Las \u00faltimas investigaciones del grupo han dado como resultado nanomotores autopropulsados impulsados por enzimas como la ureasa o la glucosa oxidasa, pero aun as\u00ed, los par\u00e1metros clave que subyacen el movimiento logrado por estas m\u00e1quinas con enzimas a\u00fan no se conocen por completo.<\/p>\n<p>Al trabajar junto con el grupo Nanoscopy for Nanomedicine de Lorenzo Albertazzi, experto en STORM &#8211; un m\u00e9todo de microscop\u00eda de s\u00faper resoluci\u00f3n que va m\u00e1s all\u00e1 de la resoluci\u00f3n de la microscop\u00eda \u00f3ptica convencional -, el grupo de Samuel pudo investigar el papel que juega la distribuci\u00f3n y la cantidad de enzimas en la generaci\u00f3n de movimiento activo.<\/p>\n<p>\u00abLa t\u00e9cnica de microscop\u00eda STORM nos permiti\u00f3 realizar un mapeo en 3D de las enzimas ureasa unidas a la superficie de los micromotores con una resoluci\u00f3n de una sola mol\u00e9cula\u00bb, dice Tania Pati\u00f1o, investigadora postdoctoral en el grupo Smart Nano-Bio-Devices y primera autora del art\u00edculo. \u00abEstudiamos dos tipos de nanomotores: uno hecho con poliestireno y el otro con poliestireno recubierto con una capa rugosa de di\u00f3xido de silicio\u00bb.<\/p>\n<p>Los investigadores hab\u00edan visto que el segundo tipo de motores ten\u00eda una propulsi\u00f3n direccional m\u00e1s r\u00e1pida en comparaci\u00f3n con sus hom\u00f3logos solo de poliestireno. Usando STORM, pudieron detectar mol\u00e9culas individuales del combustible, la enzima ureasa, en las superficies de los micromotores y ver qu\u00e9 diferencias o variaciones podr\u00edan estar contribuyendo a las diferencias de velocidad.<\/p>\n<p>\u00abAmbos tipos de motores ten\u00edan una distribuci\u00f3n asim\u00e9trica de enzimas alrededor de sus superficies, por lo que sab\u00edamos que no era solo su distribuci\u00f3n la que estaba afectando al movimiento\u00bb, dice el investigador principal del IBEC y profesor de investigaci\u00f3n ICREA Samuel S\u00e1nchez, quien dirigi\u00f3 la investigaci\u00f3n. \u00abSin embargo, cuando contamos las mol\u00e9culas de ureasa, que pudimos hacer gracias a STORM, vimos que hab\u00eda un aumento de 10 veces en el n\u00famero de enzimas en los nanomotores con recubrimiento de di\u00f3xido de silicio en comparaci\u00f3n con los que solo conten\u00edan poliestireno; en otras palabras, las superficies rugosas permiten una mayor uni\u00f3n de la enzima, lo que conduce a un movimiento m\u00e1s activo \u00ab.<\/p>\n<p>Para comprender mejor el papel del n\u00famero de enzimas en la autopropulsi\u00f3n de micromotores, los investigadores correlacionaron la cantidad de enzimas, cuantificadas por STORM, con la velocidad y la fuerza de propulsi\u00f3n, encontrando un umbral espec\u00edfico de n\u00famero de mol\u00e9culas para que los motores nadasen. \u00abEstos resultados proporcionan nuevos conocimientos sobre las caracter\u00edsticas de dise\u00f1o de los micro y nanomotores, y nos ayudar\u00e1n a lograr transportadores de carga que se puedan utilizar de forma segura y de manera espec\u00edfica y eficiente en el cuerpo\u00bb, dice Samuel.<\/p>\n<p>&#8212;<\/p>\n<p>Tania Pati\u00f1o, Natalia Feiner-Gracia, Xavier Arqu\u00e9, Albert Miguel-L\u00f3pez, Anita Jannasch, Tom Stumpp, Erik Sch\u00e4ffer, Lorenzo Albertazzi, Samuel S\u00e1nchez (2018). <a href=\"https:\/\/pubs.acs.org\/doi\/10.1021\/jacs.8b03460\" target=\"_blank\" rel=\"noopener\">Influence of enzyme quantity and distribution on the self-propulsion of non-Janus urease powered micromotors.<\/a> <em>J.
