N\u00faria Montserrat,\u00a0<\/span><span data-contrast=\"auto\">en colaboraci\u00f3n con investigadores\u00a0<\/span><span data-contrast=\"auto\">de la Universidad de Pensilvania y del Instituto de Ciencia y Tecnolog\u00eda de Gwangju,\u00a0<\/span><span data-contrast=\"auto\">han identificado los genes responsables de\u00a0<\/span><span data-contrast=\"auto\">la enfermedad renal cr\u00f3nica<\/span><span data-contrast=\"auto\">\u00a0y han\u00a0<\/span><span data-contrast=\"auto\">demostrado<\/span><span data-contrast=\"auto\">\u00a0que es posible revertir esta deficiencia<\/span><span data-contrast=\"auto\">, abriendo as\u00ed una posible ruta terap\u00e9utica contra el da\u00f1o renal.<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:220,&quot;335559740&quot;:360}\">\u00a0<\/span><\/p>\n<p>Haciendo uso de\u00a0t\u00e9cnicas\u00a0innovadoras\u00a0como la secuenciaci\u00f3n de\u00a0ARN\u00a0a nivel\u00a0de c\u00e9lula individual,\u00a0los\u00a0investigadores\u00a0descifraron\u00a0el papel crucial que desempe\u00f1an los genes del metabolismo lip\u00eddico a la hora de proteger frente a la insuficiencia renal cr\u00f3nica. Entre las diferentes\u00a0<\/span><span data-contrast=\"auto\">c\u00e9lulas\u00a0<\/span><span data-contrast=\"auto\">que conforman\u00a0<\/span><span data-contrast=\"auto\">estas\u00a0<\/span><span data-contrast=\"auto\">unidades de filtraci\u00f3n,<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-contrast=\"auto\">las\u00a0<\/span><span data-contrast=\"auto\">c\u00e9lulas epiteliales\u00a0<\/span><span data-contrast=\"auto\">del t\u00fabulo\u00a0<\/span><span data-contrast=\"auto\">proximal\u00a0<\/span><span data-contrast=\"auto\">de la nefrona\u00a0<\/span><span data-contrast=\"auto\">son las\u00a0<\/span><span data-contrast=\"auto\">encargadas de realizar<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-contrast=\"auto\">la reabsorci\u00f3n de agua y solutos<\/span><span data-contrast=\"auto\">, representando el 90% del\u00a0<\/span><span data-contrast=\"auto\">total<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-contrast=\"auto\">de la masa renal.\u00a0\u00a0<\/span><span data-contrast=\"auto\">En casi todos los casos de enfermedad renal cr\u00f3nica<\/span><span data-contrast=\"auto\">,<\/span><span data-contrast=\"auto\">\u00a0s<\/span><span data-contrast=\"auto\">e<\/span><span data-contrast=\"auto\">\u00a0produce un<\/span><span data-contrast=\"auto\">\u00a0da\u00f1o en\u00a0<\/span><span data-contrast=\"auto\">estas c\u00e9lulas<\/span><span data-contrast=\"auto\">, pero hasta<\/span><span data-contrast=\"auto\">\u00a0ahora, no se\u00a0<\/span><span data-contrast=\"auto\">sab\u00eda\u00a0<\/span><span data-contrast=\"auto\">qu\u00e9 mecanismos celulares eran los responsables de tal disfunci\u00f3n.\u00a0<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:220,&quot;335559740&quot;:360}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">Mediante la<\/span><span data-contrast=\"auto\">\u00a0secuenciaci\u00f3n\u00a0<\/span><span data-contrast=\"auto\">del ARN de cada una de\u00a0<\/span><span data-contrast=\"auto\">l<\/span><span data-contrast=\"auto\">a<\/span><span data-contrast=\"auto\">s miles<\/span><span data-contrast=\"auto\">\u00a0de\u00a0<\/span><span data-contrast=\"auto\">c\u00e9lulas\u00a0<\/span><span data-contrast=\"auto\">presentes en el<\/span><span data-contrast=\"auto\">\u00a0ri\u00f1\u00f3n<\/span><span data-contrast=\"auto\">, los investigadores\u00a0<\/span><span data-contrast=\"auto\">observaron<\/span><span data-contrast=\"auto\">\u00a0por primera vez<\/span><span data-contrast=\"auto\">,<\/span><span data-contrast=\"auto\">\u00a0diferencias importantes entre las c\u00e9lulas<\/span><span data-contrast=\"auto\">\u00a0sanas y enfermas<\/span><span data-contrast=\"auto\">\u00a0del t\u00fabulo proximal. E<\/span><span data-contrast=\"auto\">n concreto<\/span><span data-contrast=\"auto\">, en l<\/span><span data-contrast=\"auto\">os ri\u00f1ones de ratones con enfermedad renal