9lulas a lo largo del tiempo.<\/p>\n<p>Los investigadores del IBEC y sus colaboradores han observado que la sobreexpresi\u00f3n de la PrP<sup>c<\/sup> en la membrana neuronal incrementa la uni\u00f3n y expansi\u00f3n de la \u03b1-sinucle\u00edna pat\u00f3gena entre neuronas y, m\u00e1s tarde, la formaci\u00f3n de los cuerpos de Lewy. \u201cHemos visto claramente como la \u03b1-sinucle\u00edna es capaz de viajar de una regi\u00f3n cerebral a otras, dejando a su paso un n\u00famero muy alto de neuronas muertas y otras moribundas\u201d dice Laura Urrea, primera autora del estudio.<\/p>\n<p>\u201cLa PrP<sup>c<\/sup> es una pieza clave para la propagaci\u00f3n de los cuerpos de Lewy\u201d, a\u00f1ade Jos\u00e9 Antonio Del R\u00edo, coordinador del estudio e investigador principal del\u00a0grupo <a href=\"https:\/\/ibecbarcelona.eu\/neurobiotechnology\">Neurobiolog\u00eda Molecular y Celular<\/a> del IBEC\u00a0y profesor en la\u00a0Universidad de Barcelona\u00a0(UB) . \u201cSi pudi\u00e9ramos evitar que estas acumulaciones maliciosas se formaran y propagaran, podr\u00edamos detener la progresi\u00f3n de la muerte neuronal en enfermedades como el Parkinson\u201d.<\/p>\n<p>Adem\u00e1s de sus experimentos <em>in vivo <\/em>e <em>in vitro<\/em>, el equipo de Del R\u00edo ha desarrollado un dispositivo microflu\u00eddico capaz de distinguir la interacci\u00f3n espec\u00edfica entre la PrP<sup>c<\/sup> y la \u03b1-sinucle\u00edna que facilita su transporte entre neuronas. \u201cA d\u00eda de hoy, hemos identificado un dominio en la PrP<sup>c<\/sup> que podr\u00eda ser la diana terap\u00e9utica clave para contener en transporte de la \u03b1-sinucle\u00edna entre neuronas y as\u00ed frenar el avance de la enfermedad\u201d, a\u00f1ade el investigador.<\/p>\n<p>Este estudio, en el que tambi\u00e9n han participado los grupos de\u00a0<a href=\"https:\/\/ibecbarcelona.eu\/nanobioengineering\">Nanobioingenier\u00eda<\/a> y <a href=\"https:\/\/ibecbarcelona.eu\/bactinf\">Infecciones Bacterianas: terapias antimicrobianas<\/a>\u00a0del IBEC -liderados respectivamente por Josep Samitier y Eduard Torrents, ambos profesores en la Universidad de Barcelona (UB)-,\u00a0ha sido posible gracias a la financiaci\u00f3n de <a href=\"http:\/\/www.ccma.cat\/tv3\/marato\/recerca\/projectes-financats\/\" target=\"_blank\">La Marat\u00f3 de TV3<\/a>, el <a href=\"http:\/\/www.idi.mineco.gob.es\/portal\/site\/MICINN\/menuitem.00d7c011ca2a3753222b7d1001432ea0\/?vgnextoid=33881f4368aef110VgnVCM1000001034e20aRCRD\" target=\"_blank\">MINECO<\/a> y el Centro de Investigaci\u00f3n Biom\u00e9dica en Red sobre Enfermedades Neurodegenerativas (<a href=\"https:\/\/ciberned.es\/\" target=\"_blank\">CIBERNED<\/a>), entre otros.<\/p>\n<p>&#8212;<\/p>\n<p><em><strong>Article:<\/strong> <\/em>Urrea L, Segura-Feliu M, Masuda-Suzukake M, Hervera A, Pedraz L, Aznar JM, Vila M, Samitier J, Torrents E, Ferrer I, Gav\u00edn R, Hagesawa M, Del R\u00edo JA.
