En el intestino delgado, las vellosidades y criptas intestinales proporcionan al tejido una gran superficie, lo que incrementa el volumen y tiempo de residencia de los fluidos, mejorando su funci\u00f3n absortiva de nutrientes y f\u00e1rmacos.<\/p>\n<p>Sin embargo, la mayor\u00eda de los modelos in vitro no implementan estas caracter\u00edsticas 3D. Los modelos convencionales se basan en monocapas planas de c\u00e9lulas sobre sustratos duros, lo que no resulta \u00f3ptimo para la capacidad predictiva de los ensayos precl\u00ednicos actuales. Hasta ahora, el problema principal era la disponibilidad de t\u00e9cnicas de fabricaci\u00f3n simples que pudieran reproducir las geometr\u00edas complejas de los tejidos en materiales polim\u00e9ricos blandos, como son los hidrogeles. Los hidrogeles son pol\u00edmeros hidrof\u00edlicos con alto contenido de agua que mimetizan las propiedades mec\u00e1nicas de los tejidos y se utilizan como sustratos para el cultivo celular principalmente en 2D<\/p>\n<p>Este m\u00e9todo presenta una nueva tecnolog\u00eda para fabricar hidrogeles en 3D mimetizando la arquitectura del intestino delgado. \u00c9ste nuevo m\u00e9todo usa la fotolitograf\u00eda convencional para fabricar microestructuras 3D con geometr\u00edas complejas en hidrogeles de polietilenglicol (PEGDA) en un \u00fanico paso y sin necesidad de usar moldes. Este nuevo m\u00e9todo es simple y rentable para producir modelos de epitelio intestinal que pueden integrarse f\u00e1cilmente en plataformas est\u00e1ndar de cultivo celular.<\/p>\n<p>Con este nuevo procedimiento, se ha demostrado la idoneidad de la t\u00e9cnica de fotolitograf\u00eda para la producci\u00f3n de microestructuras 3D con morfolog\u00edas sin la necesidad de moldear y desmoldar.<\/p>\n<p>En este nuevo proceso se consiguen gradientes controlados de ox\u00edgeno durante la fotopolimerizaci\u00f3n. Al controlar los par\u00e1metros de fabricaci\u00f3n, como la cin\u00e9tica de difusi\u00f3n y consumo de ox\u00edgeno, la distancia de la fuente de luz y la dosis de exposici\u00f3n, las dimensiones y geometr\u00eda de las microestructuras se pueden definir a la perfecci\u00f3n. Adem\u00e1s, la copolimerizaci\u00f3n del polietilenglicol con \u00e1cido acr\u00edlico mejora el control de la din\u00e1mica de los procesos de reacci\u00f3n-difusi\u00f3n que gobiernan la polimerizaci\u00f3n. Por otra parte, el \u00e1cido acr\u00edlico permite la incorporaci\u00f3n controlada de ligandos para la adhesi\u00f3n celular mientras que conserva las propiedades mec\u00e1nicas de los hidrogeles.<\/p>\n<p>Los pol\u00edmeros sint\u00e9ticos, como por ejemplo PEGDA, tienen ventajas sobre los pol\u00edmeros naturales, entre ellas, la posibilidad de ajustar las propiedades o que no son biodegradables, y pueden soportar condiciones de cultivo celular a largo plazo. Mediante el ajuste adecuado de la composici\u00f3n del pol\u00edmero y los par\u00e1metros de fabricaci\u00f3n, es posible imitar fielmente la arquitectura 3D de c\u00e9lulas del epitelio del intestino delgado en hidrogeles sint\u00e9ticos. Adem\u00e1s, este m\u00e9todo permite la fabricaci\u00f3n de estos hidrogeles en membranas porosas que se adaptan a las placas de cultivo celular convencional y permiten medir las propiedades funcionales de la barrera celular intestinal, como por ejemplo su integridad y permeabilidad, mediante los m\u00e9todos est\u00e1ndar.
