Si pudi\u00e9ramos crear un portador de medicamentos que se uniera espec\u00edficamente a las c\u00e9lulas enfermas y a los pat\u00f3genos sin da\u00f1ar las c\u00e9lulas sanas, conseguir\u00edamos desarrollar terapias altamente dirigidas con muchos menos efectos secundarios. Ahora, gracias a un nuevo estudio en coautor\u00eda del Profesor de Investigaci\u00f3n ICREA y l\u00edder del Grupo de Bi\u00f3nica Molecular del IBEC Giuseppe Battaglia, estamos un paso m\u00e1s cerca de alcanzar este objetivo. En este importante trabajo, publicado recientemente en la prestigiosa revista cient\u00edfica Nature Communications, los autores describen c\u00f3mo lograron lo que denominan \u201cselectividad de rango\u201d en la uni\u00f3n biomolecular sint\u00e9tica. Investigadores de diez centros contribuyeron a este estudio, incluida la Universidad de Tecnolog\u00eda Qu\u00edmica de Beijing y el Imperial College de Londres.<\/p>\n<p>&nbsp;<\/p>\n<h4><strong>Imitando la naturaleza<\/strong><\/h4>\n<p>La uni\u00f3n ligando-receptor es el proceso mediante el cual las c\u00e9lulas reciben se\u00f1ales del entorno externo. Muchas biomol\u00e9culas son multivalentes, lo que significa que m\u00faltiples ligandos en una mol\u00e9cula se unen a m\u00faltiples receptores en otra, simult\u00e1neamente. Como resultado, tales mol\u00e9culas se unen con exquisita selectividad. Imitar esto con \u00e9xito abrir\u00eda una serie de nuevas posibilidades en biomedicina, como el desarrollo de portadores de f\u00e1rmacos de tama\u00f1o nanom\u00e9trico que solo liberan su carga terap\u00e9utica cuando se adhieren a c\u00e9lulas enfermas espec\u00edficas. Sin embargo, tal precisi\u00f3n selectiva sigue siendo un objetivo todav\u00eda por cumplir en bioingenier\u00eda.<\/p>\n<p>Ahora, en esta nueva publicaci\u00f3n, <strong>los investigadores describen c\u00f3mo construyeron nanopart\u00edculas que solo se unen a los receptores cuando la densidad del receptor est\u00e1 dentro de un rango preciso, pero no por debajo ni por encima, un fen\u00f3meno que denominan \u201cselectividad de rango\u201d<\/strong>. Se\u00f1alan que esto ocurre debido a un equilibrio de fuerzas atractivas y repulsivas a nivel molecular. Pasado un cierto umbral, las fuerzas repulsivas superan a las fuerzas atractivas, por lo que es mucho menos probable que se produzca la uni\u00f3n.<\/p>\n<p>Utilizando modelos mec\u00e1nicos estad\u00edsticos, demostraron que ciertos par\u00e1metros podr\u00edan explotarse para ajustar el l\u00edmite superior e inferior del rango de selectividad. As\u00ed, este estudio nos acerca un paso m\u00e1s hacia la creaci\u00f3n de nanopart\u00edculas sint\u00e9ticas que se unan exclusivamente a los receptores diana.<\/p>\n<p>Adem\u00e1s, el trabajo ofrece una visi\u00f3n importante que podr\u00eda cambiar la forma en que se realizar\u00e1n los estudios futuros en este campo: aumentar el n\u00famero de receptores no necesariamente aumenta la probabilidad de uni\u00f3n y, de hecho, puede provocar que disminuya significativamente.<\/p>\n<h4><strong>Hacia la administraci\u00f3n dirigida de medicamentos<\/strong><\/h4>\n<p>Este trabajo no s\u00f3lo allana el camino para una administraci\u00f3n de f\u00e1rmacos m\u00e1s dirigida y con menos efectos secundarios, sino que tambi\u00e9n ofrece pistas que podr\u00edan explicar por qu\u00e9 los tumores evaden el ataque del sistema inmunol\u00f3gico.