cr\u00f3nica<\/span><span data-contrast=\"auto\">,\u00a0<\/span><span data-contrast=\"auto\">detectaron<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-contrast=\"auto\">que\u00a0<\/span><span data-contrast=\"auto\">una mayor proporci\u00f3n de\u00a0<\/span><span data-contrast=\"auto\">las\u00a0<\/span><span data-contrast=\"auto\">c\u00e9lulas del t\u00fabulo proximal<\/span><span data-contrast=\"auto\">\u00a0presentaban una\u00a0<\/span><i><span data-contrast=\"auto\">firma\u00a0<\/span><\/i><span data-contrast=\"auto\">molecular\u00a0<\/span><span data-contrast=\"auto\">distinta\u00a0<\/span><span data-contrast=\"auto\">a la encontrada e<\/span><span data-contrast=\"auto\">n los ri\u00f1ones de animales sanos.\u00a0<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:220,&quot;335559740&quot;:360}\">\u00a0<\/span><\/p>\n<p>Ante estas observaciones, las investigadoras de IBEC dieron un paso m\u00e1s y, utilizando minirri\u00f1ones humanos, demostraron que estos cambios eran debidos a una disminuci\u00f3n en la expresi\u00f3n de algunos de los genes que regulan el metabolismo de los l\u00edpidos en las c\u00e9lulas del t\u00fabulo proximal.\u00a0\u00a0Adem\u00e1s, \u00abgracias a un abordaje multidisciplinar, utilizando modelos animales y minirri\u00f1ones humanos que generamos mediante bioingenier\u00eda en el IBEC descubrimos que, al corregir esta deficiencia, las c\u00e9lulas del t\u00fabulo roximal pod\u00edan recuperar su funci\u00f3n en los diferentes modelos de estudio \u00ab, explica la Dra. Montserrat.<\/p>\n<h2><strong>T\u00e9cnicas de an\u00e1lisis innovadoras: ingenier\u00eda gen\u00e9tica y celular<\/strong><\/h2>\n<p><span data-contrast=\"auto\">Combinando<\/span><span data-contrast=\"auto\">\u00a0la<\/span><span data-contrast=\"auto\">\u00a0secuenciaci\u00f3n<\/span><span data-contrast=\"auto\">\u00a0de<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-contrast=\"auto\">ARN\u00a0<\/span><span data-contrast=\"auto\">a nivel de c\u00e9lula\u00a0<\/span><span data-contrast=\"auto\">individual,\u00a0<\/span><span data-contrast=\"auto\">y e<\/span><span data-contrast=\"auto\">l uso de bioingenier\u00eda para producir<\/span><span data-contrast=\"auto\">\u00a0mini<\/span><span data-contrast=\"auto\">r<\/span><span data-contrast=\"auto\">r<\/span><span data-contrast=\"auto\">i\u00f1ones<\/span><span data-contrast=\"auto\">,<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-contrast=\"auto\">fue<\/span><span data-contrast=\"auto\">\u00a0posible identificar\u00a0<\/span><span data-contrast=\"auto\">a nivel celular\u00a0<\/span><span data-contrast=\"auto\">las diferencias clave entre los ri\u00f1ones sanos y\u00a0<\/span><span data-contrast=\"auto\">enfermos.<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:220,&quot;335559740&quot;:360}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">Los\u00a0minirri\u00f1ones\u00a0humanos \u2014tambi\u00e9n denominados\u00a0organoides\u00a0de ri\u00f1\u00f3n\u2014 est\u00e1n creados mediante t\u00e9cnicas de bioingenier\u00eda a partir de c\u00e9lulas madre humanas, y recogen<\/span><span data-contrast=\"auto\">\u00a0algunos aspectos de<\/span><span data-contrast=\"auto\">\u00a0la complejidad de este \u00f3rgano real. El pasado mes de abril, ya demostraron su eficacia como modelo de estudio al utilizarlos para descifrar en tiempo r\u00e9cord c\u00f3mo el SARS-Co-V2 interacciona e infecta las c\u00e9lulas\u00a0<\/span><span data-contrast=\"auto\">de estos\u00a0minirri\u00f1ones<\/span><span data-contrast=\"auto\">, adem\u00e1s de identificar una terapia dirigida a reducir\u00a0<\/span><span data-contrast=\"auto\">la\u00a0<\/span><span data-contrast=\"auto\">carga viral.