La mayor\u00eda de las herramientas para conseguir esta \u00abperturbaci\u00f3n\u00bb se basan en enfoques gen\u00e9ticos que interfieren en la acci\u00f3n de una prote\u00edna o se basan en la administraci\u00f3n local de compuestos. Sin embargo, ninguno de estos enfoques son adecuados para investigar eventos transitorios o irreversibles, como en el caso de la endocitosis, porque es dif\u00edcil o imposible controlar la din\u00e1mica de prote\u00ednas, la ubicaci\u00f3n espacial y el tiempo de la actividad. Estas limitaciones tambi\u00e9n pueden afectar la eficacia y especificidad de los medicamentos utilizados en la farmacoterapia.<\/p>\n<p>Ahora, el grupo \u00abNanoprobes and nanoswitches\u00bb del IBEC, liderado por el Profesor de Investigaci\u00f3n ICREA Pau Gorostiza, en colaboraci\u00f3n con el IQAC-CSIC, IPMC, IRB, IDIBELL, UB y UAB, publican en la revista Chemical Science un estudio donde desarrollan las primeras peque\u00f1as mol\u00e9culas inhibidoras de la endocitosis controladas por luz: los dynazos. Estas mol\u00e9culas, llamadas dynazos, son capaces de inhibir la dinamina, una prote\u00edna clave en endocitosis, en condiciones de \u00abno luz\u00bb. Al contrario, aplicando la luz visible, los dynazos son inactivados como inhibidores de dinamina y la endocitosis puede tener lugar. La endocitosis es un mecanismo utilizado por todas las c\u00e9lulas eucariotas para internalizar material extracelular, y regula procesos fisiol\u00f3gicos clave como la captaci\u00f3n de nutrientes y de factores de crecimiento. Adem\u00e1s, tambi\u00e9n es la v\u00eda de entrada de una variedad de pat\u00f3genos. El uso de dynazos permitir\u00e1 controlar con precisi\u00f3n la ubicaci\u00f3n espacial y la din\u00e1mica de este proceso celular estrat\u00e9gico.<\/p>\n<h4>Dynazos: los primeros inhibidores de la endocitosis controlados luz<\/h4>\n<p>Los investigadores han utilizado un dise\u00f1o racional y escalonado para obtener la primera peque\u00f1a mol\u00e9cula regulada por la luz capaz de inhibir la endocitosis a trav\u00e9s del control de la dinamina.<\/p>\n<blockquote>\n<p style=\"text-align: right;\">\u00abHemos combinado las propiedades farmacol\u00f3gicas del dynasore, un inhibidor de la dinamina, con las propiedades fotocr\u00f3micas de un grupo de azobenceno, una de las mol\u00e9culas sensibles a la luz (fotoswitcherable) m\u00e1s estudiadas\u201d.<\/p>\n<p>&nbsp;<\/p>\n<p style=\"text-align: right;\">Nuria Camarero, primera coautora del estudio.<\/p>\n<\/blockquote>\n<p>Los investigadores caracterizaron en detalle las propiedades fotocrom\u00e1ticas de los dynazos bajo diferentes espectros de luz y espectroscopia de absorci\u00f3n transitoria, y han obtenido mol\u00e9culas que responden a la luz visible violeta, lo que minimiza los efectos negativos potenciales del uso de la luz UV en sistemas vivos. Adem\u00e1s, la actividad biol\u00f3gica de los dynazos, a concentraciones micromolares, fue confirmada por microscopia de fluorescencia confocal, citometr\u00eda de flujo y microscopia de reflexi\u00f3n interna total (TIRFM) utilizando diferentes l\u00edneas celulares.<\/p>\n<p>La herramienta recientemente desarrollada, que permite activar y desactivar de forma remota y reversible la actividad biol\u00f3gica de la dinamina, ser\u00e1 sin duda muy relevante para descifrar el papel de la endocitosis en procesos celulares como el crecimiento celular, la diferenciaci\u00f3n, la motilidad y la modulaci\u00f3n de la se\u00f1alizaci\u00f3n de los receptores transmembrana y la transmisi\u00f3n sin\u00e1ptica, entre otros.
