El trabajo se ha realizado en colaboraci\u00f3n con investigadores del IDIBAPS, la UB y la Universidad de Valencia.<\/p>\n<p>El equipo de del R\u00edo, con la recientemente doctorada \u00c1gata Mata y Vanessa Gil como primeras autoras, ha identificado a Sema3E y su receptor PlexinD1 como prote\u00ednas que regulan la proliferaci\u00f3n de las c\u00e9lulas madre adultas del hipocampo. Se trata de un nuevo rol desconocido para el binomio Sema3E \u2013 PlexinD1, que hasta ahora era conocido por su papel en la formaci\u00f3n de conexiones y sinapsis espec\u00edficas durante el desarrollo del cerebro.<\/p>\n<p>El hipocampo es una pieza esencial para la memoria a corto y largo plazo, as\u00ed como para la memoria espacial. Se localiza en el interior del\u00a0l\u00f3bulo temporal del cerebro, y le debe su nombre a su forma anat\u00f3mica, que recuerda a la del caballito de mar (o hipocampo). En la\u00a0enfermedad de Alzheimer, es el hipocampo una de las primeras regiones cerebrales en sufrir da\u00f1os, causando problemas de memoria y desorientaci\u00f3n como parte de los primeros s\u00edntomas de la enfermedad.<br \/>\nDurante mucho tiempo, se crey\u00f3 que las neuronas adultas del sistema nervioso central no pod\u00edan regenerarse, y que la p\u00e9rdida de memoria era un proceso contra el que el organismo no era capaz de luchar. No obstante, en los \u00faltimos a\u00f1os el descubrimiento de neurog\u00e9nesis en algunas \u00e1reas cerebrales ha roto dicho dogma. Actualmente, ya se ha demostrado que las c\u00e9lulas del bulbo olfativo se regeneran, as\u00ed como tambi\u00e9n lo hacen las c\u00e9lulas del hipocampo, en concreto las de la zona subgranular (SGZ, del ingl\u00e9s subgranular zone). En esta regi\u00f3n del hipocampo, un nicho de c\u00e9lulas madre se encarga de generar nuevas neuronas y c\u00e9lulas gliales que posteriormente se integran funcionalmente. En los pacientes de Alzh\u00e9imer, se ha descrito que esta regeneraci\u00f3n funcional esta alterada y no es suficiente como para recuperar el da\u00f1o neurodegenerativo presente en la enfermedad, lo que contribuye, lamentablemente, al deterioro cognitivo de los pacientes. <\/p>\n<p>En su \u00faltimo trabajo, publicado recientemente en la revista Scientific Reports, los Investigadores del IBEC descifran un nuevo mecanismo molecular que controla la generaci\u00f3n de nuevas neuronas adultas en el hipocampo. Se trata de la prote\u00edna Sema3E y su receptor de membrana PlexinD1, conocidos por guiar la formaci\u00f3n de conexiones y promover la reorganizaci\u00f3n del citoesqueleto neuronal durante el neurodesarrollo. <\/p>\n<p>Los ensayos desarrollados en rat\u00f3n han revelado que, en ausencia de Sema3E o su receptor PlexinD1 en las neuronas del hipocampo, se produce una sobreproducci\u00f3n de nuevas neuronas en los nichos del hipocampo que migran de forma an\u00f3mala, provocando malformaciones en esta \u00e1rea del cerebro. Como consecuencia, las conexiones neuronales y las ondas gamma generadas por los ratones adultos tambi\u00e9n sufren cambios. \u201cSabemos que las ondas gamma tienen un papel relevante en el mantenimiento del sistema nervioso central; nuestro estudio demuestra que la ausencia de Sema3E o PlexinD1 en el hipocampo podr\u00eda promover cambios patol\u00f3gicos no s\u00f3lo a nivel morfol\u00f3gico, sino tambi\u00e9n a nivel de comportamiento\u201d, comenta \u00c1gata Mata, coprimera autora del estudio e investigadora predoctoral del IBEC y de la UB.<br \/>\nEl descubrimiento de esta nueva funci\u00f3n del binomio Sema3E-PlexinD1 podr\u00eda arrojar luz sobre los mecanismos implicados en memoria o en el desarrollo de tumores cerebrales causados por la proliferaci\u00f3n descontrolada de diversos tipos celulares. \u201cSema3E desempe\u00f1a un papel fundamental en la regulaci\u00f3n del hipocampo en desarrollo y adulto, teniendo repercusiones a nivel morfol\u00f3gico y fisiol\u00f3gico, que repercuten en la memoria a nivel conductual\u201d, a\u00f1ade Del R\u00edo.<\/p>\n<p>&#8212;<\/p>\n<p><strong>Art\u00edculo referenciado: <\/strong>Agata Mata, Vanessa Gil, Jes\u00fas P\u00e9rez-Clausell, Miguel Dasilva, Mari Carmen Gonz\u00e1lez-Calixto,\u00a0Eduardo Soriano,\u00a0Jos\u00e9 Manuel Garc\u00eda-Verdugo,\u00a0Maria V.
