Background/Objectives: The question addressed in the current work is to develop a simulation of a pharmaceutical process (DNA encapsulation within lipid nanoparticles using a microfluidic micromixer) which will be of utility to the end users (laboratory-scale formulation development). The simulation and the microfluidic approach also address sustainability issues, such as reducing the environmental impact of the process itself, and reducing the need for physical testing. The paper details the implementation and validation, taking into account key performance indicators and control parameters. Methods: The main method applied for simulation development is a novel multi-agent approach to incorporate stochastic probabilistic behavior, combined with theoretical definitions from the process experts and relevant literature, and data/results from laboratory-scale experiments with different parameter configurations. Results: The simulation was implemented as a representation of the real physical process, reproducing the relationships between process parameters (flow rates) and experimental key performance indicators (capsule diameter, poly dispersion index, encapsulation efficiency). The simulation results demonstrated a general agreement with the empirical results and provided useful predictive insights for the laboratory experiments. Conclusions: The simulation has potential as a support tool for laboratory experiments to reduce physical testing and indicate the most promising configurations on which to focus, with potential savings in time, resources and other costs.

Neurodegenerative conditions, including Alzheimer's, Parkinson's, amyotrophic lateral sclerosis, and Huntington's disease, represent a critical medical challenge due to their increasing prevalence, severe consequences, and absence of curative treatments. Beyond the need for a deeper understanding of the fundamental mechanisms underlying neurodegeneration, the development of effective treatments is hindered by the blood-brain barrier, which poses a major obstacle to delivering therapeutic agents to the central nervous system. This review provides a comprehensive analysis of the current landscape of nanoparticle-based strategies to overcome the blood-brain barrier and enhance drug delivery for the treatment of neurodegenerative diseases. The nanocarriers reviewed in this work encompass a diverse array of nanoparticles, including polymeric nanoparticles (e.g. micelles and dendrimers), inorganic nanoparticles (e.g. superparamagentic iron oxide nanoparticles, mesoporous silica nanoparticles, gold nanoparticles, selenium and cerium oxide nanoparticles), lipid nanoparticles (e.g. liposomes, solid lipid nanoparticles, nanoemulsions), as well as quantum dots, protein nanoparticles, and hybrid nanocarriers. By examining recent advancements and highlighting future research directions, we aim to shed light on the promising role of nanomedicine in addressing the unmet therapeutic needs of these diseases.Graphical AbstractSchematic representation of the different types of nanoparticles used to tackle neurodegeneration.

The use of nanocarriers in medicine, so-called nanomedicine, is one of the most innovative strategies for targeting drugs at the action site and increasing their activity index and effectiveness. Phytomedicine is the oldest traditional method used to treat human diseases and solve health problems. The recent literature on the treatment of malaria infections using nanodelivery systems and phytodrugs or supplements has been analyzed. For the first time, in the present review, a careful look at the considerable potential of nanomedicine in promoting phytotherapeutic efficacy was done, and its key role in addressing a translation through a significant reduction of the current burden of malaria in many parts of the world has been underlined.
