{"id":8080,"date":"2014-03-07T08:04:29","date_gmt":"2014-03-07T08:04:29","guid":{"rendered":"http:\/\/ibecbarcelona.eu\/new\/investigadores-crean-el-primer-bazo-en-un-chip-humano-funcional-del-mundo\/"},"modified":"2015-08-24T11:53:38","modified_gmt":"2015-08-24T11:53:38","slug":"investigadores-crean-el-primer-bazo-en-un-chip-humano-funcional-del-mundo","status":"publish","type":"post","link":"https:\/\/ibecbarcelona.eu\/es\/investigadores-crean-el-primer-bazo-en-un-chip-humano-funcional-del-mundo\/","title":{"rendered":"Investigadores crean el primer \u2018bazo-en-un-chip\u2019 humano funcional del mundo"},"content":{"rendered":"<p><span style=\"line-height: 1.3em;\">La idea original de crear un bazo-en-un-chip surgi\u00f3 de los grupos del Dr. Hernando A del Portillo, Profesor ICREA del CRESIB centro de investigaci\u00f3n de ISGlobal, quien estudia hace varios a\u00f1os el papel del bazo en la malaria y del Dr. Josep Samitier, director del IBEC y catedr\u00e1tico de la Universidad de Barcelona, quien estudia las propiedades reol\u00f3gicas de la sangre, incluyendo aquella parasitada por malaria, para desarrollar sistemas de diagn\u00f3stico. \u201dDebido a las limitaciones \u00e9ticas y tecnol\u00f3gicas de estudiar el bazo humano, conocido como la \u201ccaja negra\u201d de la cavidad abdominal, ha habido muy pocos avances en su estudio\u201d, explica del Portillo. Para romper esta barrera se inici\u00f3 una colaboraci\u00f3n para desarrollar un modelo del bazo humano-en-un-chip mediante un proyecto EXPLORA.<\/span><\/p>\n<p>\u201cEl sistema flu\u00eddico del bazo es muy complejo y adaptado evolutivamente para filtrar y destruir selectivamente gl\u00f3bulos rojos viejos, micro-organismos y gl\u00f3bulos rojos parasitados por malaria,\u201d explica el Dr. Antoni Homs, investigador del IBEC y coautor del estudio. \u201cEl bazo filtra la sangre mediante un m\u00e9todo \u00fanico, haci\u00e9ndola \u2018microcircular\u2019 a trav\u00e9s de lechos de filtraci\u00f3n formados por la pulpa roja del bazo en un compartimento especial donde el hematocrito (el porcentaje de c\u00e9lulas rojas de la sangre) se ve aumentado. De modo que los macr\u00f3fagos especializados pueden reconocer y destruir gl\u00f3bulos rojos enfermos.\u201d Adem\u00e1s, la sangre en este compartimento solo puede viajar en un \u00fanico sentido a trav\u00e9s de ranuras interendoteliales antes de llegar al sistema circulatorio, lo que representa un riguroso segundo test para asegurar la eliminaci\u00f3n de las c\u00e9lulas viejas o enfermas.<\/p>\n<p>Los investigadores de estos dos centros, pertenecientes a la red de centros CERCA, han imitado estas dos condiciones de control en su plataforma de tama\u00f1o micro para simular la microcirculaci\u00f3n de la sangre a trav\u00e9s de dos canales principales (uno lento y uno r\u00e1pido) dise\u00f1ados para dividir el flujo. En el canal \u2018lento\u2019 la sangre fluye a trav\u00e9s de una matriz de pilares simulando el ambiente real donde el hematocrito aumenta y la sangre \u201cenferma\u201d es destruida. El dispositivo ya se ha probado con gl\u00f3bulos rojos humanos sanos y en infectados por malaria, trabajo realizado mayoritariamente por los investigadores predoctorales Luis G. Rigat-Brugarolas (IBEC) y Aleix Elizalde-Torrent (CRESIB\/ISGlobal), coautores tambi\u00e9n de este trabajo. \u201cNuestro dispositivo facilitar\u00e1 el estudio de la funci\u00f3n del bazo en malaria, e incluso podr\u00eda proporcionar una plataforma flexible para la detecci\u00f3n de posibles f\u00e1rmacos contra \u00e9sta y otras enfermedades hematol\u00f3gicas,\u201d dice del Portillo.<\/p>\n<p>\u201cLa investigaci\u00f3n en \u00f3rganos-en-un-chip integrando microflu\u00eddica con sistema celulares a\u00fan est\u00e1 dando sus primeros pasos, pero ofrece enormes perspectivas hacia el futuro de los ensayos de f\u00e1rmacos para diferentes patolog\u00edas\u201d, especifica Samitier. Estos dispositivos en 3D, que imitan las interrelaciones tejido-tejido y los microambientes \u00fanicamente vistos en los \u00f3rganos vivos, permite una nueva percepci\u00f3n de las enfermedades que no puede obtenerse f\u00e1cilmente con los estudios convencionales con animales, que son costosos y consumen mucho tiempo.