<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:220,&quot;335559740&quot;:360}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">En este estudio la secuenciaci\u00f3n de ARN a nivel de c\u00e9lula individual de las muestras de los\u00a0mini<\/span><span data-contrast=\"auto\">r<\/span><span data-contrast=\"auto\">ri\u00f1ones\u00a0se ha realizado en el laboratorio del Dr. Felipe\u00a0Pr\u00f3sper\u00a0de la\u00a0<\/span><span data-contrast=\"auto\">Cl\u00ednica\u00a0<\/span><span data-contrast=\"auto\">U<\/span><span data-contrast=\"auto\">niversidad de Navarra y CIMA<\/span><span data-contrast=\"auto\">\u00a0Universidad de Navarra<\/span><span data-contrast=\"auto\">.<\/span><span data-contrast=\"auto\">\u00a0\u201cEl ri\u00f1\u00f3n tiene m\u00e1s de 23 tipos de c\u00e9lulas diferentes\u201d, explica\u00a0<\/span><span data-contrast=\"auto\">la<\/span><span data-contrast=\"auto\">\u00a0Dr. Montserrat. \u201cHace\u00a0<\/span><span data-contrast=\"auto\">unos\u00a0<\/span><span data-contrast=\"auto\">a\u00f1os, era necesario examinar<\/span><span data-contrast=\"auto\">\u00a0m\u00faltiples muestras por separado para obtener la informaci\u00f3n<\/span><span data-contrast=\"auto\">, lo que pod\u00eda llevar\u00a0<\/span><span data-contrast=\"auto\">a\u00f1os<\/span><span data-contrast=\"auto\">\u00a0de\u00a0<\/span><span data-contrast=\"auto\">trabajo<\/span><span data-contrast=\"auto\">,\u00a0<\/span><span data-contrast=\"auto\">ahora<\/span><span data-contrast=\"auto\">\u00a0esta<\/span><span data-contrast=\"auto\">\u00a0misma<\/span><span data-contrast=\"auto\">\u00a0informaci\u00f3n la podemos\u00a0<\/span><span data-contrast=\"auto\">obtener en\u00a0<\/span><span data-contrast=\"auto\">pocos\u00a0<\/span><span data-contrast=\"auto\">d\u00edas<\/span><span data-contrast=\"auto\">\u201d, y a\u00f1ade: \u201cc<\/span><span data-contrast=\"auto\">on la secuenciaci\u00f3n de ARN de\u00a0<\/span><span data-contrast=\"auto\">cada una de las c\u00e9lulas de<\/span><span data-contrast=\"auto\">l ri\u00f1\u00f3n<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-contrast=\"auto\">y utilizando<\/span><span data-contrast=\"auto\">\u00a0m\u00faltiples\u00a0<\/span><span data-contrast=\"auto\">muestras, podemos<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-contrast=\"auto\">identificar qu\u00e9 genes est\u00e1n diferencialmente expresados en cada una de las c\u00e9lulas<\/span><span data-contrast=\"auto\">\u00ab.\u00a0<\/span><span data-ccp-props=\"{&quot;201341983&quot;:0,&quot;335551550&quot;:6,&quot;335551620&quot;:6,&quot;335559739&quot;:220,&quot;335559740&quot;:360}\">\u00a0<\/span><\/p>\n<p><span data-contrast=\"auto\">Estudiando estas diferencias\u00a0<\/span><span data-contrast=\"auto\">o,<\/span><span data-contrast=\"auto\">\u00a0dicho de otra manera,\u00a0<\/span><span data-contrast=\"auto\">analizando la\u00a0<\/span><span data-contrast=\"auto\">\u201cfirma\u201d\u00a0<\/span><span data-contrast=\"auto\">de la expresi\u00f3n del ARN\u00a0<\/span><span data-contrast=\"auto\">de cada una de estas c\u00e9lulas, los investigadores identificaron que\u00a0<\/span><span data-contrast=\"auto\">la disfunci\u00f3n del t\u00fabulo proximal\u00a0<\/span><span data-contrast=\"auto\">es consecuencia de\u00a0<\/span><span data-contrast=\"auto\">la desregulaci\u00f3n de una v\u00eda metab\u00f3lica clave. Espec\u00edficamente, en las muestras enfermas<\/span><span data-contrast=\"auto\">\u00a0de ratones<\/span><span data-contrast=\"auto\">,\u00a0<\/span><span data-contrast=\"auto\">tambi\u00e9n\u00a0<\/span><span data-contrast=\"auto\">se hall\u00f3<\/span><span data-contrast=\"auto\">\u00a0una mayor proporci\u00f3n de c\u00e9lulas inmaduras. \u201cSe sabe que\u00a0<\/span><span data-contrast=\"auto\">una peque\u00f1a proporci\u00f3n de\u00a0<\/span><span data-contrast=\"auto\">las c\u00e9lulas de los t\u00fabulos proximales<\/span><span data-contrast=\"auto\">\u00a0del ri\u00f1\u00f3n<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-contrast=\"auto\">presentan\u00a0<\/span><span data-contrast=\"auto\">un<\/span><span data-contrast=\"auto\">\u00a0comportamiento similar al de las c\u00e9lulas madre\u201d, se\u00f1ala\u00a0<\/span><span data-contrast=\"auto\">la<\/span><span data-contrast=\"auto\">\u00a0<\/span><span data-contrast=\"auto\">Dra.
